Zusammenfassung (Deutsch)
Das Inflammasom ist ein Proteinverbund mit Vertretern der NALP Familie als zentrale Bestandteile. Er nimmt eine Schlüsselfunktion im Ablauf des Entzündungsprozesses ein. NALP2 und NALP3 konnten als zentrale Bestandteile des gleichnamigen NALP2/3-Inflammasoms identifiziert werden.
Ziel der vorliegenden Arbeit war es, die mRNA-Expression und Regulation verschiedener NALPs in ausgewählten Zelllinien, primären IEZ, sowie bei Patienten mit MC und UC zu untersuchen.
Von den untersuchten NALP2 bis 14 wurde in HT29- und CaCo2-Zellen jeweils die Expression von NALP2 bis 11, 13 und 14 nachgewiesen. Die Höhe der NALP-Expressionswerte in SW480- und T84-Zellen unterschieden sich nur gering und zeigten im Vergleich zu den anderen untersuchten Zellen die höchsten Expressionen. NALP12 – nachweislich in Makrophagen und eosinophilen Granulozyten exprimiert – konnte in der vorliegenden Arbeit nur in diesen beiden Zelllinien und in zwei der vier primären IEZ Proben nachgewiesen werden. Das als oozytenspezifische Protein entdeckte NALP5 konnte qualitativ und in sehr geringen Mengen auch quantitativ in allen untersuchten Zellen nachgewiesen werden.
Das NALP Expressionsniveau der untersuchten primären IEZ entsprach im Durchschnitt dem der Tumorzelllinien. Dieser Teil der Arbeit belegt die mRNA-Expression der verschiedenen NALP Proteine in Zelllinien, aber auch in primären IEZ.
Ähnlich den Expressionsuntersuchungen wurde für NALP5 und NALP12 in den Regulationsversuchen mit ausgewählten primären IEZ keine Expression detektiert. Für die übrigen untersuchten NALPs konnte durch Inkubation keine im Vergleich zur Kontrolle ohne Stimulation erhöhte Expression gemessen werden. Im Gegenteil wurde bei NALP2, 3, 6, 8, 9, 10, 11, 13 und 14 eine statistisch signifikante Minderexpression mit unterschiedlichen Konzentrationen von MDP und Pam3Cys festgestellt.
Die Regulationsversuche von NALP2 und NALP3 wurden an HT29-Zellen fortgeführt. Sowohl die mRNA-Expression von NALP2 als auch die von NALP3 wurden durch 100 µg/ml MDP signifikant erniedrigt. Diese für NALP2 beobachtete signifikante Minderexpression war auch bei primären IEZ nachweisbar. Bei einer Stimulation der HT29-Zellen mit 500 ng/ml Pam3Cys konnte für NALP2 und NALP3 jeweils eine geringe, nicht signifikante Mehrexpression gemessen werden. Aus den Ergebnissen der Regulationsuntersuchungen der vorliegenden Arbeit an HT29-Zellen im Entzündungsmodell mit INFγ Vorstimulation geht MDP als adäquater Stimulus, der zu einer signifikanten Expressionssteigerung von NALP2, 7, 11, 13 und 14 führt, hervor. Eine signifikante Erniedrigung der Expression einzelner NALPs konnte nicht gemessen werden. Die eindruckvollste Expressionssteigerung konnte für NALP2 – die zentrale Komponente des NALP2 Inflammasoms – gemessen werden. Ihre Stimulierbarkeit durch MDP weist eine Beteiligung am komplexen immunologischen Geschehen des Körpers und besonders des Darms nach. Inwieweit andere NALPs Inflammasome bilden, ist nicht bekannt.
Die mRNA-Expressionsuntersuchungen von NALP2 bei Morbus Crohn und Colitis ulcerosa zeigten im Vergleich zu nicht entzündeten Darmresektaten ein signifikant höheres Expressionsniveau. Dabei lagen die Expressionswerte von NALP2 bei MC Patienten deutlich höher als die der UC Patienten. Die Expression von NALP3 mRNA zeigte in beiden Patientengruppen signifikant niedrigere Werte im Vergleich zu nicht entzündeten primären IEZ. Die Untersuchung weist eine Beteiligung des NALP2/3 Inflammasoms bei CED nach.
Die signifikant höhere Expression von NALP2 bei Patienten mit CED wie MC und UC bekräftigt die Ergebnisse, die eine vermehrte IL-18 Sekretion bei solchen Patienten nachgewiesen haben, und bestätigt die Vermutung, dass ein Ungleichgewicht zwischen pro- und anti-inflammatorischen Zytokinen der Darmschleimhaut ursächlich für die Entstehung CED ist.

Infalmmasome is a complex of proteins with representatives of the NALP family as integral parts. It plays a fundamental role in inflammatory processes. Likewise, NALP2 and NALP3 were identified as central components of the correspondent NALP2/3-Inflammasomes.
Objective of the study was to examine the mRNA expression and regulation of various NALPs in specific cell-lines, primarily IEC (intraepithelial cells), and in patients suffering from CD (Crohn’s disease) and UC (Ulcerative colitis).
Altogether, NALP2 to 14 were analyzed, whereby the expression of NALP2 to 11, 13 and 14 was detected in HT29- and CaCo2-cells. The level of NALP-expression in SW480 and T84 cells differed only slightly; in comparison to other cells which were studied they displayed the highest rate of expression. In the present study NALP12, which is demonstrably expressed in macrophages and eosinophils, was only detected in these two cell lines and in two of the four primary IEC samples. The oozyt-specific protein NALP5, was detected qualitatively as well as in a very small amount quantitatively, in all tested cells.
On average, the level of NALP expression of the studied primary IEC corresponded to tumor cell lines. This part of the work shows the mRNA expression of the NALP proteins in different cell lines, but also in primary IEC.
As in expression studies, even in regulation experiments with selected primary IEC no expression of NALP5 and NALP12 was detected. Compared to NALPs which were subjected to incubation treatments, the negative group without specific stimulation did not show an increased level of expression. On the contrary, in NALP2, 3, 6, 8, 9, 10, 11, 13 and 14 there was a statistically significant under-expression with different concentrations of MDP and Pam3Cys.
Regulation experiments of NALP2 and NALP3 were continued in HT29 cells. Both, the mRNA expression of NALP2 and NALP3 decreased significantly after the addition of 100µg/ml MDP. This significant downregulation for NALP2 was also detectable in primary IEC. By stimulating HT29 cells with 500ng/ml Pam3Cys, the expression measured for NALP2 and NALP3 did not rise significantly. From the results of regulation tests regarding HT29 cells in an inflammatory model with INFγ pre-stimulation emerges that MDP is an adequate stimulus to obtain a significant increase in the expression level of NALP2, 7, 11, 13 and 14. A significant reduction of NALP-expression could not be measured. The most striking increase in the expression level was detected for NALP2, the central component of the NALP2 inflammasomes. The fact that they can be stimulated by adding MDP shows their involvement in the complex immunological system of the body, especially of the intestine. But it is unknown to what extent other NALPs form infalmmasomes.
By comparing the expression of mRNA for NALP2 in individuals suffering from Crohn's disease or ulcerative colitis and individuals with a non inflamed bowel wall we can see a significantly higher level of expression in the former. The expression levels of NALP2 in CD patients were significantly higher than those of UC patients. The expression of NALP3 mRNA in both groups showed significantly lower values compared to non-inflamed primary IEC. The study proves the involvement of the NALP2 / 3 inflammasome in IBD (inflammatory bowel disease).
The significantly higher expression of NALP2 in patients suffering from IBD, such as MC and UC, supports the findings that have demonstrated an increased IL-18 secretion in such patients, and confirms the suspicion that an imbalance of pro-and anti-inflammatory cytokines in the intestinal mucosa is involved in the development of IBD.
