Abstract (German)
Das Mammakarzinom ist das häufigste Malignom der Frau. Gerade deshalb besteht ein großes Interesse daran, die Therapiemöglichkeiten ständig zu erweitern und zu verbessern. Insbesondere endokrine und „target“-spezifische Therapien gewinnen immer mehr an Bedeutung. Das Mammakarzinom ist das häufigste Malignom der Frau. Gerade deshalb besteht ein großes Interesse daran, die Therapiemöglichkeiten ständig zu erweitern und zu verbessern. Insbesondere endokrine und „target“-spezifische Therapien gewinnen immer mehr an Bedeutung. In den letzten Jahren kristallisierte sich der erst 1996 entdeckte, zweite Östrogenrezeptor β als möglicher Apoptose-Auslöser bzw. Tumorsuppressor heraus.
Wir untersuchten deshalb, inwiefern die Östrogenrezeptor-β-Modulatoren Liquiritigenin, Erb-041, WAY200070, FERb-033, 3-β-Adiol, (R-R)-THC und PHTPP die durch die Chemotherapeutika Etoposid, Doxorubicin, Docetaxel und Staurosporine ausgelöste Apoptose an den vier Brustkrebszelllinien MDA-MB-231, MCF-7, T47D und ZR-751 beeinflussen können bzw. ob die ERβ-Modulatoren eventuell auch bei alleiniger Anwendung die Apoptoserate der Zellen beeinflussen können. Die Ergebnisse sind durchaus positiv zu werten. Es konnte für einige Fälle ein deutlicher Effekt im Sinne einer Veränderung der Apoptoserate durch die ERβ-Modulatoren ausgelöst werden.
Es scheint jedoch von diversen Faktoren (z.B. ERα-Status, vorhandene ERβ-Isoformen, Coaktivatoren, Corepressoren, Phosphorylierungsstatus von ERβ, Einwirkdauer, Apoptose-Auslöser) abzuhängen, inwiefern eine ERβ-Aktivierung die Apoptose beeinflusst. Gegenstand weiterer Forschung sollte sein, all diese Faktoren aufzudecken und deren Wirkung zu analysieren.

Breast cancer is the most common cancer in females. For this reason, there is a great interest in constantly expanding and improving the therapy options. Breast cancer is the most common cancer in females. For this reason, there is a great interest in constantly expanding and improving the therapy options. In particular, endocrine and targeted therapies are becoming increasingly important. In recent years the second estrogen receptor, named ER β and discovered in 1996, crystallized out as a possible trigger of apoptosis or at least as tumor suppressor.
We therefore investigated to what extent the estrogen receptor β modulators Liquiritigenin, Erb-041, WAY200070, FERb-033, 3-β-adiol, (RR) -THC and PHTPP may influence the by chemotherapeutic agents, such as etoposide, doxorubicin, docetaxel and Staurosporine, induced apoptosis of four breast cancer cell lines (MDA-MB-231, MCF-7, T47D and ZR-751), or whether the ERβ modulators may possibly also influence the apoptotic rate of these cells on their own. The results are quite positive. In some cases, a clear effect in terms of a change in the rate of apoptosis by the ERβ modulators could be triggered.
However, it seems to depend on various factors (e.g., ERα status, existing ERβ isoforms, coactivators, corepressors, phosphorylation status of ERβ, duration of exposure, apoptosis inducer) to what extent ERβ activation affects apoptosis. The subject of further research should be to uncover all these factors and to analyze their effects.
