Doublecortin/DCX ist ein X-chromosomal kodiertes Protein, das während der Embryonalentwicklung des zentralen Nervensystems und während adulter Neurogenese in Säugetieren spezifisch in neuronalen Vorläuferzellen exprimiert wird. Das Protein ist nur vorübergehend in diesem Zelltyp und weder in Stammzellen, noch in adulten Nervenzellen nachweisbar. Dieses Expressionsmuster macht DCX zu einem einzigartigen Marker für Neurogenese. DCX stabilisiert Mikrotubuli, interagiert auch mit anderen Zytoskelett-Komponenten, verschiedenen Proteinkinasen sowie weiteren intrazellulären und membranständigen Molekülen. Darüber hinaus ist die molekulare Funktion Doublecortins jedoch weitgehend unklar. DCX-Mutationen führen beim Menschen zu neuronalen Migrationsstörungen, daraus resultieren während der Entwicklung charakteristische Gehirnfehlbildungen (Lissenzephalie / Bandheterotopie-Spektrum). DCX wird einerseits für neuronale Migration z.B. bei der Entstehung des zerebralen Kortex benötigt, findet sich andererseits aber in den kaum migrierenden, neuronal determinierten Zellen des Hippocampus, was auf zusätzliche Funktionen des Proteins über neuronale Migration hinaus hindeutet.
Die vorliegende Promotion beschreibt neue Aspekte zur Funktion von DCX und etabliert basierend auf dem Vorläuferzell-spezifischen Expressionsmuster den DCX-Promotor als Neurogenese-Reporter. Die Arbeit 1) zeigt die molekulare Funktionsweise des Proteins DCX anhand seiner Affinität zum Mikrotubuli-System im Spindelapparat; an Patienten beschriebene DCX-Mutationen setzten die Fähigkeit zur Mikrotubuli-Bündelung herab und bewirkten einen verminderten Einfluss von DCX auf Mitose. 2) identifiziert regulatorische Sequenzen des DCX-Gens und beschreibt deren spezifische Aktivität in neuronalen Vorläuferzellen in vitro. 3) etabliert mittels Expression von Reporterproteinen unter Kontrolle der DCX-regulatorischen Region ein Reportersystem für Neurogenese im Tiermodell; dieses Modell ermöglicht in Zukunft bildgebende Verfahren zur Analyse von Neurogenese, sowie weiterführende Untersuchungen zur Molekular- und Zellbiologie neuronaler Vorläuferzellen.

Translation of the abstract (English)

Doublecortin/DCX is encoded on the X-chromosome and specifically expressed in neuronal precursors of the developing and adult mammalian central nervous system. Doublecortin protein is only transiently expressed in neuronal precursor cells, but absent in stem cells and in adult neurons. Due to its specific expression pattern, attention was drawn to DCX as a marker of neurogenesis. DCX stabilizes the microtubule network inside the cells, has affinity to other cytoskeletal proteins besides microtubuli, has been shown to interact with different kinases and, moreover, with additional intracellularly located as well as with membrane bound molecules. Despite of these reported interactions, the exact molecular function of doublecortin is largely unknown. Doublecortin mutations result in aberrant migration of neuronal precursor cells in humans, entailing distinct malformations of the human brain taking place during development (lissencephaly/band heterotopia). Doublecortin is therefore necessary for the migration of neuronal precursors during the formation of the mammalian neocortex. In contrast, newly generated neurons in the adult hippocampus express DCX even if they hardly undergo any cellular migration, thereby suggesting a role of DCX in additional aspects of neurogenesis besides neuronal migration.
This dissertation describes new aspects concerning the molecular role of DCX and establishes DCX as a marker of neurogenesis, based on its specific expression in neuronal precursor cells. This dissertation 1) contains data on DCX affinity for the microtubule network of the mitotic spindle apparatus resulting in defective COS7 cell mitosis; DCX mutations identified in patients were shown to reduce DCX protein activity on mitosis. 2) identifies regulatory sequences of the DCX gene and shows their specific activity in neuronal precursors in vitro. 3) describes a transgenic mouse neurogenesis reporter system expressing reporter proteins under control of the DCX regulatory region; this model system might soon allow molecular imaging of neurogenesis and lead to further knowledge on molecular and cellular biology of neuronal precursor cells.
