Ein wichtiger Kontrollmechanismus des Zellzyklus ist die irreversible Proteolyse von Zellzyklus-Regulatoren. Dabei markieren E3-Ligasen Zielproteine durch Ubiquitinmoleküle. Für den Abbau in der Mitose und der G1-Phase reguliert der APC/C-Komplex (Anaphase-Promoting-Complex/Cyclosome) als E3-Ligase den zeitlichen Verlauf des Zellzyklus. Die Aktivität des APC/C wiederum wird in den übrigen Zellzyklusstadien durch Phosphorylierung und durch Proteine der Rca1/Emi1-Proteinfamilie inaktiv gehalten.
Für eine vollständige APC/CFzr-Aktivität in der G1-Phase muss Rca1 seine inhibitorische Funktion auf den APC/CFzr verlieren. Es konnte gezeigt werden, dass Rca1 über einen anderen Mechanismus abgebaut wird als das Ortholog Emi1. Während Emi1 bereits in der Mitose SCFβTRCP-vermittelt abgebaut wird, ist bei Rca1 ein Abbau erst in der frühen G1-Phase zu erkennen.
Für die Identifizierung der Abbausequenz wurden in dieser Arbeit live-cell-imaging und Durchflusszytometrie verwendet. Dazu wurden Schneiderzellen transient mit Fluoreszenz-markierten Rca1-Deletions-Derivate transfiziert und die Stabilität in G1 näher untersucht. Auf diese Weise wurden eine Abbausequenz im N-terminalen Bereich sowie eine im zentralen C-terminalen Bereich von Rca1 identifiziert. Letztere beinhaltet neben einer für Rca1 neu entdeckten Linkerregion eine KEN-Box-Domäne, die als bekannte Erkennungssequenz für APC/CFzr-Substrate gilt und somit eine direkte Verbindung zum APC/CFzr-Komplex herstellt.
In verschiedenen Experimenten konnte gezeigt werden, dass der Rca1-Abbau mit der Aktivierung des APC/CFzr zusammen fällt. In der G1-Phase beginnt der Abbau von Rca1 zeitgleich mit anderen APC/C-Substraten. Eine verfrühte Aktivierung des APC/CFzr durch die Überexpression von Fzr führt ebenfalls zu einem zeitgleich beginnenden Abbau in der Interphase. Des Weiteren konnte gezeigt werden, dass die Stabilität von Rca1 direkt von seiner Fähigkeit abhängt, den APC/CFzr zu inaktivieren. Während ein aktives Rca1 seinen eigenen Abbau verzögern kann, wird der Abbau eines inaktiven Rca1 durch die Überexpression von Fzr beschleunigt.
Diese Ergebnisse lassen darauf schließen, dass Rca1 während der S- und G2-Phase ein Inhibitor des APC/CFzr ist, jedoch im Verlauf der G1-Phase zu einem Substrat des APC/CFzr wird.
Um einen endgültigen Nachweis zu erhalten, dass Rca1 sowohl Inhibitor als auch Substrat des APC/CFzr ist, wurde ein Drosophila-spezifischer, biochemischer in vitro-Ubiquitinylierungsassay etabliert. In diesem Assay konnte eine Ubiquitinylierung von APC/C-Substraten beobachtet werden und soll nun für die Analyse von Rca1 als Inhibitor des APC/C als auch als Substrat des APC/C eingesetzt werden.

Translation of the abstract (English)

For progression through the cell cycle, alternate degradation and accumulation of regulatory proteins are essential. Proteolysis of cell cycle regulators is initiated by E3 ligases that mediate polyubiquitination of substrate molecules. The anaphase-promoting complex/cyclosome (APC/C) is such an ubiquitin-ligase that is active during mitosis and G1-phase but needs to be inactive during S- and G2-phase. Inactivation of the APC/C involves proteins of the Emi1/Rca1 family. Inhibition of the APC/C by Rca1 has to be reversed to allow full APC/C activity. In the case of the vertebrate Emi1, this is achieved by its proteolysis during mitosis, initiated by SCFßTrcP dependent ubiquitination. Rca1 is stable throughout mitosis and gets rapidly degraded when cells enter the G1-phase, suggesting a different degradation mechanism.
In this work, live-cell-imaging and flow cytometry of transfected Drosophila tissue culture cells were used to identify the sequences as well as the mechanism responsible for the degradation of GFP- and Cherry-tagged Rca1-proteins. Two distinct degradation sequences could be identified in the N- and C-terminal part of Rca1, respectively. In the latter case, the KEN-box together with a newly identified linker-region is necessary for Rca1 degradation. The KEN-box is a well characterized recognition site of APC/CFzr-substrates and implies the involvement of APC/C in the degradation mechanism of Rca1. Several experiments could show that the degradation of Rca1 resembles the degradation of other substrates of the APC/CFzr-complex. This was seen in direct comparison to Geminin and CyclinB in G1 but also after APC/CFzr activation during interphase following overexpression of Fzr. Furthermore, it could be shown that the stability of Rca1 depends on its ability to inhibit the APC/CFzr. An active Rca1 can reduce its own degradation while the degradation of an inactive Rca1 can be speed up by a higher concentration of Fzr.
These findings suggest that Rca1 can be switched from an inhibitor of the APC/CFzr-complex during S- and G2-phase to a substrate of APC/CFzr in G1. This switch could be induced by a modification for example a phosphorylation as it is seen by the human ortholog Emi1.
In order to test biochemically, whether and how Rca1 can act as both inhibitor and substrate for APC/CFzr, an assay was established to analyse APC/C-dependent ubiquitination in vitro. This assay can now be used in further experiments to examine the way how Rca1 acts as an inhibitor and as a substrate for the APC/C.
