Abstract (English)

The synthesis of different cispentacin derivatives as well as of a conformational restricted gamma-amino acid was accomplished starting from furan methyl ester and the thereof derived trans-substituted gamma-butyrolactone respectively. The keystep in this synthetic approach is a [3+2]-nitrone-cycloaddition reaction affording two diastereomeric products which were subsequently transformed in a straightforward manner. These amino acids have a broad range of potential applications like in the field of medicinal chemistry as new antiviral agents (cispentacin analogues), in organocatalysis, as constricting elements in peptides and as building blocks for foldamers.

Furthermore, two diketopiperazine (DKP) esters with proline and the analogous amides were prepared. In this course, differently protected aspartate and serine derivatives were condensed to a DKP alcohol scaffold, which can be subsequently transformed to the corresponding DKP amine via a Mitsunobu reaction followed by Staudinger ligation. Adducts of these to DKPs with (L)-and (D)-proline were applied as novel organocatalysts in the conjugate addition of aldehydes and ketones to nitroolefines. In this course, the proline DKP amides were shown to be highly active and selective in the reaction of aldehydes. However, the addition of ketones was achieved in reasonable to good yields with comparably low stereoinduction.

Die Darstellung verschiedener Cispentacin-Derivate, als auch einer gamma-Aminosäure wurde ausgehend von Furancarbonsäuremethylester bzw. eines davon abgeleiteten gamma-Butyrolactons erreicht. Der Schlüsselschritt dieser Synthese war eine [3+2]-Nitron-Cycloaddition welche zwei diastereomere Produkte ergab, die hierauf zu den entsprechenden Zielverbindungen umgesetzt wurden. Diese Aminosäuren haben ein breites Spektrum möglicher Anwendungen, wie z.B. im Bereich der Wirkstoffchemie aufgrund der antifungizid wirksamen Cispentacin-Grundstruktur, in der Organokatalyse, als konformationsinduzierter Baustein in Peptiden sowie in der Synthese von Foldameren.

Des Weiteren, wurden zwei Diketopiperazin-(DKP)-Prolin-Ester als auch die entsprechenden Amide hergestellt. Hierzu wurden orthogonal geschützte Aspartat- und Serin-Derivate miteinander gekuppelt und zum DKP-Alkohol zyklisiert, welcher mit Hilfe einer Mitsunobu-Reaktion und anschließender Staudinger-Ligation zum entsprechenden DKP-Amin umgesetzt werden konnte. Die Addukte dieser DKP-Strukturen mit (L)- und (D)-Prolin wurde in ihrer Eigenschaft als potentielle neuartige Organokatalysatoren in der Addition von Aldehyden an Nitroolefine untersucht. Dabei stellten sich im Besonderen die DKP-Amide als äußerst aktiv wie auch selektiv heraus. Die Addition von Ketonen an Nitroolefine hingegen konnte zwar in guten Ausbeuten, jedoch nur mit geringer Stereoselektivität erreicht werden.
