Aus humanen induzierten pluripotenten Stammzellen ausdifferenzierte retinale Organoide bilden die neuronale Retina in wichtigen Bereichen in ihrer typischen Schichtung ab. Das retinale Pigmentepithel (RPE) wird in den bestehenden Differenzierungsprotokollen für retinale Organoide jedoch lediglich ektopisch als kugelförmige, mehrschichtige Struktur ausgebildet. Sowohl die Reifung als auch die Funktionalität der neuronalen Retina wird in vivo aber entscheidend vom direkten Kontakt des RPE zu den Photorezeptoraußensegmenten beeinflusst. Die Rekonstruktion der wichtigen physiologischen Beziehung zwischen RPE und Photorezeptoren in vitro wäre deshalb der nächste Schritt, um ein präziseres Modell der Netzhaut für die Forschung zu etablieren.
Am Institut für Humangenetik Regensburg wurde das existierende Differenzierungsprotokoll für retinale Organoide so angepasst, dass zusätzlich zu den retinalen Organoiden als Nebenprodukt RPE (bRPE) gewonnen werden kann. Die gleichzeitige Ausdifferenzierung von bRPE und retinalen Organoiden über ein einziges Protokoll spart erheblich Ressourcen und Zeit, es garantiert aber auch eine identische epigenetische Herkunft der verschiedenen Zelltypen.
Im ersten Teil der vorliegenden Arbeit sollte die Reproduzierbarkeit dieses Protokolls überprüft werden. Durch eine fungale Kontamination von 4 der 5 bRPE-Zelldifferenzierungen konnte dies allerdings nur begrenzt bestätigt werden.
Im zweiten Teil sollten die in einer Vorarbeit bereits differenzierten bRPE-Zellen näher charakterisiert werden. Dazu wurde für diese Zellen die Expression von verschiedenen RPE-Marker-Genen mithilfe der qRT-PCR Analyse und die Proteinexpression mithilfe der Immunzytochemie untersucht. Weiterhin wurde der transepitheliale elektrischen Widerstand und die Fähigkeit zur Phagozytose von Photorezeptor-Außensegmenten bestimmt. In allen durchgeführten Experimenten zeigten die bRPE-Zellen charakteristische Eigenschaften von RPE-Zellen, jedoch mit einer hohen Variabilität zwischen den einzelnen bRPE-Differenzierungen.
In Hinblick auf eine spätere Co-Kultur der retinalen Organoide mit den bRPE-Zellen wurde im dritten Teil der Arbeit der Einfluss des Mediums der retinalen Organoide auf die bRPE-Zellen untersucht. Hier zeigte sich, dass die Kultivierung der bRPE-Zellen im Medium für die retinalen Organoide über 3 Wochen einen signifikanten Abfall der TEER-Messwerte und eine signifikant verringerte Expression des RPE-Marker-Gens RPE65 verursachte. Die Kultivierung der bRPE-Zellen im Standardmedium nach Krohne et al. (2012) und eine vorangegangene Kryokonservierung schienen dagegen keinen Einfluss auf die Eigenschaften der bRPE-Zellen zu nehmen. Um bei einer Co-Kultur ein möglichst physiologisches bRPE garantieren zu können, sollte in künftigen weiteren Studien überprüft werden, ob eine schnellere oder langsamere Umstellung der bRPE-Zellen auf das Medium der retinalen Organoide oder alternativ der Zusatz von Nicotinamid-Mononukleotid eine positive oder negative Auswirkung auf die bRPE-Zellen zeigt.

Translation of the abstract (English)

Retinal organoids differentiated from human induced pluripotent stem cells recapitulate the typical laminar organization of the neural retina in key areas. However, in current differentiation protocols for retinal organoids, the retinal pigment epithelium (RPE) is only formed ectopically as a spherical, multilayered structure. In vivo, both the maturation and functionality of the neural retina are critically dependent on direct contact between the RPE and the photoreceptor outer segments. Reconstructing this essential physiological relationship between RPE and photoreceptors in vitro would therefore represent an important step toward establishing a more accurate retinal model for research purposes.
At the Institute of Human Genetics in Regensburg, the existing differentiation protocol for retinal organoids was modified to additionally yield RPE cells as a byproduct alongside retinal organoids (bRPE). This simultaneous differentiation of bRPE and retinal organoids using a single protocol not only saves considerable time and resources but also ensures an identical epigenetic origin of the different cell types.
The first part of this study aimed to evaluate the reproducibility of the modified differentiation protocol. Due to fungal contamination in 4 out of 5 bRPE differentiations, this could only be confirmed to a limited extent.
The second part focused on a detailed characterization of bRPE cells that had been previously differentiated in earlier work. For this purpose, the expression of various RPE marker genes was analyzed using qRT-PCR, and protein expression was assessed through immunocytochemistry. Additionally, transepithelial electrical resistance (TEER) and the ability to phagocytose photoreceptor outer segments were evaluated. In all experiments, the bRPE cells showed characteristic features of RPE cells, though with considerable variability between individual bRPE differentiations.
With a future co-culture of retinal organoids and bRPE cells in mind, the third part of this study investigated the effect of the retinal organoid medium on bRPE cells. Culturing bRPE cells in retinal organoid medium for three weeks led to a significant decrease in TEER values and a significantly reduced expression of the RPE marker gene RPE65. In contrast, culturing the cells in standard medium according to Krohne et al. (2012), as well as prior cryopreservation, appeared to have no adverse effects on bRPE properties. To ensure a physiologically functional bRPE in future co-culture systems, further studies should investigate whether a more gradual or more rapid transition to the retinal organoid medium—or alternatively, supplementation with nicotinamide mononucleotide—has a positive or negative impact on bRPE cell properties.
