Abstract
Zusammenfassung: Der Metabolismus der antipsoriatisch
wirksamen Verbindungen 1,8,9-Triacetoxyanthracen und
1,8-Diacetoxy-9-anthron wird untersucht. Es bilden sich 1,8-
Dihydroxy - 9 - anthron, 1 - Hydroxy - 8 - acetoxy-9-anthron,
1,8,1 ',8'- Teträhydroxy-bianthron, 1,8-Dihydroxy-anthrachinon,
1,8-Diacetoxy-anthrachinon und 1-Hydroxy-8-acetoxy-
anthrachinon. Quantitative Bestimmungen dieser „Metaboliten"
zeigen, daß die hydrolytischen Reaktionen vor den
Oxidationen stattfinden. Im Gegensatz zu 1,8-Dihydroxy-9-
anthron lassen 1,8,9-Triacetoxyanthracen und 1,8-Diacetoxy-
9-anthron psoriatische Läsionen verschwinden, ohne Entzündungen
hervorzurufen. Die Unwirksamkeit des 1,8,9-
Trimethoxyanthracens spricht ebenfalls dafür, daß es sich
zumindest beim 1,8,9-Triacetoxy-anthracen um eine Verbindung
mit „prodrug"-Charakter handelt. — Die hier beschriebenen
10,10-dialky Herten 1,8- Dihydroxy -9- anthrone
sind in Übereinstimmung mit der Krebs'sehen Hypothese
unwirksam bei Psoriasis.
Summary: Studies on the Metabolism of Antipsoriatic
Anthrone-Derivatives
The metabolisatiön of the antipsomatically active molecules
1,8,9-triacetoxy-anthracene und 1,8-diacetoxy-9-anthrone
by serum is described. Under these conditions 1,8-dihydroxy -
9-anthrone, l-hydroxy-8-acetoxy-9-anthrone, 1,8 ,V ,8'-tetrahydroxy-
bisanthrone, 1,8-dihydroxy-anthraquinone, 1,8-diacetoxy-
anthraquinone and 1-hydroxy-8-acetoxy-anthraquinone
arise from both educts. Quantitative determinations
of these metabolites indicate that hydrolytic reactions
occur prior to oxidation. Contrary to l,8-dihydroxy-9-
anthrone, 1,8,9-triacetoxyanthracene and 1,8-diacetoxy-9-
anthrone are effective against psoriatic lesions without accompanying
inflammations of the skin. 1,8,9-Trimethoxyanthracene,
however, is ineffective, also indicating that at
least 1,8,9-triacetoxy-anthracene is a prodrug. — In agreement
with Krebs' hypothesis 10,10-dialky lated 1,8-dihydroxy-
9-anthrones described in this paper are ineffective against
psoriasis.
