Abstract (English)
This thesis was focused on the interactions of Layer-by-Layer (LbL) thin films with biological counterparts during a drug delivery scenario. Several physiological barriers of an organism, the adsorption of serum proteins, the cellular association or the barrier function of biological hydrogels, limit the transport efficacy and hence the therapeutic effect of a drug delivery system. All those processes are mainly influenced by the physicochemical properties, such as the size and surface charge, of the drug delivery system. Here, thin film technologies, like the LbL strategy are an ideal tool to fine-tune these properties on the nanoscale and hence can lead to a profound knowledge of the ideal parameters for the design of future drug delivery systems.
At the starting of this work a suitable nanoparticle material, which can easily be modified by an LbL strategy had to be found and characterized in detail. Gold nanoparticles were found to be an interesting colloidal material, due to their distinct optical properties. Gold nanoparticles of different sizes were synthesized according to the Frens method and their size-dependent extinction coefficients were calculated (Chapter 2). This connection was the fundamental basis for all further quantitative experiments, for example the cell association and protein adsorption studies. Those gold nanoparticles were used as templates for Layer-by-Layer (LbL) thin film surface modifications. As the coating procedure, especially the purification, of nanoparticles is troublesome, due to their high tendency against aggregation, a general protocol for the LbL coating of gold nanoparticles was established (Chapter 3). This protocol was later transferred to various other polyelectrolyte pairs and other core materials such as silica nanoparticles.
These tools were the basis for a comparative study on the cell association and protein adsorption behavior of different LbL coated gold nanoparticles (Chapter 4). Here, the multilayer formation of three different polycations poly(ethylene imine) (PEI). poly(allylamine hydrochloride) (PAH) and poly(diallyl dimethyl ammonium chloride) (PD) in combination with the polyanion poly(styrene sulfonate) (PSS) was investigated with surface plasmon resonance spectroscopy. Those polycations were chosen because of their comparable chemical composition and, in case of PEI, because of its known transfection capacity. But, an unusual deviation of the LbL deposition, a so called overshoot-effect, was found for the PEI/PSS polyelectrolyte pair. Atomic force microscopy revealed that this overshoot phenomenon increased the surface roughness by 11-fold upon adsorption of the third polymer layer and at the same time sharp surface features appeared. It was further assumed that the overshoot effect happened also upon the coating of gold nanoparticles, due to a decrease of the particle size at the same coating step. Hence, it was also concluded that a comparable surface structure was present on the trilayered PEI/PSS coated particles. This surface structure had a major influence on two known physiological barriers; on the adsorption of serum proteins and consequently on the association to cells. The protein adsorption increased significantly for the trilayered PEI/PSS particles, compared to the corresponding bilayered particle species. And this elevated protein shell hindered the association to HeLa cell in vitro. As all other physicochemical properties of the particles, the particle size and the surface charge, were comparable for all particle types, it was concluded that the surface topography had a major influence on the interactions with the biological counterparts. As a consequence, this property should be considered for the design of future nanoparticle based drug delivery systems.
Further specialties of LbL systems were highlighted in Chapter 5. LbL multilayers are known to be dynamic constructs with an internal conformation of the polymer chains that highly depends on the applied fabrication conditions. The internal conformation of the above mentioned polyelectrolyte pairs were investigated with an approach based on Foerster resonance energy transfer. It was found that the combination of PAH/PSS led to a highly interpenetrated polymer network. Those interpenetrations acted as crosslinks between the polymer chains and increased the stability of this polymer films against swelling in a biological milieu. Hence, it was found that PAH/PSS multilayers are disadvantageous for drug delivery systems, where a charged macromolecular drug is incorporated into the multilayer itself. In contrast, PAH/PSS films might be suitable as coating for drug reservoirs, because they showed a good permeability for small molecular drugs. Additionally the mode of release of drugs could be fine-tuned by the degree of ionization of PAH.
The extracellular matrix as another important physiological barrier for colloidal drug carriers was addressed in Chapter 6. So far, the interstitial connective tissue, which contains large amounts of collagen I, was less regarded as an effective molecular filter. But with fluorescence recovery after photobleaching studies in collagen I hydrogels it was found that cationic LbL coated particles and unbound polycations were effectively trapped inside those model matrices. It was further shown, that the addition of a high concentrated PEI-solution of 0.5 mg/ml increased the mobility of the trapped species. But at the same time, these high concentrated polycations altered the constitution of the collagen I fibrils, as it was found with turbidity assays. Hence, it was concluded, that an effective electrostatic filter can also be assumed for the interstitial connective tissue and collagen I hydrogels are a suitable model systems, if the appropriate control experiments are performed side-by-side.

In der vorliegenden Arbeit wurden die Wechselwirkungen von sogenannten Layer-by-Layer Dünnfilmen und möglichen biologischen Barrieren in einem Wirkstofftransportmodell untersucht.
Verschiedene physiologische Barrieren eines Organismus, die Adsorption von Serumproteinen, die Zellassoziation oder die Barrierefunktion biologischer Hydrogele, limitieren die Transporteffizienz und letztendlich den therapeutischen Effekt eines Wirkstofftransportsystems. Diese Prozesse werden hauptsächlich von den physikochemischen Eigenschaften der Transportsysteme, wie der Partikelgröße oder der Oberflächenladung, bestimmt. In diesem Zusammenhang sind Dünnfilmtechniken, wie die Layer-by-Layer (LbL) Strategie, ideale Werkzeuge um diese Eigenschaften nanometergenau einzustellen. Des Weiteren führen Sie zu einem fundamentalen Verständnis der idealen physikochemischen Parameter um effiziente Wirkstofftransportsysteme zu entwickeln.
Am Beginn der Arbeit stand die Auswahl und Charakterisierung eines für die LbL-Technik geeigneten Nanopartikelmaterials. Hierbei stellten sich Goldnanopartikel aufgrund ihrer besonderen optischen Eigenschaften als ein interessantes Kolloidsystem heraus. Goldnanopartikel verschiedener Größen wurden mit der Methode nach Frens synthetisiert und die dazugehörigen größenabhängigen Extinktionskoeffizienten berechnet (Kapitel 2). Dieser Zusammengang bildete die Grundlage für alle folgenden quantitativen Untersuchungen, zum Beispiel zur Proteinadsorption und Zellassoziation. Die hergestellten Goldnanopartikel wurden anschließend als Substrate für die LbL-Methode verwendet. Da dieses Überzugsverfahren insbesondere durch die notwendigen Aufreinigungsschritte für Nanopartikel herausfordernd ist, wurde zunächst ein allgemeines Verfahren zum Überziehen von Nanopartikeln entwickelt (Kapitel 3). Dieses Verfahren wurde später auf weitere Polyelektrolytpaare und Nanopartikelkerne übertragen.
All diese Werkzeuge bildeten die Basis für eine vergleichende Untersuchung zur Zellassoziation und Proteinadsorption verschiedener mittels LbL überzogener Goldnanopartikel (Kapitel 4). Die Bildung von Multischichten aus drei unterschiedlichen Polykationen, nämlich Polyethylenimin (PEI), Polyallylaminhydrochlorid (PAH) und Polydiallylamindimethylammoniumchlorid (PD), kombiniert mit Polystyrolsulfonat (PSS) als Polyanion, wurde mit der Oberflächenplasmonresonanzspektroskopie untersucht. Die genannten Polykationen wurden aufgrund ihres ähnlichen chemischen Aufbaus, und im Fall von PEI, auch auf Grund seiner bekannten Transfektionsfähigkeit, ausgewählt. Eine Besonderheit im LbL-Aufbau, ein sog. „overshoot-Effekt“ wurde dabei für das Polyelektrolytpaar PEI/PSS gefunden. Rasterkraftmikroskopie auf planaren Oberflächen zeigte, dass dieses Phänomen zu einer zwölffach erhöhten Oberflächenrauigkeit bei der Adsorption der dritten Polyelektrolytschicht führte. Des Weiteren wurde vermutet, dass dieser Effekt auch bei der Beschichtung von Nanopartikeln auftrat, da hier der Partikelradius ebenfalls beim Aufbringen der dritten Schicht abnahm. Folglich wurde auch eine ähnliche Oberflächenstruktur auf den Nanopartikeln angenommen. Diese Oberflächenstruktur hatte einen großen Einfluss auf die Serumproteinadsorption und die Zellassoziation. Einerseits stieg die Serumproteinadsorption für die dreischichtigen PEI/PSS Partikel, im Vergleich zur zweischichtigen Variante, signifikant an. Andererseits behinderte die stark ausgebildete Proteinhülle die Assoziation an HeLa Zellen in vitro. Da alle anderen physikochemischen Eigenschaften, wie Partikelgröße und Oberflächenladung zwischen allen Partikelarten vergleichbar waren kann auf einen großen Einfluss der Oberflächentopographie auf die biologischen Wechselwirkungen geschlossen werden. Folglich sollte diese Eigenschaft für die zukünftige Entwicklung neuer nanopartikelbasierter Wirkstofftransportsysteme stärker berücksichtigt werden.
Weitere Eigenschaften von LbL Systemen wurden in Kapitel 5 beleuchtet. LbL-Multilayer sind bekanntermaßen sehr dynamische Konstrukte mit einer inneren Konformation der Polymerketten, die von den Beschichtungsbedingungen abhängt. Die innere Konformation der oben genannten Polyelektrolytpaare wurde durch einen Ansatz basierend auf der Foerster Resonanzenergieübertragung untersucht. Die Kombination aus PAH und PSS konnte damit als ein hochverzahntes Polymernetzwerk beschrieben werden. Diese Verzahnungen dienen offenbar der Quervernetzung zwischen den Polymerketten und erhöhten die Stabilität dieses Polymerfilms gegenüber Quellung in einem biologischen Milieu. PAH/PSS-Multilayer sind daher unvorteilhaft für Wirkstofftransportsysteme in denen ein geladenes Makromolekül im Polymernetzwerk eingebunden ist. Im Gegensatz dazu sind PAH/PSS Schichten geeignet für die Beschichtung von Wirkstoffdepots, da sie eine gute Permeabilität für niedermolekulare Wirkstoffe zeigten. Zusätzlich konnte die Wirkstofffreisetzung durch den Ionisierungsgrad von PAH gesteuert werden.
Die extrazellulare Matrix, eine weitere wichtige physiologische Barriere für kolloidale Wirkstoffträger, wurde in Kapitel 6 behandelt. Bis dahin wurde das interstitielle Bindegewebe, das eine große Menge Kollagen I enthält, als wenig effektiver molekularer Filter angesehen. Mit der Technik des „fluorescence recovery after photobleaching“, wurde jedoch gezeigt, dass kationisch beschichtete Nanopartikel sowie ungebundene Polykationen, in Kollagen I-Modellmatrizes effektiv aufgehalten wurden. Des Weiteren erhöhte der Zusatz einer hochkonzentrierten PEI-Lösung von 0,5 mg/ml die Mobilität der gefangenen Spezies. Gleichzeitig veränderten diese hochkonzentrierten Polykationen aber in Trübungsversuchen die Beschaffenheit der Kollagenfibrillen. Letztendlich konnte gezeigt werden, dass das interstitielle Bindegewebe einen effektiven elektrostatischen Filtereffekt besitzt und Kollagen-I-Hydrogele hierfür geeignete Modellmatrizes darstellen, sofern geeignete Kontrollen mitgeführt werden.
