Despite continuing efforts to find new therapeutic strategies for glioblastoma, the prognosis of affected patients remains poor. Tumor recurrence is an unfortunate reality, largely due to the malignant nature of glioblastoma including its intrinsic heterogeneity and immunosuppressive microenvironment. The urgent need for innovative therapeutic approaches and new molecular targets has brought attention to the 18 kDa translocator protein, TSPO. This mitochondrial protein is overexpressed in glioblastoma and during neuroinflammation. Functionally, TSPO has been linked to apoptosis, mitochondrial metabolism, and immunomodulation, however its role in glioblastoma remains enigmatic. Intriguingly, dysregulation of these potential TSPO functions drives glioblastoma therapy resistance and tumor progression.
Therefore, we hypothesized that TSPO plays a pivotal role in these malignant traits of glioblastoma. The overarching goal was to gain a functional understanding of TSPO in glioblastoma with a particular focus on its involvement in tumor-intrinsic mechanisms following cytotoxic T cell attacks. This includes tumor cell apoptosis and the secretion of immunomodulatory cytokines in response to T cell effector molecules. Additionally, the impact of TSPO on mitochondrial energy metabolism, which is crucial for glioblastoma growth, was explored. To address this critical research question, BTIC13 patient-derived mesenchymal glioblastoma cells underwent a wide array of molecular and functional analyses, including state of the art genetic loss-of-function approaches, comprehensive examination of apoptosis pathways, multilayer examination of pro-inflammatory cytokine effects, and analysis of mitochondrial respiration.
Key findings of this thesis include the selective upregulation of TSPO in tumor cells in response to direct contact with T cells and following exposure to the pro-inflammatory cytokines IFNγ and TNFα. The death ligand TRAIL has been identified as potential mechanism through which cytotoxic T cells induce extrinsic and intrinsic apoptosis in BTIC13. However, the TRAIL-induced apoptotic response of BTIC13 did not solely depend on TSPO expression but was rather influenced by a multitude of tumor cell-intrinsic mechanisms in monoculture conditions. Additionally, TSPO appeared to play no substantial role in the induction of the immune-modulatory cytokines IL-6 and IL-8 by BTIC13 in response to IFNγ and TNFα stimulation. However, TSPO-deficiency significantly impaired cancer cell migration and elevated maximal mitochondrial respiration and reactive oxygen production.
Taken together, this thesis positions TSPO as a potential marker for effector T cell activity and OXPHOS regulation in glioblastoma. However, concrete conclusions regarding TSPO’s potential as a therapeutic target remain elusive at this point. The established methodology and results presented here set the groundwork for unravelling a putatively tightly regulated role of TSPO in glioblastoma and serve as a basis for future experiments.

Trotz der Bemühungen um neue Therapiestrategien für Glioblastoma bleibt die Prognose der betroffenen Patienten nach wie vor düster. Tumorrezidive sind eine bedauerliche Realität, die größtenteils auf die bösartige Natur des Glioblastoms, einschließlich seiner intrinsischen Heterogenität und seiner immunsuppressiven Mikroumgebung, zurückzuführen sind. Der dringende Bedarf an innovativen Therapieansätzen und neuen molekularen Zielen hat die Aufmerksamkeit auf das 18 kDa Translocator Protein TSPO gelenkt. Dieses mitochondriale Protein wird in Glioblastoma und bei der Neuroinflammation übermäßig stark exprimiert. Funktionell wird TSPO vor allem mit der Apoptose, dem mitochondrialen Stoffwechsel und der Immunmodulation in Verbindung gebracht, doch die Funktion im Glioblastom ist noch nicht hinreichend erforscht. Interessanterweise ist die Fehlregulation der potenziellen TSPO-Funktionen maßgeblich an der Therapieresistenz und der Progression des Tumors beteiligt.
Die Grundlage der vorliegenden Arbeit bildete die Hypothese, dass TSPO eine zentrale Rolle bei diesen malignen Eigenschaften des Glioblastoms spielt, und das übergeordnete Ziel war es, ein funktionelles Verständnis von TSPO im Glioblastom zu erlangen. Dabei lag ein besonderer Fokus auf der Beteiligung von TSPO an tumoreigenen Mechanismen nach einem zytotoxischen T-Zell-Angriff wie zum Beispiel der Tumorzell-Apoptose und der Sekretion immunmodulatorischer Zytokine als Reaktion auf T-Zell-Effektor Moleküle. Darüber hinaus wurden die Auswirkungen von TSPO auf den mitochondrialen Energiestoffwechsel untersucht, welcher für das Wachstum des Glioblastoms mit entscheidend ist. Im Rahmen dieser Arbeit wurde die vom Patienten stammende, mesenchymale BTIC13-Glioblastom Zelllinie einer breiten Palette von Analysen unterzogen. Dazu gehörten der genetische Knockdown und Knockout von TSPO in BTIC13, die umfassenden Untersuchungen der Apoptose Wege, die mehrschichtigen Untersuchungen der Auswirkungen von proinflammatorischen Zytokinen und Analysen der mitochondrialen Atmung.
Nach direktem Kontakt mit zytotoxischen T-Zellen oder nach der Stimulation mit den proinflammatorischen Zytokinen IFNγ und TNFα, wurde TSPO selektiv in den Tumorzellen hochreguliert. Der Todesligand TRAIL wurde als potenzieller Mechanismus identifiziert, durch den zytotoxische T-Zellen die extrinsische und intrinsische Apoptose in BTIC13 auslösen. Die Empfindlichkeit gegenüber der TRAIL-induzierten Apoptose von BTIC13 hing jedoch nicht allein von der TSPO-Expression ab, sondern wurde unter Monokulturbedingungen durch eine Vielzahl intrinsischer Resistenzmechanismen der Tumorzellen beeinflusst. Darüber hinaus schien TSPO keine wesentliche Rolle bei IFNγ- und TNFα-stimulierten Induktion und Sekretion der immunmodulatorischen Zytokine IL-6 und IL-8 zu spielen. Knockdown und Knockout von TSPO in BTIC13 beeinträchtigten die Migration von Krebszellen erheblich und erhöhten die maximale mitochondriale Atmung sowie die Produktion von reaktiven Sauerstoffspezies.
Zusammenfassend positioniert diese Arbeit TSPO als potenziellen Marker für die Aktivität von Effektor-T-Zellen und der OXPHOS Regulation im Glioblastom. Dennoch bleiben konkrete Schlussfolgerungen hinsichtlich des therapeutischen Potenzials von TSPO zum jetzigen Zeitpunkt aus. Die hier vorgestellte Methodik und die Ergebnisse legen jedoch den Grundstein für die Aufklärung einer vermutlich streng regulierten Rolle von TSPO beim Glioblastom und dienen als Grundlage für zukünftige Experimente.
