Social anxiety disorder (SAD) and post-traumatic stress disorder (PTSD) are anxiety disorders characterized by excessive fear and avoidance of social situations and of stimuli associated with the trauma, respectively. The medication for SAD and PTSD is rather unspecific and, despite considerable efforts, their efficacy is unsatisfactory. The neuropeptide oxytocin (OT) has been suggested as a possible therapeutic agent for SAD and PTSD, as it facilitates a wide variety of social behaviors and decreases stress and anxiety in both humans and rodents.
This thesis describes the social and cued fear conditioning paradigms as animal models of SAD and PTSD, respectively, which were established to induce and study social and cued (non-social) fear in rodents. We describe the effects of central and local OT on social and non-social fear. Based on the results described in this thesis, we can conclude that OT represents a promising therapeutic approach for disorders associated with social deficits, such as SAD and possibly PTSD due to social trauma. However, caution is needed before recommending OT for the treatment of PTSD due to non-social trauma.

Sowohl Soziale Angsterkrankungen (SAD) als auch Post-traumatische Belastungsstörungen (PTSD) sind Angststörungen. Während SAD durch exzessive Angstzustände und der Vermeidung sozialer Situationen charakterisiert ist, zeichnet sich PTSD zum Einen durch die Vermeidung der Situationen, die ein Trauma herbeigeführt haben, als auch der Situationen, die an das Trauma erinnern, aus. Therapiemaßnahmen für SAD und PTSD sind relativ unspezifisch und die Effektivität meist nicht zufriedenstellend. Das Neuropeptid Oxytocin (OT) dient als mögliche therapeutische Maßnahme für SAD und PTSD, da es sowohl eine Bandbreite von sozialen Verhalten hervorruft, als auch das Stress- und Angstverhalten in Menschen und Nagetieren verringert.
Die hier vorliegende Arbeit beschreibt zwei verschiedene Tierversuchsmodelle (social fear conditioning, cued fear conditioning), welche etabliert wurden, um soziale und nicht-soziale (cued) Angst in Nagern zu induzieren und untersuchen. Die Effekte von zentraler und lokaler OT-Applikation auf soziale und nicht-soziale Angst werden hierin beschrieben. Basierend auf den Ergebnissen dieser Studie kann man schlussfolgern, dass OT einen vielversprechenden therapeutischen Ansatz für Erkrankungen, welche mit sozialen Defiziten (wie SAD und durch soziales Trauma hervorgerufene PTSD) einhergehen, darstellt. Jedoch sollte man mit Bedacht vorgehen, wenn man OT für die Behandlung von PTSD, welche durch nicht-soziale Traumata ausgelöst wurden, empfiehlt.
