In neurodegenerativen und autoimmunologischen Erkrankungen des zentralen Nervensystems und der Netzhaut werden verschiedene neurotoxische Prozesse beobachtet. Hierbei scheinen überaktivierte Mikrogliazellen eine erhebliche Rolle in der Pathogenese zu spielen. Mikrogliazellen präsentieren sich dabei mit einem zweischneidigen Wirkprofil. Mikroglia können zytotoxisch wirken und so degenerative und autoimmunologische Mechanismen unterstützen, aber anderseits besitzen sie eine Reihe von protektiven Funktionen. Im Rahmen einer auf Mikroglia gezielten pharmakologischen Therapie wäre also eine differenzierte Modulation dieser Zellen wünschenswert, die die schädlichen Wirkungen dämpft und die protektiven Eigenschaften fördert.
Interferon-β (IFNβ) wird schon länger als Medikament bei der Autoimmunerkrankung Multiple Sklerose verwendet. Die exakten Wirkmechanismen sind bis dato nicht komplett entschlüsselt, aber ein Teil der förderlichen Wirkung wird auf ein modulierendes Eingreifen in das überaktivierte Immunsystem zurückgeführt.
Ziel dieser Arbeit war herauszufinden welchen Effekt IFNβ auf Mikrogliazellen hat, und wie sich dieser auf transkriptioneller- und funktioneller Ebene äußert.
In dieser Arbeit konnte gezeigt werden, dass IFNβ in Mikroglia der BV-2 Zellinie und primären murinen Mikrogliazellen des Gehirns einen deutlichen Effekt zeigt. IFNβ aktiviert Mikrogliazellen durch die Induktion einer Vielzahl von, sowohl pro- wie auch antiinflammatorischen, Genen. Eine Steigerung der Phagozytoseaktivität durch IFNβ-Behandlung wurde ebenfalls beobachtet. Im Gegensatz zu LPS-stimulierten Mikroglia, welche gleichermaßen eine erhöhte Phagozytoseleistung zeigten, nahmen die Zellen aber durch die IFNβ-Stimulation keinen Schaden. Die Ergebnisse zur Änderung der Zellmorphologie ergaben einen ramifizierten, „protektiven“ Phänotyp für IFNβ-behandelte Mikrogliazellen, ein Effekt der sich auch bei gleichzeitiger Stimulierung mit LPS durchsetzen konnte. Zusammenfassend lassen die Ergebnisse im Kontext der aktuellen Literatur die Induktion eines alternativ aktivierten, potentiell protektiven Mikrogliaphänotyps durch IFNβ vermuten.
Darum stellt sich die Frage ob IFNβ nicht als potentielle Therapieoption bei neurodegenerativen und retinalen Erkrankungen mit Mikrogliaaktivierung in der Pathogenese in Betracht zu ziehen ist. Hier wäre insbesondere der Schritt zu in vivo Experimenten von großem Interesse, weil nur so die langfristigen Effekte von IFNβ beobachtet werden könnten. Aus der Behandlung von Multiple Sklerose-Patienten mit IFNβ weiss man, dass die Wirkung klinisch erst nach einiger Zeit einsetzt und auch die Änderungen auf Transkriptomebene längerfristig abweichend von den initialen Genexpressionsänderungen zu Beginn der Therapie sind. Mit Hinblick auf die besondere Ausrichtung dieser Arbeitsgruppe ist die Wiederholung der Ergebnisse in retinalen primären Mikrogliazellen ein weiterer wichtiger Schritt, insbesondere in Mausmodellen retinaler Degeneration.

Translation of the abstract (English)

In neurodegenerative and auto-immune disorders of the central nervous system and the retina various neurotoxic processes can be observed. In this hyper activated microglia cells seem to play a significant role in pathogenesis. Here microglia cells present themselves with a dual activity profile. On the one hand microglia can have cytotoxic effects and thereby facilitate degenerative and auto-immune mechanisms, but on the other hand they also have a number of protective functions. Therefore a differentiated modulation of these cells which lessens the damaging effects and enhances the protective characteristics would be desirable in the course of pharmacological therapy targeted at microglia.
Interferon-β (IFNβ) has been in use as medication for the auto-immune disorder multiple sclerosis for some time now. The exact modes of action have currently still not been decoded, nonetheless a portion of the beneficial effect has been traced back to modulating intervention in the hyper activated immune system.
The goal of this thesis was to find out the effects of IFNβ on microglia cells and how this presents on transcriptional and functional levels.
This thesis demonstrated that IFNβ has significant effects in microglia cells of the BV-2 cell line and primary murine microglia cells of the brain. IFN activates microglia cells via induction of a variety of pro- as well as anti-inflammatory genes. An increase in phagocytosis activity due to IFNβ-treatment was also observed. The cells were not damaged by the IFNβ-treatment in contrast to LPS-stimulated microglia, which showed equivalent increase in phagocytosis performance. The results regarding the changes in cell morphology showed a ramified "protective" phenotype for IFNβ-treated microglia cells, an effect which prevailed even with simultaneous stimulation with LPS. In summary the results, seen in the context of the current literature, suggest the induction of an alternatively activated, potentially protective microglia phenotype via IFNβ.
This poses the question whether IFNβ might be considered as potential therapy option for neurodegenerative or retinal diseases with microglia activation in pathogenesis. In this, the move to in vivo tests would be very interesting, because this is the only way to observe the long-term effects of IFNβ. Based on the treatment of multiple sclerosis patients with IFN one knows that the effects clinically only begin after some time and that the changes on transcriptome level differ from the gene expression changes at the start of the therapy for the long term. In regard to the specific focus of this work group a repetition of the results in retinal primary microglia cells is an important next step, in particular in mouse models of retinal degeneration.
