Parvovirus B19 ist der einzige bekannte humanpathogene Vertreter der Parvoviridae. Ziel der vorliegenden Arbeit war die detaillierte biophysikalische Beschreibung der Interaktion von B19 mit seinem vorgeschlagenen, zellulären Rezeptor, dem Glycospingolipid Globosid. Es wurde eine spezifische IgG-Fraktion gegen das virale Strukturprotein VP2 zum Einsatz als immobilisierender Träger für Bindungsassays hergestellt und die Präparation rekombinanter VP2-Proteinkapside optimiert. Zur Charakterisierung der B19/Globosid-Wechselwirkung wurden Bindungstests mit fluoreszenz-markierten Liposomen und 125I-markierten Kapsiden etabliert. Die direkten Testsysteme lieferten keine Hinweise auf eine spezifische Kapsid/Globosid-Interaktion. Die Ergebnisse konnten durch Biosensormessungen und kalorimetrische Untersuchungen verifiziert werden. Hämagglutinationsuntersuchungen im Rahmen dieser Arbeit bestätigten die publizierte Hemmaktivität von Globosid, zeigten aber eine vergleichbare Aktivität von Globopentaosylceramid. Dies stellt eine hohe Spezifität des Globosid-Kapsid-Kontaktes in Frage. Die Ergebnisse implizieren, daß Globosid nicht als singulärer Rezeptor für B19 agieren kann, aber vielleicht im Kontext einer Interaktion des Virus mit einem anderen Rezeptormolekül erkannt wird. Daher wurden erste Untersuchungen zur Identifizierung anderer Rezeptormoleküle in Membranproteinfraktionen von humanen Zellinien durchgeführt und die Bindung 125I-markierter Kapside an mehrere Proteine von 30 bis 100 kD Größe detektiert. Die Identität der Banden ist weitgehend unklar, wobei die Spezifität einer Interaktion mit Carboanhydrase noch überprüft werden muß. Das Kapsid von Parvovirus B19 besteht aus nur zwei Strukturproteinen. Für weitere Bindungstests und Strukturuntersuchungen wurden heterogene VP1/VP2-Partikel im Baculo/Sf9-System produziert. Durch Mutation des Promoterbereiches des bicistronischen Baculovirus konnten Mischkapside mit VP1-Anteilen von 45 bzw. 30 hergestellt werden.

Parvovirus B19 is the only humanpathogenic member of the Parvoviridae known. Aim of this studies was the detailled biophysical characterization of the interaction of B19 with ist putative receptor globoside. A specific IgG fraction against the viral structural protein VP2 as immobilizing agent for binding assays was produced, and the preparation of recombinant VP2 protein shells was optimized. For the characterization of the B19/globoside interaction two binding assays with fluorescence labeled liposomes and 125I-labeled capsids were established. In the direct binding assays no specific capsid/globoside interaction was detected. These results have been verified by biosensor measurements and microcalorimetry. Hemagglutination studies reported herein confirmed the published inhibitory activity of globoside, but revealed a comparable activity of globopentaosylceramide. Therefore, the high specificity of the globoside recognition is questionable. The results implicate that globoside does not function as the direct singular receptor of Parvovirus B19 but could be recognized in a complex with other receptor molecules. For the identification of such molecules a virus overlay binding assay using membrane protein fractions of human cell lines was performed. In this assay the binding of 125I-labeled capsids to several proteins between 30 and 100 kD was detected. The identity of the bands is still unclear. However, the specificity of the detected interaction with carboanhydrase has to be verified. The Parvovirus B19 capsid consists of only two structural proteins. For comparison in binding assays and for further structural determination heterogeneous VP1/VP2 capsids were produced in the Baculovirus/Sf9 expression system. Mutations in the promoter regions of the bicistronic baculovirus accomplished the preparation of capsids with 45 or 30 VP1.
