Die vorliegende Dissertation befasst sich mit der histologischen, genetischen und funktionellen Charakterisierung des Gens headcase (hdc) innerhalb der Entwicklung des visuellen Systems von Drosophila melanogaster. Die Bedeutung von headcase für das optische System wurde im Rahmen einer Suche nach neuen Mutanten mit defekter retinaler Projektion innerhalb einer Sammlung letaler P-Elementstämme (Deak et al., 1997) entdeckt. Dabei zeigte die lebensfähige Linie 75/04 grobe Störungen im Projektionsmuster der Photorezeptoraxone (PR-Axone), deren P-Element im zytogenetischen Bereich von headcase inseriert war. Die angeforderte headcase Deletionsmutante hdc50 bestätigte die beobachteten Defekte und Komplementationskreuzungen mit 75/04 wiesen den kausalen Zusammenhang zwischen der P-Elementinsertion und dem mutanten Phänotyp nach. Die pupal letale hdc50-Mutante zeigte ein komplexes Störungsmuster innerhalb des visuellen Systems: Larval traten besonders Faszikulierungsdefekte und eine gestörte Topographie in den ersten beiden optischen Ganglien hervor, wobei die Augenscheibe kaum gestört war; im fast adulten Zustand zeigten nicht nur die optischen Ganglien, sondern auch die Retina starke Defekte, mit dem prominenten Bypass-Phänotyp der retinalen Projektionen in der Medulla.
Die folgenden Expressionsstudien gaben erste Hinweise auf den Wirkort von headcase. Es stellte sich heraus, dass HDC fast ubiquitär im larvalen bzw. frühpupalen, aber nicht mehr im adulten visuellen System exprimiert wird. Es konnte auch nicht im Neuropil, sondern nur im Zytoplasma von Neuronen, Glia und anderen Zellen im Hirn und im Auge nachgewiesen werden. Die Untersuchung von Mosaiktieren, in denen hdc50-mutante Photorezeptoraxone in ein wildtypisches Hirn projizieren, zeigte die funktionelle Notwendigkeit von headcase für die Entwicklung der Retina und des optischen Lobus. Eine partielle Rettung der Projektionsdefekte von hdc50 durch eine photorezeptorspezifische Expression der headcase-cDNA bestätigte die Mosaikexperimente und damit die ubiquitäre Notwendigkeit der headcase-Funktion für die Entwicklung des optischen Systems. Diese Aussagen manifestierten sich in der weiteren Detailuntersuchung des pleiotropen hdc50-Phänotyps. Spezifische Markierungen der PR-Axone belegten, dass alle acht Photorezeptortypen die hdc-Funktion für ihre korrekte Projektion ins Hirn benötigen. Ferner lässt die Fehlexpression von Zelladhäsionsmolekülen in hdc50-mutanten PR-Axonen darauf schließen, dass headcase mitbeteiligt ist an deren wildtypischer Faszikulierung. Desweiteren bewies eine Untersuchung der Zielregion retinaler Projektionen die Notwendigkeit von headcase sowohl für die wildtypische Verteilung von Gliazellen zur korrekten Terminierung der PR-Axone, als auch für eine geordnete Proliferation von Neuroblasten im optischen Lobus im Hinblick auf eine normale Entwicklung des ersten optischen Ganglions.

Translation of the abstract (English)

The present dissertation deals with the histological, genetic and functional characterisation of the gene headcase in the development of the visual system of Drosophila melanogaster. The importance of the function of headcase in the optical system was found during a search for new mutants with defects in the retinal projections within a collection of lethal P-element insertion lines (Deak et al., 1997). Particularly the viable line 75/04, with a P-element insertion at the cytogenetic interval of headcase, reveals defects in the photoreceptor axon (PR-axons) projection pattern. The observed defects were verified by the use of a headcase deletion mutant, hdc50 , and complementation analysis with 75/04 confirmed the causality between the P-element insertion and the mutant phenotype. The pupal lethal mutant hdc50 display a complex pattern of disorders in the visual system: In larvae defects in fasciculation and topography are especially significant, whereas the eye disc appeares quite normal. Adult mutant flies exhibit strong defects not only in the optic ganglions, but also in the retina, including a prominent medulla bypass-phenotype of the retinal projections.
First hints referring to spatial distribution of the headcase function came from expression studies. Headcase is almost ubiquituosly expressed in larvae and early pupae, but not in the adult visual system. Moreover, there was no expression in the neuropil, only in the cytoplasm of neurons, glia and other cells of the brain and the eye. Experiments with mosaic flies, whose hdc50-mutant PR-axons project into a wildtype brain, have demonstrated the requirement of headcase function for the development of the retina and the optic lobe. A partial rescue of the hdc50 projection defects via photoreceptor specific expression of a hdc-cDNA validated the mosaic experiments indicating the ubiquitous requirement of the headcase function for the development of the optic system. Specific labelling of PR-axons has shown, that all eight types of photoreceptors need the headcase function for their correct projection into the brain. The misexpression of cell adhesion molecules in hdc50-mutant PR-axons revealed the participation of headcase in their fasciculation. The investigation of the target region of retinal projections has proven the requirement of headcase for both, the wild type distribution of glia cells to ensure a correct termination of the PR-axons and the orderly proliferation of neuroblasts in the optic lobe concerning a normal development of the first optic ganglion.
