Weidenrinde besitzt eine lange Tradition als gut verträgliches pflanzliches Arzneimittel gegen Schmerzen und Fieber. Dabei wurde lange Zeit das Phenolglucosid Salicin als wirksamkeitsbestimmender Inhaltsstoff betrachtet. Neuere Untersuchungen belegen jedoch, dass Salicin nicht allein für die antiinflammatorischen Eigenschaften ausschlaggebend sein kann. So rückten vor allem die Polyphenole in den Fokus wissenschaftlicher Untersuchungen. Die vorliegende Arbeit leistet einen Beitrag, das Polyphenolspektrum von STW 33-1 bzw. S. purpurea L. genauer zu beschreiben und prominente Flavonoide zu quantifizieren. Zudem wurden mögliche Wirkmechanismen und die Resorption von Extrakt bzw. Extraktfraktionen in vitro untersucht.
Der standardisierte Trockenextrakt STW 33-I (S. purpurea L., DEV 16-23:1, 23-26% Gesamtsalicingehalt, Auszugsmittel: Wasser) wurde durch Flüssig-Flüssig-Verteilung fraktioniert und phytochemisch charakterisiert. Die Ethylacetatfraktion wies den höchsten Flavonoid-Anteil auf. 20 Substanzen, darunter Flavanone, Flavanonole und deren Glucoside, Chalkonglucoside und korrespondierende Cumarsäureester wurden aus dieser Fraktion isoliert und ihre Strukturen aufgeklärt. NMR-Daten und eine stereochemische Zuordnung der epimeren 6''-O-p-trans-Cumaroyl-(2S/2R)-naringenin-5-O-β-D-glucoside wurden vorgelegt. Weiterhin lieferte die Ethylacetatfraktion Salicylalkoholderivate und einfache phenolische Verbindungen, wobei Populosid B erstmals in der Gattung Salix und Trichocarposid erstmals in Salix purpurea L. gefunden wurden. Die Isolierung von Minorkomponenten wie cis-1,2-Dihydroxycyclohexanderivaten lieferte die bisher unbekannte Substanz 6'-O-4-Hydroxybenzoylgrandidentin sowie Isograndidentatin A, eine bisher nicht beschriebene Substanz innerhalb der Gattung Salix. Darüber hinaus gelang erstmals die Gewinnung von Phelligrin A aus einer Salicaceae. Phelligrin A nimmt strukturell eine Zwischenstellung zwischen Flavanonen und Salicylalkoholderivaten ein und wird vermutlich durch symbiotische Effekte zwischen einem Rindenpilz und der Weide als Wirtspflanze synthetisiert.
Flavanonglucoside und deren acylierte Derivate stellen neben den Salicylalkoholderivaten die Hauptinhaltsstoffe des untersuchten Trockenextraktes dar. Laut Monographie des Europäischen Arzneibuches ist der Gehalt an Salicylalkoholderivaten das Hauptkriterium für die Qualität von Weidenrinde. Um die Qualitätsansprüche den neuesten wissenschaftlichen Untersuchungen anzupassen, werden Methoden zur Polyphenolbestimmung von Weidenextrakten benötigt. Deshalb wurde in einem ersten Schritt eine Methode zur Quantifizierung von Naringenin und Naringeninderivaten, den Hauptvertretern der Weidenrindenflavonoide, entwickelt und validiert. Es handelt sich um eine quantitative hydrolytische Spaltung der Naringeninderivate, die teilweise durch Trifluoressigsäure erfolgt und durch enzymatische Deglucosidierung vervollständigt wird. In Anlehnung an die Gehaltsbestimmung von Ginkgo biloba werden die zu Aglyca umgesetzten Flavanonderivate durch hochleistungsflüssigkeitschromatographische Analyse gemeinsam als Naringenin erfasst. Der Gehalt des Gesamtextraktes an Naringeninderivaten beträgt nach dieser
Methode 13,6 ± 0,5%, berechnet als Naringeninmonoglucosid.
Weiterhin wurden zellbiologische Untersuchungen durchgeführt. Im ICAM-1 (intercellular adhesion molecule-1) -Assay wird das antiinflammatorische Potential von Substanzen untersucht, die ICAM-1-Expression auf stimulierten mikrovaskulären Endothelzellen zu unterdrücken. Die Aktivität des Gesamtextraktes konnte größtenteils auf die Ethylacetatfraktion und den darin enthaltenen NF-κB-Hemmstoff Catechol zurückgeführt werden. Catechol ist als Abbauprodukt bestimmter Salicylalkoholderivate im untersuchten Gesamtextrakt zu etwa 2% enthalten. Weitere isolierte Substanzen wurden ebenfalls untersucht, wobei glucosidische Verbindungen keine Aktivität aufwiesen. Eine deutliche Hemmung bewirkte außerdem das Flavanon Eriodictyol. Es konnte gezeigt werden, dass Catecholstrukturelemente die Aktivität von Molekülen im ICAM-1-Assay erhöhen. Um Hinweise auf die Bioverfügbarkeit der im Extrakt enthaltenen phenolischen Substanzen zu erhalten, wurde die Resorption der Ethylacetatfraktion im Caco-2-Zellkulturmodell untersucht. Lediglich Catechol und Naringenin waren nach 90 min im basolateralen Kompartiment in quantifizierbaren Konzentrationen detektierbar (Papp= 46,2 ± 4,6 bzw. 23,3 ± 5,9 x 10-6 cm/s). Demgegenüber wurde die Resorption der Einzelsubstanzen betrachtet (Papp= 39,4 ± 3,2 bzw. 34,2 ± 3,0 x 10-6 cm/s). Für Catechol wurden erstmals Permeabilitätskoeffizienten beschrieben. Statistisch signifikante Unterschiede, die für eine erhöhte Resorption der Substanzen infolge positiver Matrixeffekte durch die Ethylacetatfraktion sprechen, waren in diesem Testsystem bei Naringenin, jedoch nicht bei Catechol festzustellen.
Die Bedeutung von Catechol und Flavonoiden mit Catecholstrukturelement an der antiinflammatorischen Wirkung von Weidenrindenextrakten sowie deren Resorption und Metabolismus, bieten Ansatzpunkte für weitere Untersuchungen. Aufgrund des hohen Catecholgehaltes dieses Extraktes werden zudem Studien zur chronischen Toxizität von Weidenrindenextrakten dringend benötigt.

Willow bark preparations hold a long tradition as well-tolerated herbal remedies for pain and fever. For the longest time, the phenolic glucoside salicin was regarded as the main active principle. However, recent studies indicate that salicin alone cannot be responsible for the observed antiinflammatory effects. Therefore, mainly the attendant polyphenols gained scientific attention. This study entailed the identification and quantification of prominent flavonoids of Salix purpurea L. In addition, possible modes of action as well as the absorption of a polyphenol-rich fraction were investigated in vitro.
The standardized willow bark extract STW 33-I (S. purpurea L., DER 16-23:1, 23-26% total salicin content, extracting agent: water) was subjected to liquid-liquid partition and subsequent phytochemical characterization. The largest amount of flavonoids was found in the ethyl acetate fraction. Twenty compounds, including flavanones and flavanonols, their glucosides, along with chalcone glucosides and corresponding coumaric derivatives were isolated and their structures elucidated. The stereochemical assignment of the epimeric 6''-O-p-trans-coumaroyl-(2S/2R)-naringenin-5-O-β-D-glucosides was determined. The ethyl acetate fraction yielded salicylic alcohol derivatives and simple phenolic compounds of which populoside B was detected in Salix and trichocarposide in S. purpurea L. for the first time. The isolation of minor compounds like cis-1,2-dihydroxycyclohexane derivatives yielded the hitherto unknown compound 6'-O-4-hydroxybenzoylgrandidentin, as well as isograndidentatin, a compound previously not observed within Salix genera. Furthermore, phelligrin A was isolated for the first time from a Salicaceae. This molecule combines structural elements of a salicylic alcohol and a flavonoid. It is synthesized most probably due to a symbiosis between a wood decaying fungus and its willow tree host.
Together with salicylic alcohol derivatives, flavanone glucosides and their acylated derivatives represent the principal compounds of the investigated dry extract. According to the monograph of the European Pharmacopeia the content of salicylic alcohol derivatives is the main criterion determining the pharmaceutical quality of willow bark preparations. With regard to the possible contribution of the polyphenols to the effect, a procedure for quantification of the principal flavonoid naringenin and its derivatives was developed and validated. First, most of the glucosides were cleaved by acid hydrolysis followed by an enzymatic deglucosidation of the remaining derivatives. Adapting the general idea of the procedure used to assay the flavonoid glycosides of Ginkgo biloba leaves (Ph. Eur.), naringenin glucosides and their corresponding chalcones were quantified as aglycones by high pressure liquid chromatography. The content of naringenin derivatives in the dry extract was determined to be 13.6 ± 0.5% (m/m), calculated as naringenin monoglucoside.
Further, cell biological investigations were carried out. The ICAM-1 (intercellular adhesion molecule-1) assay evaluates antiinflammatory potential of substances by determination of the suppression of ICAM-1 expression on the surface of stimulated microvascular endothelial cells. The major activity of the whole extract was found in the ethyl acetate fraction and assigned in large part to the simple phenol and NF-κB-inhibitor catechol. This compound is a degradation product of some salicylic alcohol derivatives and is present in the extract to an extent of approximately 2%. Isolated glucosidic compounds were shown to be inactive. A considerable inhibition of ICAM-1 expression was caused by the flavanone eriodictyol. It was demonstrated, that catechol moieties enhance the activity of compounds in the ICAM-1 assay.
In order to investigate the bioavailability of willow bark flavonoids in vitro, the absorption and transport of the ethyl acetate fraction was determined in a caco-2 cell culture model. Only catechol and naringenin were detected in quantifiable amounts in the basolateral compartment after 90 min (Papp= 46.2 ± 4.6 and 23.3 ± 5.9 x 10-6 cm/s, respectively). The transport of each single compound (Papp= 39.4 ± 3.2 and 34.2 ± 3.0 x 10-6 cm/s, respectively) was determined. An apparent permeability coefficient for catechol was determined for the first time. An increase in transport velocity due to matrix effects of the ethyl acetate fraction was demonstrated for naringenin, but not for catechol at a statistically significant level.
The relevance of catechol and flavonoids with catechol moieties, their contribution to the antiinflammatory properties of willow bark extracts, as well as their absorption and metabolism represent interesting fields for future research. Moreover, due to the relatively high content of catechol, data regarding the in vivo metabolism of this compound are needed.
