In humans, neuropeptide Y (NPY), a linear 36-amino acid peptide abundantly present in the CNS and PNS, is involved in the regulation of numerous physiological processes. NPY exerts its actions by activation of four G protein-coupled receptors constituting the neuropeptide Y receptor family (Y1R, Y2R, Y4R, Y5R). Several studies revealed an overexpression of Y1Rs in different malignant tumors with the highest levels found in breast cancer tissue, suggesting the Y1R as a potential target for tumor imaging and cancer therapy. Therefore, fluorescence and radiolabeled ligands with high selectivity and affinity for the Y1R are needed as tool compounds for the development of functionalized Y1R ligands to study Y1R expression in cells and tissues, and as tumor imaging agents themselves.
This work focused on the design of novel molecular tools for the Y1R with markedly improved binding affinity compared to previously reported ligands. The approach was guided by the reported X-ray crystal structure of the Y1R in complex with the high-affinity Y1R antagonist UR-MK299, which represents an Nω-carbamoylated derivative of the argininamide BIBP3226. According to the crystal structure, the introduction of bulky substituents at the guanidino group in BIBP3226 is incompatible with the binding mode, but the attachment of large substituents to the diphenylacetyl moiety of the argininamides should be well tolerated. Therefore, derivatives of UR-MK299 (2.14 and 2.30) and BIBP3226 (3.6), containing a 4-aminobutoxy substituent at one of the phenyl rings, were synthesized and the absolute stereochemistry of the generated second chiral center was determined. The obtained amine precursors exhibited similar binding affinity (pKi = 10.30-10.89) as UR-MK299 (pKi = 10.11). Conjugation of (S,R)-2.14 and (R,R)-2.14 to different fluorophores yielded a set of fluorescently labeled Y1R antagonists (2.35-2.39) with subnanomolar affinities (pKi = 9.37-9.95) and excellent Y1R selectivity (at least 1000-fold over Y2R, Y4R and Y5R). Flow cytometric saturation binding assays performed with intact MCF-7-Y1 mammary carcinoma cells afforded dissociation constants (pKD = 9.57-9.87), which were in agreement with the pKi values obtained from radioligand competition binding studies. The characterization of the 5’-TAMRA-labeled ligand 2.39 in fluorescence anisotropy-based binding assays, which take into account ligand depletion, suggested an even higher Y1R affinity (pKD = 11.02). The binding kinetics of the fluorescent probes at the Y1R were also studied by flow cytometry (2.35, 2.37 and 2.39) and fluorescence anisotropy (2.39). Furthermore, 2.37 and 2.39 were successfully used to visualize Y1R receptors in the plasma membrane of live MCF-7-Y1 cells using widefield and TIRF microscopy.
Prompted by the promising results of the fluorescent ligand approach, the amine-functionalized precursors were also used to synthesize potential Y1R PET ligands, exhibiting higher Y1R affinity and increased hydrophilicity than previously reported argininamide-type Y1R PET ligands. The PET ligand candidates were obtained by attachment of a fluoroglycosyl moiety via click chemistry (3.11, 3.12, 3.19 and 3.22) or by conjugation to a DOTA chelator and subsequent incorporation of Ga3+ (3.25). As anticipated, the potential PET ligands displayed high Y1R binding (pKi = 8.87-10.20). Radiosyntheses were performed to afford the PET-tracers [18F]3.11, [18F]3.12 and [68Ga]3.25, which were studied in nude mice with subcutaneous MCF-7-Y1 tumors (PET imaging, biodistribution). All PET ligands enabled the imaging of the tumor by PET with [68Ga]3.25 representing the most promising tracer due to a pronounced renal clearance.
To obtain high-affinity homobivalent Y1R antagonists (4.17-4.20, pKi = 9.67-10.13) as potential tools to study receptor homodimerization, amine precursor (S,R)-2.14 was dimerized by conjugation to spacers with varying lengths (8, 15, 25 and 37 atoms). Notably, the radioligand competition binding data, specifically the slopes of the sigmoidal displacement curves, did not indicate binding to Y1R homodimers.
Finally, a synthetic route towards argininamide-type potential “radiohybrid” Y1R ligands was explored, which will support the future development of theranostic agents targeting the Y1R.
In conclusion, this work presents the structure-guided design of amine-functionalized argininamide-type Y1R ligands that could be converted to fluorescently labeled ligands, PET ligands and homobivalent ligands displaying subnanomolar Y1R affinity and excellent Y1R selectivity. Based on the presented approach, a broad variety of specifically functionalized tools compounds for the Y1R, including imaging agents for cancer diagnosis, will be accessible in future studies.

Neuropeptid Y ist ein aus 36 Aminosäuren bestehendes, im Menschen im zentralen und peripheren Nervensystem exprimiertes Peptid, das an der Regulierung von zahlreichen physiologischen Prozessen beteiligt ist. Die Signale werden durch die Aktivierung von vier G-protein gekoppelten Rezeptoren der Neuropeptid Y-Rezeptorfamilie erzeugt (Y1R, Y2R, Y4R und Y5R). Studien zeigten eine Überexprimierung von Y1Rs in diversen malignen Tumorgeweben mit den höchsten Expressionsraten in Mammakarzinomen, weswegen der Y1R als potenzielles Target für Imaging und Therapie von Brustkrebs diskutiert wird. Um Y1R-Liganden als Tumordiagnostika entwickeln zu können, sind Fluoreszenz- und Radioliganden mit hoher Affinität und Selektivität für den Y1R vonnöten, um die Y1R-Expression in Zellen und Geweben untersuchen zu können.
Diese Arbeit konzentrierte sich auf das Design neuartiger molekularer Werkzeuge für den Y1R mit verbesserter Bindungsaffinität verglichen mit zuvor beschriebenen Liganden. Dabei stellte eine kürzlich veröffentlichte Röntgenkristallstruktur des Y1Rs im Komplex mit dem hochaffinen Y1R-Antagonisten UR-MK299 – ein Nω-carbamoyliertes Derivat des Argininamids BIBP3226 – die Grundlage für den Ansatz.
Die Kristallstruktur zeigte, dass eine Einführung von sperrigen Resten an der Guanidinogruppe von BIBP3226 nicht mit dem Bindungsmodus in Einklang zu bringen war, wohingegen deren Anknüpfung an der Diphenylacetyl-Gruppe gut toleriert werden sollte.
Daher wurden Derivate von UR-MK299 (2.14 and 2.30) und BIBP3226 (3.6) hergestellt mit einem 4-Aminobutoxy-Rest an einem der Phenylringe. Weiterhin wurde die absolute Stereokonfiguration am dadurch erzeugten zweiten Chiralitätszentrum bestimmt.
Die aminofunktionalisierten Vorstufen wiesen mit UR-MK299 (pKi = 10.11) ähnliche Bindungsaffinitäten auf (pKi = 10.30-10.89). Die Kopplung von (S,R)-2.14 und (R,R)-2.14 an diverse Fluorophore resultierte in einer Reihe von fluoreszenzmarkierten Y1R-Antagonisten (2.35-2.39) mit subnanomolaren Bindungsaffinitäten (pKi = 9.37-9.95) und exzellenter Selektivität für den Y1R (> 1000-fach gegenüber Y2R, Y4R und Y5R).
Sättigungsexperimente am Durchflusszytometer wurden an intakten MCF-7-Y1-Mammakarzinomzellen durchgeführt und ergaben pKD-Werte, die gut mit den pKi-Werten aus den Radioligand-Verdrängungsexperimenten übereinstimmten (pKD = 9.57-9.87).
Die Charakterisierung des 5’-TAMRA-gelabelten Liganden 2.39 in Fluoreszenzanisotropie-Assays, welche Ligandendepletion berücksichtigen, deutete auf eine noch höhere Bindungsaffinität hin (pKD = 11.02).
Die Bindungskinetiken der Fluoreszenzliganden wurden mit Durchflusszytometrie (2.35, 2.37 and 2.39) und Fluoreszenzanisotropie (2.39) untersucht. Weiterhin konnten durch Zugabe von 2.37 und 2.39 erfolgreich in Weitfeld- und TIRF-Mikroskopieexperimenten membranständige Y1R-Rezeptoren in MCF-7-Y1-Zellen visualisiert werden.
Angeregt durch diese vielversprechenden Ergebnisse wurden die aminofunktionalisierten Vorstufen auch dazu genutzt, potenzielle Y1R-PET-Liganden mit erhöhter Hydrophilie und verbesserter Bindungsaffinität im Vergleich zu zuvor beschriebenen Y1R-PET-Liganden des Argininamid-Typs synthetisiert.
Die potenziellen PET-Tracer konnten hergestellt werden durch Kopplung eines Fluoroglycosyl-Rests (3.11, 3.12, 3.19 and 3.22) oder durch Konjugation eines DOTA-Chelator, gefolgt vom Einschluss von Ga3+ (3.25).
Erwartungsgemäß wiesen die potenziellen PET-Liganden eine hohe Y1R-Bindungsaffinität (pKi = 8.87-10.20) auf. Die radioaktiv markierten PET-Liganden [18F]3.11, [18F]3.12 und [68Ga]3.25 wurden in Nacktmäusen mit subkutan transplantierten MCF-7-Tumoren hinsichtlich PET-Imaging und Biodistribution untersucht. Alle PET-Tracer ermöglichten die Visualisierung des Tumors im PET-Scan, wobei [68Ga]3.25 aufgrund der bevorzugt renalen Ausscheidung den aussichtreichsten Kandidaten darstellte.
Um hochaffine homobivalente Y1R-Antagonisten als potentielle Werkzeuge zur Untersuchung von Rezeptor-Homodimerisierung zu erhalten (4.17-4.20, pKi = 9.67-10.13), wurde (S,R)-2.14 durch Konjugation an unterschiedlich lange Spacer dimerisiert (8, 15, 25 und 37 Atome). Jedoch ergaben radiochemische Assays, im Speziellen die Steigungen der sigmoidalen Verdrängungskurven, kein Indiz auf eine Bindung an Y1R-Homodimere.
Zuletzt wurde eine Syntheseroute für potenzielle Y1R-‘Radiohybrid’-Liganden vorgestellt, die die künftige Entwicklung von an Y1Rs bindenden theranostischen Mitteln ermöglichen kann.
Zusammenfassend stellt diese Arbeit ein strukturbasiertes Design von aminofunktionalisierten Y1R-Liganden des Argininamid-Typs vor. Diese konnten für die Synthese von Fluoreszenzliganden, PET-Liganden und homobivalenten Liganden genutzt werden, die sich durch eine hohe Y1R-Affinität im subnanomolaren Bereich und eine herausragende Selektivität für den Y1R auszeichneten. Dadurch werden Syntheserouten für eine Vielfalt von funktionalisierten Werkzeugen für den Y1R ermöglicht, die auch die Entwicklung von Tumordiagnostika weiter unterstützen können.
