Polymeric cell carriers (scaffolds) play a pivotal role in attempts to engineer living tissue from isolated cells for tissue reconstruction and basic research. This thesis aimed at fabricating functional, biodegradable scaffolds. The functional cell carrier design concept involved a biodegradable copolymer that allows for instant biomimetic surface modification. A suitable fabrication technique was developed to process lab-scale amounts of the biomaterial into biocompatible, macroporous cell carriers with controlled architectural properties. Furthermore, an injectable hydrogel matrix was synthesized as part of a new concept to induce neovascularization of tissue engineered constructs.
Recently developed, low-adhesive, amine- and thiol-reactive biodegradable diblock copolymers were employed as functional scaffold materials. To demonstrate that cell adhesion to these polymers can be specifically promoted through the attachment of adhesion peptides, prefabricated films of both amine- and thiol-reactive copolymer films were modified with cyclic, αvβ3/αvβ5 integrin subtype specific, cyclic RGD peptides employing a simple incubation protocol. While only a small percentage of human osteoblasts adhered to the control films, cell adhesion was promoted on the RGD-modified copolymer surfaces. The adhered cells were well spread, while cells on the control surfaces exhibited a round shape. Further control groups proved the covalent attachment of the peptides, demonstrating the tested polymers potential to achieve tailored surface modification and selective cell adhesion for biomedical applications.
To process these functional diblock copolymers into tissue engineering scaffolds, a suitable fabrication technique was required that accounts for the physicochemical properties of the polymers, the hydrolytic sensitivity of the reactive groups, and the general demands on scaffold microstructure, namely high porosity, control of pore size and high pore interconnectivity. To this end, an anhydrous technique was developed, which utilizes solid lipid microspheres dispersed in an organic solution of the polymer as pore forming devices. Simultaneous polymer precipitation and porogen extraction on contact with warm n-hexane generated macroporous scaffolds. The adjustment of the porogen particle�s melting point to the polymer�s glass transition temperature was shown to be a key step in generating interconnected networks. Scaffolds from amine-reactive polymers were fabricated and insulin, as a model protein, was covalently attached to the scaffold surface in a simple incubation step. To dispose the halogenated solvent chloroform from polymer processing, an alternative solvent mixture, thermodynamically equal to the established acetone-chloroform-mixture (65:35 (v/v)), was systematically determined using Hansen Solubility Parameters (HSPs). The HSPs of a methyl ethyl ketone-tetrahydrofurane-mix (59:41 (v/v)) were found to fit best. Using this mixture, polymer processing into interconnected scaffolds was successful without further changes to the established process parameters. A follow-up study focused on determining the HSPs of poly(lactic acid) (PLA) and two of its copolymers. Theoretical and experimental methods, namely group contribution methods, solubility and viscosity measurements, were employed.
During polymeric scaffold fabrication, control over architectural and mechanical properties is mandatory for adaptation to different applications and types of tissue. Parameters that allow for the processing of various biodegradable polymers of different compositions and molecular weights (22 - 160 kDa) were determined for the developed solid lipid templating technique. Pore interconnectivity, was controlled by adjusting the extraction temperature and the porogen�s melting point. The pore size of the scaffolds could be predetermined by the size distribution of the porogen microspheres. The rheological behavior of the dispersion prepared from the polymer solution and lipid microspheres, which had influence on the scaffolds micro- and macrostructure, was characterized by an oscillating rheometer. The scaffolds proved suitable for cartilage cell culture.
To overcome limitations in the nutrient and oxygen supply inside tissue engineered cell-polymer constructs, strategies to promote the ingrowth of blood vessels are under intensive investigation. Towards a novel strategy, an injectable artificial extracellular matrix was synthesized from gelatin and N isopropylacrylamide. Different type A gelatins were tested and conjugates were synthesized with varying gelatin contents. The conjugates were characterized by GPC, NMR, DSC, UV spectroscopy and rheology. The gels underwent thermo-reversible gelation around 31°C. High cell viability (90%) in the gels was determined for mesenchymal stromal cells. Encapsulated chondrocytes could be stimulated for glycosaminoglycan production over 14 days demonstrating the cytocompatibility of the injectable matrix.

Zellträger (Scaffolds) aus Polymeren spielen eine bedeutende Rolle bei der Gewebezüchtung aus isolierten Zellen zur Geweberegeration und für die Grundlagenforschung. Ziel dieser Dissertation war die Herstellung funktioneller, bioabbaubarer Scaffolds. Bioabbaubare Copolymere, die nach Modifikation gezielt mit Zellen wechselwirken können, dienten als funktionelles Scaffoldmaterial. Eine Herstellungsmethode wurde entwickelt, um dieses Biomaterial im Labormaßstab zu biokompatiblen, makroporösen Zellträgern mit kontrollierbaren strukturellen Eigenschaften verarbeiten zu können. Schließlich wurde eine injizierbare Matrix, als Bestandteil eines neuartigen Konzeptes zur Neovaskularisierung der gezüchteten Gewebe, synthetisiert.
Neuartige, die Zelladhäsion unterdrückende, amino- und thiolreaktive Diblock-Copolymere wurden als funktionelles Material eingesetzt. Um zu zeigen, daß eine spezifische Adhäsion von Zellen auf diesen Materialien durch Anbindung von Adhäsionssequenzen erreicht werden kann, wurden Integrinrezetor-spezifische RGD-Peptide an Filme aus den reaktiven Copolymeren durch Inkubation mit der Peptidlösung angebunden. Während auf den Kontrolloberflächen nur ein geringer Anteil der ausgesäten Osteoblasten adhärierte, wurde eine nahezu vollständige Zelladhäsion auf den RGD-modifizierten Polymerfilmen gefunden. Die adhärierten Zellen waren stark gespreitet, während die Zellen auf nichtmodifizierten Oberflächen abgerundet waren. Mittels geeigneter Kontrollgruppen konnte auf eine kovalente Anbindung der Peptide geschlossen werden.
Um diese funktionellen Copolymere zu Scaffolds für das Tissue Engineering zu verarbeiten, wurde eine Herstellungstechnik entwickelt, welche die physiko-chemischen Eigenschaften der Polymere, die Hydrolyseempfindlichkeit der aktiven Gruppe und die generellen strukturellen Anforderungen, d.h. hohe Porosität, Kontrolle über die Porengröße und hohe Interkonnektivität, berücksichtigt. Das entwickelte wasserfreie Verfahren nutzt Mikropartikel aus Hartfetten als Porenbildner, die in einer organischen Lösung des Polymers dispergiert werden. Durch das gleichzeitige Fällen des Polymers und Herauslösen des Porenbildners bei Kontakt mit warmen Hexan bilden sich makroporöse Scaffolds. Das Anpassen der Schmelztemperatur der Porogenpartikel an die Glasübergangstemperatur des Polymers war dabei entscheidend für die Herstellung interkonnektiver Porenstrukturen. Gerüste aus amino-reaktiven Polymeren wurden hergestellt und das Modellprotein Insulin wurde in einer einfachen Inkubationsprozedur kovalent an die Gerüstoberfläche angebunden.
Um Chloroform aus dem Verarbeitungsprozess zu eliminieren, wurde mittels der Hansen Löslichkeitsparameter (HLP) systematisch eine alternative, der etablierten Aceton-Chloroform-Mischung thermodynamisch equivalente, Lösungsmittelmischung bestimmt. Die HLP einer Mischung aus Methylethylketon und Tetrahydrofuran (59:41 (V/V)) entsprachen am Besten. Mittels dieser Mischung konnten die bioabbaubaren Polymere zu Zellgerüsten mit interkonnektiven Poren ohne weitere Parameteranpassung verarbeitet werden.
Die Anschlußstudie zielte auf die Bestimmung der HLP von Polylaktid (PLA) and zweier PLA-haltiger Copolymere. Zu diesem Zweck wurden theoretische und experimentelle Methoden, nämlich verschiedene Gruppenbeitragsmethoden, sowie Löslichkeits- und Viskositätsmessungen eingesetzt.
Bei der Scaffoldherstellung ist die Kontrolle über die architektonischen und mechanischen Eigenschaften eine grundsätzliche Voraussetzung, um Anpassungen an verschiedene Anwendungen und Gewebtypen zu ermöglichen. In einer weiteren Studie wurden die Prozessparameter für die Verarbeitung bioabbaubarer Polymere von verschiedener Zusammensetzung und unterschiedlichem Molekulargewichts (22-160 kDa) mittels dem Solid Lipid Templating bestimmt. Die Interkonnektivität der Poren wurde dabei durch die Anpassung der Extraktionstemperatur an die Schmelztemperatur der Porogene gewährleistet. Die Porengröße der Scaffolds konnte durch die Größenverteilung der eingesetzten Porogenpartikel vorgegeben werden. Die rheologischen Eigenschaften der Dispersion aus Polymerlösung und Lipidpartikeln, welche die Makro- und Mikrostruktur der Scaffold beeinflussen, wurden mittels eines oszillierenden Rheometers bestimmt. Die Zellträger wurden erfolgreich zur Kultivierung von Chondrozyten eingesetzt.
Als Teil einer neuartigen Strategie zur Förderung der Vaskularisierung in vitro gezüchteter Gewebe wurde eine künstliche, injizierbare Extrazellularmatrix aus Gelatine und N-Isopropylacrylamid synthetisiert. Verschiedene Gelatinen vom Typ A wurden getestet und Konjugate verschiedener Zusammensetzung synthetisiert und charakterisiert. Die Konjugate zeigten thermoreversible Gelbildung bei 31°C. In diesen Gelen konnte eine hohe Viabilität (90%) für mesenchymale Stammzellen gezeigt werden. Eingebettete Chondrozyten konnten über 14 Tage zur Produktion von Glucosaminoglykanen stimuliert werden, was die Zytokompatibilität dieses Materials zeigt.
