Synthetische biokompatible bioabbaubare Polymere werden erfolgreich für verschiedenste medizinisch-pharmazeutische Applikationen eingesetzt. In der Arzneimitteltherapie dienen sie als Wirkstoff-Träger, mit denen Ziele wie kontrollierte Wirkstofffreisetzung und Freigaben bis hin zu mehreren Monaten realisiert werden können. Entscheidend für den Einsatz dieser Polymere ist ihre Eigenschaft, im Kontakt mit Körperflüssigkeiten zu erodieren, d.h. Masse zu verlieren. Jede operative Entfernung nach Erreichen des Einsatzzieles wird damit überflüssig. Der Erosionsprozess ist gleichzeitig entscheidend für eine kontrollierte Wirkstofffreisetzung und für die mechanische Stabilität polymerer Körper. Trotz der Bedeutung des Erosionsverhaltens ist dieses für viele Polymere nur ungenügend untersucht. Das Anwendungspotential dieser Materialien kann damit nur unzureichend genutzt werden.
Um das Verständnis über Erosionsverhalten und Erosionsmechanismen bioabbaubarer Polymere zu verbessern, wurde der Erosionsprozess repräsentativ am Beispiel von Poly(D,L-laktiden), Poly(D,L-laktid-co-glykoliden) und Diblock-Copolymeren aus Poly(D,L-laktid) und Poly(ethylenglykol) mittels GPC, DSC, WAXD, NMR und SEM untersucht. Basierend auf den Ergebnissen wurde ein Modell entwickelt, anhand dessen bioabbaubare Polymere nach ihrem Erosionsmechanismus klassifiziert werden können. Das Modell zeigt, daß alle abbaubaren Polymere sowohl nach dem Mechanismus der Bulk- als auch nach dem der Oberflächenerosion erodieren können. Welcher Mechanismus abläuft, ist entscheidend abhängig von den Erosionsbedingungen, der chemischen Struktur des Polymers u. den Abmessungen des polymeren Körpers. Mit diesem Modell ist ein grundlegendes System geschaffen, das die Vorhersage des Erosionsverhaltens ermöglicht. Damit trägt diese Arbeit zu einem besseren Verständnis von Erosionsverhalten u. Erosionsmechanismen bioabbaubarer Polymere u. zu einer Optimierung ihres Einsatzes für pharmazeutische u. medizinische Zwecke bei.

Synthetic biocompatible biodegradable polymers are successfully applied for various medical-pharmaceutical purposes. In drug delivery they serve as carrier materials for drugs which can be used for aims such as controlled release and release for periods up to months. Decisive for the use of these polymers is their property to erode by the contact to body fluids which means that they loose mass. Due to this any removal by surgery, after having reached the goal of application, is superfluous. The process of erosion is at the same time decisive for a controlled release of drugs and for the mechanical stability of polymeric devices. Despite the importance of the process of erosion, for many polymers this process is only insufficiently examined. The potential possibilities of application of these materials can only be used insufficiently.
To improve the knowledge on the process and the mechanisms of erosion of biodegradable polymers, the process was examined by GPC, DSC, WAXD, NMR and SEM. Poly(D,L-lactides), poly(D,L-lactide-co-glycolides) and diblock-copolymers from poly(D,L-lactide) and poly(ethylenglykol) served as model polymers. Basing on the results a theoretical model was developed which allows to classify biodegradable polymers according to their mechanism of erosion. The model shows that all biodegradable polymers can erode both according to the mechanism of bulk erosion as well as according to the mechanism of surface erosion. Which mechanism proceeds depends decisively on the conditions of erosion, the chemical structure of the polymer and the dimensions of the polymeric device. With this model a basic system is created which allows to predict the process of erosion. Thus, this work helps to improve the knowledge on the process and the mechanisms of erosion of biodegradable polymers and to optimise their application for pharmaceutical and medical purposes.
