Abstract (German)

Der Hautkrebs ist eine der am weitesten verbreiteten Krebsformen. Seine Neigung zur Metastasierung ist für die Agressivität des Tumors verantwortlich und führt schließlich zum Tod der betroffenen Patienten. Das Protein MIA (Melanoma Inhibitory Activity) wird nur von Melanomzellen, nicht jedoch von Melanozyten synthetisiert und sezerniert und ist mit dem Metastasierungsverhalten von Melanomzellen assoziiert.
In dieser Studie wurde die Rolle und Funktion von MIA auf die Entstehung von malignen Melanomen untersucht. Hierzu wurden Melanomzellen generiert, die aufgrund stabiler antisense-cDNA-Transfektion das Protein MIA nicht mehr exprimieren. Diese MIA-defizienten Melanomzellen wurden dann mit Hilfe von cDNA- und Antikörper-Arrays sowie mittels Powerblot-Analyse auf Unterschiede in der Gen- und Protein-Expression im Vergleich zu MIA-positiven Melanomzellen untersucht. Unterschiede in der Expression der zwei Zelllinien wurden anschließend durch PCR- beziehungsweise Western Blot-Analyse bestätigt. Vor allem Gene und Proteine, die direkt oder indirekt mit der Matrixdegradation (tPA, MT1-MMP), dem Übergang vom �radial growth phase�-Melanom zum �vertical growth-phase�-Melanom (Integrin beta3), oder der Proliferation assoziiert sind (Fibronektin) und für die Tumorentstehung und -progression eine wichtige Rolle spielen, wurden in MIA-positiven Zellen verstärkt exprimiert.

MIA is a protein produced and secreted by melanoma cells but not by melanocytes. It has been shown that MIA plays an important role in tumor progression in vivo and in regulation of differentiation in vitro. Therefore, we established a cell system in which MIA expression is inhibited via stable antisense MIA cDNA transfection. MIA-deficient cell clones showed changes in cell morphology like enhanced cell-cell contact and re-expression of E-Cadherin. MIA-deficient cell clones showed a re-induction of pigment synthesis in comparison to the amelanotic parental cell line HMB2. Molecular analysis revealed re-expression of TRP-1 and tyrosinase in the MIA-deficient cell clones responsible for melanin synthesis. Analyzing expression of MITF, a transcriptional regulator of Trp-1 and tyrosinase, showed reduced expression of MITF on protein level in melanoma cells. Further, differential expression of PIAS3 and PAX3, regulators of MITF activity and Trp-1 expression, was observed. Further investigating gene expression of the parental melanoma cell line HMB2 compared to MIA-deficient cell clones HMB2-5 and HMB2-8, respectively, in a micro array analysis revealed down regulation of MT1-MMP, tPA, fibronectin, MCAM (MUC18), Cathepsin D, Nestin und Integrin beta 3 (ITGB3) in the MIA-deficient cell clones. In summary, we can show that MIA-expressing melanoma cells compared to MIA-deficient cell clones differ in cell morphology, cell-cell-contact and pigmentation - which is due to the expression of MIA.
