Abstract (English)

Vision loss in hereditary retinal degeneration or glaucoma is caused by apoptotic death of photoreceptors or retinal ganglion cells (RGC), respectively. In animal models of both diseases, treatment with glucocorticoids (GC) has been shown to protect against neuronal cell death. We could show that 10 h of psychosocial stress induced by chronic subordinate colony housing (CSC) protects mouse photoreceptors from light-induced damage, a well-defined model to study the mechanisms of apoptotic photoreceptor cell death. The protective effects correlate with a marked elevation in plasma corticosterone (CORT). In addition, we will analyze the effects of 10 h and 19 d of CSC on RGC death after excitotoxic damage following treatment with N-methyl D-aspartate (NMDA). To clarify the neuroprotective role of GC in psychosocial stress, adrenalectomized mice will be treated similarly and analyzed. Finally, we will analyzed the activation of the phosphatidylinositol 3-Akt-kinase pathway, and the retinal expression of neurotrophic factors.

Sehverlust bei erblichen Netzhautdegenerationen oder beim Glaukom wird durch Apoptose von Photorezeptoren oder retinalen Ganglienzellen (RGC) hervorgerufen. Tiermodelle beider Krankheiten zeigen, dass Glucocorticoide (GC) protektive Effekte auf den neuronalen Untergang zeigen. Wir konnten beobachten, dass 10h psychosozialer Stress, induziert durch chronisch subordinierte Koloniehaltung (CSC), der zu einem hohen Spiegel von Corticosteron (CORT) im Plasma von Mäusen führt, im Tiermodell des Licht-induzierten Photorezeptorschadens protektive Wirkungen zeigt.. Schließlich haben wir den Einfluss von 10 h oder 20 d CSC auf den exzitotoxischen Tod von RGC nach Injektion von N-methyl D-Aspartat untersucht. Um die neuroprotektive Rolle von GC zu analysieren, haben wir die Experimente auf Tiere mit Adrenalektomie ausdehnent. Schließlich werden wir die Aktivierung des Phosphatidylinositol 3-Akt-Kinase Signalweges und die retinale Expression von neurotrophen Faktoren untersucht.
