Abstract (German)
In den 50er Jahren finden A. L. Hodgkin und R. D. Keynes bei Flussmessungen von Kaliumionen am Riesenaxon des Tintenfischs Sepia Officinalis, dass das Verhältnis der unidirektionalen Flüsse, des Kaliumflusses vom Zellinneren nach außen zum Kaliumfluss von der Zellumgebung in das Axon, von dem Wert abweicht, der bei freier Diffusion der Ionen zu erwarten ist. Als Maß dieser Abweichung führen sie den Flussverhältnis-expoenenten n ein, der bei freier Diffusion den Wert 1 annimmt. Den experimentellen Wert von n = 2,5 interpretieren sie mit Hilfe der Single-File Diffusion [4]. Ein Flussverhältnis-exponent n > 1 ist charakteristisch für Single-File Diffusion, während ein Wert n < 1 typisch für den Transport mittels Carrier mit einem Bindungsplatz ist [5,6,8]. Schagina und Lev et al. bestimmen am Ionenkanal Gramicidin-A die Flussverhältnis-exponenten von Natrium und Rubidium: Liegen beide Kationen gemeinsam in Lösung vor, nehmen deren Flussverhältnis-exponenten n(Na) und n(Rb) gleiche Werte an, die zwischen den Werten liegen, die man findet, wenn nur eine Kationenspezies vorliegt [9,3]. Die analytische Beschreibung dieses Befundes mit Hilfe eines vereinfachten kinetischen Modells einer symmetrischen Single-File-Zweiplatzpore gelingt Aityan et al. nur teilweise: Die Flussverhältnis-exponenten der gemeinsam vorliegenden Kationen sind identisch, liegen jedoch über den Werten für Lösungen mit nur einem Kation [1]. Die Erweiterung des Modells mit der Annahme, dass der Ionenkanal in zwei Konformationen unterschiedlicher Ionenleitfähigkeit vorliegen kann, ermöglicht die Wiedergabe des experimentellen Ergebnisses [2]. Zur Klärung der Frage, ob das nicht vereinfachte Modell der symmetrischen Single-File-Zweiplatzpore gegebener Konformation in der Lage ist, die experimentellen Flussverhältnisexponenten zu beschreiben, werden in dieser Arbeit numerische Berechnungen durchgeführt. Für die Geschwindigkeitskonstanten werden Werte eingesetzt, die mit Hilfe einer Modellanpassung gewonnen sind [7]. Die Rechnungen zeigen, dass die experimentellen Ergebnisse weitgehend richtig wiedergegeben werden können. Das Modell der symmetrischen Single-File-Zweiplatzpore gegebener Konformation reicht völlig aus.
Im Anhang werden die unabhängigen kinetischen Parameter bestimmt, die die untersuchten Flussverhältnis-exponenten festlegen. Weiterhin wird untersucht, welche allgemeine Bedingung die Geschwindigkeitskonstanten erfüllen müssen, damit die Flussverhältnis-exponenten in Gegenwart zweier Kationen identisch werden und unter welchen Voraussetzungen Parallelverschiebungen der Funktionen der Flussverhältnis-exponenten der Art n(i)=n(i)(lg(c(i)/(mol/l))) (Kation der Spezies i; Konzentration c) bezüglich der Abszisse beobachtet werden. Es werden auch einige Kurvenverläufe der Funktionen n(i)=n(i)(lg(c(i)/(mol/l))) in Gegenwart zweier Kationen phänomenologisch vorgestellt. Darunter werden Beispiele für den überraschenden Befund gezeigt, dass bei geeigneter Wahl der Geschwindigkeitskonstanten der Flussverhältnisexponent einer Kationenspezies in Gegenwart einer zweiten von Werten n > 1 zu Werten n < 1 übergehen kann. Dies ist überraschend, weil die Vorstellung vorherrscht, dass bei Single-File Systemen nur Werte n >= 1 auftreten [5,6,8].

In the 50s A. L. Hodgkin and R. D. Keynes, determining the fluxes of potassium ions through the cell membrane of the giant axon of sepia officinalis, find that the ratio of the unidirectional fluxes (the directional flux from the inner of the cell to the outside divided by the flux from outside into the cell) differs from the ratio expected in the case of free ion diffusion. As a measure of this difference they introduce the flux ratio exponent n, which, in the case of free diffusion, equals 1. To interpret the experimental value of n = 2,5 they use the model of single-file diffusion [ Hodgkin 1955 ]. A value n > 1 typically occurs in single-file diffusion, while a value n < 1 is characteristic for a carrier with a single binding site [ Kohler 1979, 1980; Läuger 1980 ]. For the gramicidin A channel Schagina and Lev determine the flux ratio exponents of different alkali ions in solution. In the mixtures of sodium and rubidium they find that the flux ratio exponents of the two cations assume the same value lying between the respective values of the single-salt solutions [ Lev 1988; Blasko 1988 ]. Aityan et. al., using a simplified kinetic model of a symmetrical single-file pore, succeed only partially in describing this experimental finding analytically. The values of the flux ratio exponents in mixture are found to be identical but greater than the values for the single-salt cases. [ Aityan 1991 ]. Extending the model by the assumption of two different channel conformations of different ion conductivities, Aityan succeeds in describing the experimental finding [ Aityan 1991a ]. The failure of the one-conformation model may be due to simplifying assumptions concerning the rate constants of the model. These assumptions were necessary to achieve an analytical solution. In this work numerical calculations are performed using the unrestricted kinetic model of a symmetrical single-file pore with one conformation with rate constants derived from the single-ion cases [ Kohler 1983 ]. We find good agreement with the experimental values. It is concluded that a normal symmetrical single-file pore is sufficient to describe the flux ratio exponents in both the single-ion and the two-ion cases. In the appendix the independent kinetic parameters are listed determining the flux ratio exponents. General conditions are established under which the flux ratio exponents of two different cations simultaneously present become equal. Our numerical analysis of two-salt cases shows a surprising result: With suitable values of the kintetic parameters, the flux ratio exponent of one species may become smaller than one. Commonly it is believed that single-file systems always show flux ratio exponents n >= 1 [ Kohler 1979, 1980; Läuger 1980 ].
