Ziel der vorliegenden Arbeit war, den Einfluss des TGF β - Signalwegs auf das Überleben Retinaler Ganglienzellen im adulten unbehandelten Tier und im NMDA - Schadensmodell zu untersuchen. Durch eine Konditionelle Deletion von Smad7 mithilfe der α - Cre - Maus konnte der inhibitorische Einfluss von Smad7 auf den TGF β - Signalweg in retinalen Neuronen und Müllerzellen entfernt und somit ein verstärktes TGF β - Signal erzielt werden.
Zum Ausschluss eines möglichen Einflusses der verwendeten Cre - Rekombinase selbst auf die Befunde, wurden α Cre - Mäuse mit ihren wildtypischen Geschwistertieren verglichen.
Die Auswirkungen des verstärkten TGF β - Signals auf Retinale Ganglienzellen wurden anschließend in acht Wochen alten Tieren und sechs Monate alten Tieren anhand von Axonzählungen der Nn. optici untersucht. In Sehnerven von Tieren mit einem verstärkten TGF β - Signal zeigte sich eine signifikant höhere Anzahl an Axonen, was auf einen neuroprotektiven Effekt des verstärkten TFG β - Signalwegs für Retinale Ganglienzellen schließen lässt. Zusätzlich wurde die retinale Gefäßsituation durch in vivo - Fundusaufnahmen und Fluoreszenzangiographien der Augen und die Funktion der Retinalen Ganglienzellen durch Elektroretinographien auf mögliche Effekte durch das erhöhte TGF β - Signal beurteilt. Hier zeigten sich keine signifikanten Veränderungen.
Zur Untersuchung einer neuroprotektiven Rolle des verstärkten TGF β - Signalwegs im adulten Tier wurde als Modell ein experimentell induzierter Netzhautschaden angewendet, der exzitotoxische Schaden von Retinalen Ganglienzellen durch Injektion von N Methyl – D Aspartat (NMDA). In diesem Modell konnte kein Hinweis auf eine neuroprotektive Rolle des verstärkten TGF β - Signalwegs für Retinale Ganglienzellen gezeigt werden.
Zusammenfassend kann gesagt werden, dass eine zelltypspezifische Konditionelle Deletion des inhibitorischen Smad7 - Proteins in retinalen Neuronen und Müllerzellen Retinale Ganglienzellen nicht vor einem NMDA - induzierte Exzitotoxischen Zelltod schützt. Jedoch schützt das verstärkte TGF β - Signal Retinale Ganglienzellen während der Entwicklung vor Apoptose, was sich letztlich in einer signifikant höheren Anzahl von Axonen im N. opticus des adulten Tieres widerspiegelt.

Translation of the abstract (English)

The role of TGF-β signaling in the eye is complex, as pro- and anti-apoptotic properties have been described. Recently, we demonstrated that mice with an increased TGF-β signal in cells derived from the inner layer of the optic cup were protected against developmental programmed cell death (Braunger et al., J Neurosci. 2013). To learn more about the role of TGF-β signaling in retinal neuroprotection in the adult animal, we used mice with a conditional deletion of the inhibitory mothers against decapentaplegic homolog 7 (Smad7) in cells derived from the inner layer of the optic cup.
The animals showed a significant increase in the amounts of phosphorylated Smad3 indicating an elevated activity of TGF-β-signaling. Furthermore, adult mice carrying a conditional deletion of Smad7 showed a significant higher number of retinal ganglion cells axons, as determined after morphometric analysis of optic nerve sections, indicating a higher number of retinal ganglion cells. However, the higher number of RGCs did not result in an improved retinal functional as evidenced by electroretinography.
To challenge the survival of retinal ganglion cells in adult animals, we experimentally induced apoptosis of retinal ganglion cells by intravitreal injections of N methyl D aspartate (NMDA). The procedure resulted in a significant decrease in the number of retinal ganglion cell axons in the NMDA injected samples. However, we could not determine a difference in the number of the remaining retinal ganglion cell axons in NMDA injected mice carrying a conditional deletion of Smad7 compared to NMDA injected controls.
Taken together we provide evidence that a cell type specific conditional deletion of the inhibitory Smad7 protein in cells derived from the inner layer of the optic cup does not protect retinal ganglion cells from NMDA induced excitotoxic cell death. On the other hand, an increased TGF-β-signaling serves an important role during developmental programmed cell death in the mouse retina, resulting in an increased number of retinal ganglion cells in the adult animal.
