Abstract (German)

Bei Norrin handelt es sich um ein sezerniertes Protein welches u.a. die Entwicklung von Kapillaren im Bereich der Retina und des Innenohres reguliert und welches in der Lage ist den Wnt/β-Catenin-Signalweg zu aktivieren. Beobachtungen an gentechnisch veränderten Mäusen zeigten darüber hinaus, dass Norrin auch einen protektiven Effekt auf retinale Ganglienzellen ausübt. Gegenstand dieser Arbeit war daher die Untersuchung einer möglichen neuroprotektiven Wirkung von Norrin auf geschädigte retinale Ganglienzellen. Hierfür wurde sowohl ein in vivo als auch ein in vitro Schadensmodell etabliert. In vitro zeigte sich nach der Differenzierung von immortalisierten retinalen Ganglienzellen (RGC-5) mittels Staurosporin und dem anschließenden Entzug von Serum und letztlich von Wachstumsfaktoren, dass die Zugabe von Norrin zu einer konzentrationsunabhängigen Zunahme der Anzahl der RGC-5 führt. In vivo stellte die intravitreale Injektion von 10 mM NMDA (N-Methyl-D-Aspartat) ein verwendbares Schadensmodell dar, welches zur exzitotoxischen Schädigung der retinalen Ganglienzellen führte und anhand dessen die neuroprotektive Wirkung von Norrin in vivo untersucht werden konnte. Die zusätzliche Injektion von Norrin führte dabei zu einem signifikanten Anstieg der überlebenden retinalen Ganglienzellen, welcher durch die Behandlung mit Dkk-1 (Dickkopf-1) fast vollständig aufgehoben werden konnte. Diese Ergebnisse legen den Schluss nahe, dass Norrin seine neuroprotektiven Eigenschaften überwiegend durch die Aktivierung des kanonischen Wnt/β-Catenin-Signalwegs vermittelt. Des Weiteren zeigte sich, dass Norrin eine gesteigerte Synthese von neuroprotektiven Faktoren, wie beispielsweise BDNF (Brain Derived Neurotrophic Factor), CNTF (Ciliary Neurotrophic Factor) oder FGF-2 (Fibroblast Growth Factor-2) induziert. Insgesamt konnte in der vorliegenden Arbeit gezeigt werden, dass Norrin einen bedeutenden neuroprotektiven Effekt auf das Überleben geschädigter retinaler Ganglienzellen besitzt.

Norrin is a secreted protein that controls the development of capillaries in retina and inner ear and ist able to activate the Wnt/β-Catenin-pathway. Furthermore this study provides evidence that Norrin has neuroprotective properties on retinal ganglion cells after excitotoxic damage. The function of Norrin was investigated in a mouse model of excitotoxic retinal ganglion cell damage after intravitreal injection of NMDA (N-Methyl-D-Aspartat) and in cultured immortalized retinal ganglion cells (RGC-5) treated with Staurosporin and cultured in growth factor-deprived serum-free culture medium. When Norrin was added to Staurosporin treated RGC-5 cultures, which kept in growth factor-deprived serum-free culture medium, the number of surviving cells significantly increased. This effect of Norrin was not dose dependent. Furthermore intravitreal injections of Norrin significantly increased the number of surviving retinal ganglion cell axons in the optic nerve and decreased apoptotic death of retinal ganglion cells following NMDA-mediated damage. This effect could be blocked by adding Dkk-1 (Dickkopf-1), an inhibitor of the Wnt/β-Catenin-pathway, indicating that the neuroprotective properties of Norrin are mediated via the Wnt/β-Catenin-pathway. Treatment of eyes with combined Norrin/NMDA increased the retinal expression of neurotrophic growth factors such as BDNF (Brain Derived Neurotrophic Factor), CNTF (Ciliary Neurotrophic Factor) and FGF-2 (Fibroblast Growth Factor-2). In conclusion Norrin has neuroprotective properties on retinal ganglion cells with the distinct potential to decrease the damaging effects of NMDA-induced retinal ganglion cell loss. The effects of Norrin involve activation of Wnt/β-Catenin signaling and induction of neurotrophic growth factors.
