Abstract (German)
1995 beschrieben Sakaguchi et al. eine Subpopulation von CD4+ T-Zellen, die die a-Kette des IL-2 Rezeptors (CD25) exprimiert und eine starke regulatorische Aktivität aufweist (Sakaguchi S et al., 1995, J.Immunol. 155:1151-1164). In zahlreichen Studien konnte seither bestätigt werden, dass CD4+ CD25+ T-Zellen entscheidend an der Aufrechterhaltung der immunologischen Selbsttoleranz beteiligt sind. Neben den CD4+ CD25+ T-Zellen wurden auch weitere Zellpopulationen mit einer regulatorischen Funktion nachgewiesen, darunter unter anderem die TCRab+ CD4- CD8- (doppeltnegativ, DN) T-Zellen. Im Mausmodell konnte gezeigt werden, dass DN T-Zellen Effektorzellen Antigen-spezifisch supprimieren und dadurch eine Abstoßung von Transplantaten verhindern können. In der vorgelegten Arbeit konnte beschrieben werden, dass die Frequenz an humanen DN T-Zellen im peripheren Blut von gesunden Spendern circa 1% aller T-Lymphozyten beträgt. Durch quantitative TREC-Analysen wurde nachgewiesen, dass humane DN T-Zellen nicht frisch aus dem Thymus ausgewandert sind, sondern eine proliferative Vorgeschichte aufweisen. Zudem wurde ein DN T-Zellklon mit bekannter Antigenspezifität charakterisiert. Dieser Klon konnte aus einem Melanom-Patienten nach Vakzinierung mit dem gp100 Peptid isoliert werden. Nach einer spezifischen Stimulation produzierte der DN Klon überwiegend die Zytokine IFN-g und TNF. Dieses Zytokinprofil konnte auch nach Aktivierung von frisch isolierten humanen DN T-Zellen beobachtet werden. Eine Untersuchung der zytotoxischen Aktivität des DN T-Zellklons zeigte eine starke Antigen-spezifische Zytotoxizität sowohl gegen exogen beladene Zielzellen als auch gegen Tumorzellen, die das Protein endogen prozessieren und auf ihrer Oberfläche präsentieren. Im Gegensatz zu den Daten im Maussystem wird diese zytotoxische Aktivität nicht über Fas/FasL Interaktion sondern über die Effektormoleküle Perforin und Granzyme-B vermittelt. Im zweiten Teil der vorliegenden Arbeit wurde die immunregulatorische Funktion humaner DN T-Zellen analysiert. Wir konnten erstmalig zeigen, dass humane DN T-Zellen nach repetitiver Stimulation mit allogenen DCs in der Lage sind, alloreaktive CD4+ und CD8+ T-Zellen zu supprimieren. Zudem konnten humane DN T-Zellen auch die Proliferation von aktivierten Effektorzellen inhibieren. Mit Hilfe von Transwell-Experimenten konnte nachgewiesen werden, dass der suppressive Effekt humaner DN T-Zellen nicht über Zytokine oder andere lösliche Faktoren sondern über einen Zell-Zell-Kontakt abhängigen Mechanismus vermittelt wird. Weiterhin zeigen unsere Ergebnisse, dass humane DN T-Zellen im Gegensatz zu murinen DN T-Zellen nicht die reaktiven Zellen durch Fas/FasL Interaktion eliminieren, sondern die Effektorzellen aktiv supprimieren.

Regulatory T lymphocytes play an important role in the maintenance of immune tolerance to self antigens, and are involved in downregulating immune responses in autoimmunity, transplant rejection, tumor immunity and infectious diseases. Numerous studies demonstrated that many distinct T cell subsets possess an immunoregulatory function. Among them, the population of CD4+ CD25+ T cells is currently the most highly investigated subset and their role has been studied in a wide range of animal models and in humans. Recently, a novel subset of TCRab+ CD4- CD8- (double negative, DN) T cells was characterized to specifically suppress immune responses in both mouse and humans. In the present thesis, we show that about 1% of human peripheral T cells offer the DN phenotype. Quantitative TREC analysis indicated that DN T cells are not recent thymic emigrants and have a proliferative history. Furthermore, we analyzed a human DN T cell clone isolated from the peripheral blood of a melanoma patient vaccinated with a gp100 peptide. We found that the DN T cell clone predominantly secretes IFNg upon antigen recognition. This cytokine profile was also detected after allogenic stimulation of freshly isolated DN T cells. Although lacking the CD8 coreceptor, the DN T cell clone exhibit a strong antigen-specific cytotoxic activity against gp100-pulsed target cells as well as gp100-expressing tumor cell lines. This cytolytic effect is mediated via the effector molecules perforin and granzyme-B. Moreover, we examined the immunoregulatory function of human DN T cells. In our studies we could show that human DN T cells are able to suppress alloreactive CD4+ or CD8+ T cells in a mixed lymphocyte reaction. Furthermore, DN T cells also inhibit the proliferation of activated effector T cells. Using transwell-assays we demonstrated that human DN T cells are not able to suppress CD4+ T cells through secretion of cytokines or other soluble factors. However, the mechanism by which human DN T cells mediate suppression still remains unclear. In contrast to murine DN T cells, which eliminate effector T cells via fas/fasL interaction, human DN T cells suppress the proliferation of CD4+ or CD8+ T cells in an active manner.
