Abstract (German)
In der vorliegenden Dissertation wurde die Expression und Funktion des Glykoproteins Gp96 in intestinalen Makrophagen bei Patienten mit und ohne chronisch entzündliche Darmerkrankungen untersucht. Dabei wurde zunächst die Expression in humaner intestinaler Mukosa untersucht und später die Wirkung von Gp96 auf den Entzündungsverlauf im Transfer-Kolitis-Maus-Modell. Weiterhin wurde die Expression der im NF-kappaB Signalweg eingreifenden Proteine TANK und TBK1 in intestinalen Makrophagen betrachtet. Konstrukte für einen Targeting-Vektor einer konditionellen Gp96-knock-out-Maus wurden hergestellt.
Die Expression von Gp96 in intestinalen Makrophagen wurde mittels Affymetrix GeneChip® Analysen, RT-PCR, real-time-PCR (TaqMan®) und immunhistochemischer Färbungen in intestinalen Makrophagen, sowohl im humanen, als auch im murinen System untersucht. Gp96 wird in intestinalen Makrophagen exprimiert und bei der Differenzierung zu diesen induziert. Um dies zu prüfen, wurde die Gp96-Expression in Monozyten und in in vitro differenzierten Makrophagen mittels real-time-PCR und RT-PCR analysiert. Im MZS-Modell zur Differenzierung von Monozyten in intestinale Makrophagen wurde gezeigt, dass Gp96 differenzierungsspezifisch für intestinale Makrophagen ist. Gp96-Protein ist in intestinalen Makrophagen bei Morbus Crohn nicht nachzuweisen, bei anderen intestinalen Entzündungen, wie Colitis ulcerosa und Divertikulitis, ist es exprimiert. Daher kann man vermuten, dass Gp96 essenziell für die Aufrechterhaltung der Toleranz gegenüber luminalen Antigenen ist.
Gp96 wurde zur Behandlung von immundefizienten Scid-Mäusen, bei denen durch den Transfer von CD4+ CD62L+-Zellen eine Kolitis induziert wurde, eingesetzt. Des Weiteren wurden Balb/c-Donortiere mit Gp96, peptidbeladenem Gp96 oder denaturiertem Gp96 behandelt und die T-Zellen auf Scid-Tiere übertragen. Es konnte festgestellt werden, dass die Behandlung der T-Zell-Spendertiere mit Gp96 einen positiven Effekt auf die Ausprägung der intestinalen Entzündung mit sich bringt. Auch die Behandlung der Tiere nach dem T-Zell-Transfer mit Gp96 führte zu diesem Ergebnis. Vor allem der histologische Score dieser Tiere war signifikant besser als der von Kontrolltieren. Auch die Analyse der Zytokinprofile der Donorzellen und der mesenterialen Lymphknotenzellen aus den Scid-Tieren zeigte, dass durch Gp96 T-Zellen generiert werden, die hauptsächlich TH2-Zytokine produzieren (IL-4, IL-10, IL-13) und somit regulatorische Funktion haben können. Die Transfer-Experimente lassen den Schluss auf eine essenzielle Rolle von Gp96 in der Aufrechterhaltung der Toleranz gegenüber luminalen Antigenen zu. Da Gp96 in intestinalen Makrophagen bei Morbus Crohn und der Transfer-Kolitis fehlt, kann vermutet werden, dass der Verlust von Toleranz für die Pathophysiologie bei Morbus Crohn wichtig ist.
Die für den Targeting-Vektor benötigten drei Fragmente aus dem Maus-Genom, die Neomycin-Resistenz und das lox p-Element wurden per PCR amplifiziert und in jeweils einen Topo-Vektor kloniert. Die Sequenzierungen ergaben, dass es sich um die gewünschten Gensequenzen handelt, so dass die Klonierung des Targeting-Vektors fortgesetzt werden kann.
Die Expression von TANK und TBK1 in intestinalen Makrophagen wurde zunächst mit Affymetrix GeneChip® Analysen untersucht. Dabei zeigte sich, dass die Expression von TANK in intestinalen Makrophagen, im Vergleich zu in vitro differenzierten Makrophagen, erhöht, die von TBK1 erniedrigt war. Durch Western Blot wurde gezeigt, dass TANK in LPMNZ der intestinalen Mukosa sowohl bei Entzündung als auch ohne Entzündung exprimiert wird. Durch Immunhistochemie konnte gezeigt werden, dass TANK in intestinalen Makrophagen vorkommt, wenn keine Entzündung vorherrscht. Bei Morbus Crohn wurde TANK in anderen noch nicht näher charakterisierten Zellen gefunden. Die Heraufregulation von TANK, bei gleichzeitiger Herabregulation von TBK1, in normalen intestinalen Makrophagen könnte zur Inhibierung einer TRAF2 vermittelten NF-kappaB-Aktivierung führen, was die Differenzierung in einen anergen Makrophagentyp unterstützt. Das Fehlen von TANK in intestinalen Makrophagen bei Morbus Crohn könnte eine erhöhte TRAF2 vermittelte NF-kappaB Aktivität zu Folge haben.

Thereby the expression of gp96 was first analysed in human intestinal mucosa and afterwards the effect of gp96 on the development of intestinal inflammation was studied in the transfer-colitis-mouse-model. Further the expression of two proteins, TANK and TBK1, which are involved in NF-kappaB-signalling, was analysed in intestinal macrophages. A targeting-vector for a conditional gp96-knock-out-mouse was constructed.
Expression of gp96 was analysed by Affymetrix Genechip arrays, RT-PCR, real-time-PCR (Taqman) and immunohistochemical staining in human and murine intestinal macrophages. Gp96 is expressed in intestinal macrophages and it is induced during differentiation of monocytes into intestinal macrophages. To test this, gp96 expression was analysed in monocytes and in vitro differentiated macrophages by real-time-PCR and RT-PCR. In the multicellular spheroid model for differentiation of monocytes into intestinal macrophages gp96 expression was shown to be differentiation specific for intestinal macrophages. Gp96-protein is not detectable in intestinal macrophages from patients with Crohn�s disease, but it is detectable in intestinal macrophages from patients with ulcerative colitis and diverticulitis. Therefore gp96 might be essential for the maintenance of tolerance towards luminal antigens.
In the CD4+ CD62L+ T-cell-transfer-model of colitis gp96 was tested to treat mice after T-cell-transfer. Furthermore Balb/c donor-mice have been treated with gp96, peptide loaded gp96 or denatured gp96 and T-cells were transferred to Scid-mice. Gp96-treatment of donor-mice ameliorated the intestinal inflammation and also treatment of recipient-mice with gp96 improved intestinal inflammation. First of all the histological score showed significant improvement in treated mice than in untreated controls. Analysis of secreted cytokines from donor-T-cells and mesenteric lymphnode-cells of Scid-mice showed enhanced TH2-cytokine-release (IL-4, IL-10, IL-13) after gp96-treatment and these cells might have regulatory functions. Because of these transfer-experiments it is obvious that gp96 plays an essential role in the maintenance of tolerance towards luminal antigens. Because gp96-protein is not expressed in intestinal macrophages in patients with Crohn�s disease and in mice with T-cell-induced colitis, one can conclude that loss of tolerance is an important factor in the pathophysiology of Crohn�s disease.
For the knock-out-mouse-targeting-vector three genomic fragments of the mouse genome have been amplified by PCR and also the neomycin-resistance-gene and the lox p element were amplified. All fragments have been cloned into TOPO-vectors. We performed sequence analysis with all fragments and showed that no mutations occurred during PCR so that cloning of the targeting-vector is continued.
Expression of TANK and TBK1 was first analysed by Affymetrix Genechip arrays in intestinal macrophages. Thereby it turned out that TANK-expression was enhanced in intestinal macrophages in comparison to in vitro differentiated macrophages. Expression of TBK1 was reduced in intestinal macrophages. Western blot analysis showed that TANK-protein was expressed in LPMNC of the intestinal mucosa in patients with and without intestinal inflammation. By immunohistochemical staining it was shown that TANK-protein is expressed in intestinal macrophages in patients without intestinal inflammation. In patients with Crohn�s disease TANK-protein is expressed in other cells than macrophages, but these cells have be to further analysed. Upregulation of TANK and simultaneous downregulation of TBK1 in intestinal macrophages might be a mechanism to inhibit TRAF2-mediated NF-kappaB-activation, which leads to the differentiation of an inactive type of macrophage. Absence of TANK in intestinal macrophages in patients with Crohn�s disease might lead to enhanced TRAF2-mediated NF-kappaB-activity.
