The present work was aimed at the development of copper-(I)-bisoxazoline catalyzed cyclopropanation of furans as a key step towards the stereoselective synthesis of a cis-fused furo[2,3-b]furan intermediates which can be broadly applied for the synthesis of spongiane diterpenoids such as norrisolide.
Norrisolide was isolated in 1983 by Faulkner, Clardy and coworkers in a study of the chemical defense system of nudibranch molluscs and has also been reported to vesiculate the Golgi apparatus irreversibly. However, the detailed synthetic as well as biological studies of such spongiane diterpenoids is rather unexplored.
Asymmetric cyclopropanation of the 3-methyl-2-furoic ester was developed with excellent regio-, diastereo- and enantioselectivity. Conversion of the resulting 1,2-cyclopropanecarboxylated dihydrofuran to the corresponding allylic bromide allowed to enhance the enantiomeric excess to >99% ee. Moreover, the allylic bromide, being a versatile functionality could be further manipulated to various enantiomerically pure 1,2-cyclopropanecarboxylated dihydrofurans.
The stereoselective synthesis of cis-fused furo[2,3-b]furans was achieved as an application of these derivatives, which gave intermediates, relevant for the synthesis of norrisolide as well as various spongiane diterpenoids containing distinctive furo[2,3-b]furan framework. The strategy followed was predominantly based on acid-mediated one-pot ring expansion of the corresponding cyclopropanes. The main advantage of the strategy is the orientation of the functional group in such a way that the side-chains could be introduced on concave as well as convex face of the bicycle.
Furthermore, applications of 1,2-cyclopropanecarboxylated furanoids in a variety of cyclopropane rearrangements giving stereocontrolled access to THF-containing unnatural amino acids as well as enantiomerically pure dihydrofuran derivatives were investigated as an additional part of this work.
In conclusion, the stereoselective synthesis of 1,2-cyclopropanecarboxylated furanoids and their applications in the preparation of marine natural products and unnatural amino acids was carried out.

Die vorliegende Arbeit beschreibt die Entwicklung Kupfer(I)-Bisoxazolin katalysierter Cyclopropanierungen von Furanen als Schlüsselschritt der stereoselektiven Synthese von cis-verknüpften Furo[2,3-b]furan Zwischenstufen welche weite Anwendung in der Synthese von Diterpenoiden wie Norrisolid finden.
Norrisolid wurde 1983 von Faulkner, Clardy et al. während Studien über das chemische Verteidigungssystem von Nacktschnecken und Mollusken isoliert. Jedoch sind detaillierte Synthesen und biologische Studien dieser Art von Diterpenoiden noch weitgehend unbekannt.
Asymmetrische Cyclopropanierung von 3-Methyl-2-furancarbonsäureestern wurde mit exzellenter Regio-, Diastereo-, und Enantioselektivität erreicht. Die Umsetzung des entstehenden 1,2-Cyclopropancarboxylierten Dihydrofuran zum entsprechenden Allylbromid ermöglichte eine Erhöhung des enatiomeren Überschusses auf über 99%. Desweiteren konnte das vielseitig einsetzbare Allylbromid weiter zu verschiedenen enantiomerenreinen 1,2-cyclopropancarboxylierten Dihydrofuranen umgesetzt werden.
Die stereoselektive Synthese von cis-verknüpften Furo[2,3-b]furanoiden wurde als eine der Anwendungen genutzt. Diese Verbindungen sind interessante Zwischenstufen in der Synthese von Norrisolid und anderen Diterpenoiden welche dieses Gerüst enthalten. Die hauptsächlich verfolgte Stratgie war eine säure-vermittelte Ringerweiterung der entsprechenden Cyclopropane. Der Hauptvorteil dieser Strategie liegt in der Orientierung der funktioniellen Gruppen welche die Einführung von Seitenketten von der konvexen Seite des Bicyclus ermöglichen.
Weitere Untersuchungen der Anwendungsgebiete von 1,2-Cyclopropancarboxylierten Dihydrofuranoiden ergab ein weites Spektrum von Cyclopropanumlagerungen zu THF-beinhaltenden unnatürlichen Aminosäuren und enantiomerenreinen Dihydrofuran Derivaten.
In dieser Arbeit wurde also zusammenfassend die stereoselektive Synthese von 1,2-cyclopropancarboxylierten Dihydrofuranoiden und deren Anwendung in der Darstellung von marinen Naturstoffen und unnatürlichen Aminosäuren behandelt.
