Abstract (German)

Das Ziel dieser Arbeit war es herauszufinden, ob es möglich ist, ein unbekanntes Füllungsmaterial zu analysieren und darin einzelne Bestandteile bzw. das Ausgangsprodukt zu identifizieren. Zuerst musste gezeigt werden, dass bekannten Füllungswerkstoffen typische Basismonomere zugeordnet werden können.
Damit die Ergebnisse gedeutet werden können, mussten zuerst bekannte, mögliche Bestandteile charakterisiert werden. Dazu wurden elf typische Basismonomere soweit gelöst und verdünnt, dass sie mittels einer chemischen Analysetechnik, der HPLC analysiert werden konnten. In meiner Arbeit werden dann von elf Füllungswerkstoffen die Monomere und die Füller isoliert und diese Monomere mit der HPLC analysiert.
Die isolierten Füllkörper wurden unter einem Rasterelektronenmikroskop (REM) betrachtet und ausgewertet.
Das Basismonomer Bis-GMA konnte in sieben von elf getesteten Kompositmaterialien nachgewiesen werden. Das Monomer Ethoxy Bis-MA wurde in drei untersuchten Füllungsmaterialien gefunden. Es zeigte sich, dass die Hersteller in allen ihren Produkten meist identische bzw. gering abgewandelte Monomerzusammensetzungen verwendeten.
Der große Unterschied der verschiedenen Produkte kam erst durch die unterschiedlichen Füllkörper zustande. Anhand der REM-Bilder konnten ebenfalls Füllungswerkstoffe zugeordnet werden. Dazu müssen aber alle für diesen Stoff charakteristischen Füllkörper auf dem Objektträger zu finden sein, damit sie abgebildet werden können.
Diese Arbeit hat gezeigt, dass es auf diese Art und Weise prinzipiell möglich ist, unbekannte Komposite zu erkennen. Um genauere Ergebnisse zu bekommen, müsste aber ein ausführliches Register von HPLC-Messungen und REM-Bildern aller Füllungsmaterialien unter den gleichen Parametern angelegt werden.

The aim of this investigation was to analyze monomer and filler components of different restorative composites. High-performance liquid chromatography (HPLC) was used to characterize the monomeric matrix and scanning electron microscopy (SEM) pictures were used to describe the inorganic phase of the restoratives.
HPLC reference runs were made for eleven isolated methacrylate monomers, giving standards for comparison of the components of the composites.In this work, the monomers and the fillers from eleven commercially available composites were than isolated and the monomers analyzed by the HPLC and compared with the standards. Filler were separated and the isolated particelswere observed in the scanning electron microscope (SEM) and analyzed.
Seven composites were tested positive forbis-GMA and three materials for ethoxybis-MA. It became apparent, that most manufacturers use identical monomers or small modified compositions for different materials of their portfolio.The main difference of the individual products was presented by the different filler variations. SEM-images of the filler particles therefore may help for further identifying of different composites.
This work is a first but limited assay identifying unknown composites with the help of standard HPLC and SEM analytics. Restrictions have to be made for the resolution of the used HPLC system, pureness and quality of the standard monomers, the solution and separation of the monomer systems or the manual differentiation of filler particles for SEM. For allowing a detailed differentiation, a more elaborate catalogue of HPLC measurements and SEM images of all composites should be provided.
