Abstract (German)
Residente T-Zellen in Lymphknoten werden kurz nach der subkutanen Infektion mit Leishmanien aktiviert und beginnen zu proliferieren in drainierenden Lymphknoten von C57BL/6 Mäusen. Katakai et. al. postulierten 2004, dass diese Aktivierung von T-Zellen eventuell in distinkten "immunologischen Plattformen" an der Grenze zwischen B-Zellfollikel und T-Zellbereich stattfindet. Bisher konnte eine solche Plattform in einer T-Zell-vermittelten Immunantwort gegen einen intrazellulären Erreger nicht dargestellt werden. In meiner Dissertation konnte ich zeigen, dass die Aktivierung von CD4+ T-Zellen im "cortical ridge" stattfindet.
Follikuläre T-Zellen sind für eine erfolgreiche humorale Immunantwort notwendig. In meiner Dissertation konnte ich zeigen, dass Langerin+ Zellen aus der Haut für die Induktion von follikulären T-Zellen verantwortlich sind.
Translation of the abstract (English)
Lymph node resident T cells are activated and start to proliferate within skin draining-lymph nodes (SDLNs) of C57BL/6 mice priming early after subcutaneous infection with Leishmania parasites. Katakai et al. postulated in 2004 that T cell priming might take place inside distinct “immunological platforms” located at the interface of the B cell follicle and T cell area. However, the existence of such a distinct micro-compartment in T cell mediated immunity against intracellular parasites has not been described so far. In my thesis I could show that priming of CD4+ T cells occur within the cortical ridge.
Follicular T cells are necessary to maintain a successful humoral immuneresponse. In my thesis I could show that Langerin+ cells from skin a responsible for the induction of follicular T cells.
