Abstract (German)

Die Bedeckung cardiovaskulärer Bioprothesen mit autologen Endothel hat zum Ziel die Hämokompatibilität zu verbessern. Ziel dieser Arbeit ist es das Endothelialisierungspotential Glutaraldehyd-fixierter Herzklappenprothesen, sowie die zellulären Funktionen adhärenter Endothelzellen zu untersuchen. Weiterhin wurde der Einfluß einer zweizeitigen Besiedelung von humanen Fibroblasten und Endothelzellen auf deren fuktionelle Eigenschaften untersucht. Adhärenz und Morphologie der adhärierten Zellen wurde mittels Rasterelektronenmikroskopie und Immunhistochemie untersucht. Es konnte reproduzierbar eine vollständige Bedeckung von Glutaraldehyd-fixierten und detoxifizierten Herzklappenprothesen erreicht werden. Eine Analyse der direkt auf dem Material adhärenten Zellen ergab nicht-proliferative Zellen, die jedoch in der Lage waren auf inflammatorische Stimulation mit Interleukin-6 Sekretion, sowie mit Adhärenz inflammatorischer Zellen zu reagieren. Die Endothelzellen zeigten weiterhin erhaltene antithrombogene Eigenschaften, was mittels quantitativer Thrombozytenadhäsion und Sekretion von Prostacyclin gezeigt werden konnte. Eine Vorbesiedelung des Materials mit vaskulären Myofibroblasten gefolgt von Endothelzellen induzierte deren Proliferation und verbesserte inflammatorische sowie antithrombotische Eigenschaften.
Cardiovaskuläre Biomaterialien unterscheiden sich deutlich bezüglich des Potentials die Adhärenz humaner Endothelzellen zu ermöglichen. Erfolgreich endothelialisierte Biomaterialien zeigen zelluläre Eigenschaften die die klinische Effektivität dieser Materialien verbessern könnten. Zweizeitige Besiedelung ermöglicht regeneratives Potential und verbessert die zellulären Eigenschaften der adhärenten Endothelzellen.

Coverage of cardiovascular bioprostheses with autologous endothelium is used for the purpose of improving blood compatibility. Aim of our study was to analyze endothelialization potential of glutaraldehyde-fixed heart valves, cellular functions of seeded endothelial cells (EC) and the impact of a two-stage seeding protocol using human fibroblasts (FB) and EC from saphenous veins (HSVEC) on functional properties. Adherence and morphology of adhered cells were assessed by scanning electronic microscopy and immunohistochemistry. A reproducible complete coverage of the surface with EC was established on glutaraldehyde-fixed and decellularized bovine pericardium. Analyzing functional properties of directly adhered cells revealed non-proliferative cells, which were capable of maximum inflammatory stimulation in terms of increased interleukin-6 secretion and adhesion of inflammatory cells. Furthermore, EC showed sustained anti-thrombotic properties quantified by platelet adhesion onto EC and prostacyclin secretion of EC. Preseeding with vascular myofibroblasts using a two-stage seeding protocol resulted in induction of EC proliferation and improved inflammatory and anti-thrombotic functions. Cardiovascular biomaterials differ largely in their potential to allow adhesion of human EC. Successfully endothelialized biomaterial revealed cellular properties which might be favourable to improve performance of biomaterials. Two-stage seeding adds regenerative potential and improves cell functions of adherent EC.
