Abstract (English)

Several reports have identified the H4R as an important receptor in inflammatory mechanisms and thus, as a therapeutic target for several diseases. The focus of this work was to identify and further characterize the role of mH4R on mouse myeloid immune cells.
A novel finding of this thesis is the expression of mH4R in DC macrophages, and T cells (CD4+, CD8+) which was determined on the mRNA level. The expression was down-regulated during bone marrow differentiation to BMDC as well as to BMDM, and both cell types expressed lower mH4R levels when stimulated by LPS, but IFN-gamma seemed to be an inducer of mH4R expression.
A novel function of mH4R is the chemotactic effect on BMDM in response to 4MEH. This finding supported other findings from human and mouse cells.
In the functional analysis, histamine was found not to have any direct effect on BMDM functions such as phagocytosis, TNF, IL-6 and NO production. Conversely, the absence of the mH4R on macrophages revealed a partial functional defect in phagocytosis and the production of cytokine. These findings indicate a possible role of mH4R in bone marrow differentiation to macrophages.
Based on the findings in this thesis several topics should be further investigated, and the exact role of mH4R on myeloid cells should be analyzed on the molecular level with the advantage of the H4R -/- mice.

Der H4R wurde in mehreren Berichten als Rezeptor, der eine wichtige Rolle bei entzündlichen Mechanismen spielt, beschrieben. Aus diesem Grund stellt er einen therapeutischen Angriffspunkt für verschiedene Krankheiten dar. Der Fokus dieser Arbeit war die Identifizierung und weitere Charakterisierung der Rolle des H4R auf myeloiden Immunzellen in der Maus.
In dieser Doktorarbeit konnte erstmals die Expression des H4R in dendritischen Zellen, Makrophagen und T-Zellen (CD4+, CD8+) auf mRNA-Ebene gezeigt werden. Während der Differenzierung von Knochenmarkszellen zu BMDC bzw. BMDM konnte eine Herunterregulation der Expression des Rezeptors beobachtet werden. Außerdem zeigten beide Zelltypen (BMDC und BMDM) eine verringerte mH4R-Expression nach Stimulation mit LPS; IFN-gamma hingegen scheint die H4R-Expression zu erhöhen.
Weiterhin konnte in dieser Doktorarbeit gezeigt werden, dass BMDM eine Chemotaxis hin zu 4MEH zeigen und diese durch den H4R vermittelt wird. Dieses Ergebnis stützt andere Berichte, die von humanen bzw. murinen Zellen stammen und ähnliche Beobachtungen beschreiben. Bei der funktionellen Analyse zeigte sich, dass Histamin keinen direkten Effekt auf die Phagozytoseaktivität, TNF-, IL-6- und NO-Produktion der BMDM hat. Im Gegensatz dazu führt die Abwesenheit des mH4R auf Makrophagen zu einem funktionellen Defekt, wenn man die Phagozytose und die Zytokinproduktion betrachtet. Diese Ergebnisse deuten darauf hin, dass der mH4R eine Rolle bei der Differenzierung von Knochenmarkszellen zu Makrophagen spielt.
Basierend auf den Ergebnissen dieser Doktorarbeit sollte weiterhin die Rolle des mH4R auf myeloiden Zellen untersucht werden. Hierbei sollte ein besonderes Augenmerk auf der Untersuchung des mH4R auf molekularer Ebene unter Verwendung der H4R-/--Mäuse liegen.
