Abstract (English)

The synthesis of Fmoc protected SAAC and their use in solid-phase synthesis is reported. Peptide � metal complex conjugates were either obtained by incorporation of the metal coordinated SAAC followed by mild nucleophilic resin-cleavage or by complexation in metal salt solution after cleavage from the resin. The reported solid phase peptide synthesis protocols are suitable for automation and the position and number of the modified amino acid within the peptide chain may vary. This allows the synthesis of libraries of modified peptides with metal complexes as binding sites in synthetic receptors or inhibitors.

Die Synthese von Fmoc geschützten Amino Säuren mit Chelatliganden und ihre Anwendung in der Festphasensynthese wird berichtet. Peptid � Metallkomplex Konjugate wurden entweder durch Inkorporation von Metall gebundenen, auf Amino Säure basierenden Chelatliganden erhalten, welche dann anschließend unter milden Bedingungen vom Resin abgespalten wurden. Alternativ konnten Peptid � Metallkomplex Konjugate dargestellt werden indem die Komplexierung in Lösung stattfand and am Ende vom Resin abgespalten wurde. Die beschriebenen Festphasen-Protokolle eignen sich zur Automatisierung. Die Position und die Anzahl der modifizierten Amino Säuren mögen innerhalb der Peptidkette variieren. Dies ermöglicht die Synthese von Peptid-Bibliotheken mit Metallkomplexen als Bindungsstellen für synthetische Rezeptoren oder Inhibitoren.
