Abstract (German)

Die circadiane Rhythmik von Drosophila ist ein weites Forschungsgebiet, das in der Vergangenheit viel dazu beigetragen hat, die Mechanismen von inneren Uhren im Allgemeinen zu verstehen. Leider gilt dies kaum im Bereich der Weitergabe des zentralen Rhythmus an untergebene Strukturen oder der Kommunikation von Rhythmusgebern untereinander. Diese Arbeit sollte als Grundlagenforschung die Funktionen und Eigenschaften von dem Neuropeptid Pigment Dispersing Factor (Pdf) genauer beleuchten, welches in der Literatur als potentieller Kopplungs- und Outputfaktor gehandelt wird. Es galt, unterschiedlichste Herangehensweisen an das Thema PDF miteinander zu verbinden und damit ein klareres Bild von der Aufgabe dieses Neuropeptids im circadianen Gefüge zu erhalten.
Als sicherlich erstaunlichstes Ergebnis ist die Tatsache zu werten, dass nach dem Verlust von PDF nur bei hohen Temperaturen und dem Verlust der sLNv bereits bei Raumtemperatur Nachtaktivität im LD auftritt, die möglicherweise durch eine Entkopplung im dorsalen Gehirn dieser Komponente vom Rest der Aktivität zustande kommt. Ein Nebenergebnis dieser Versuche ist die Erkenntnis, dass es noch weitere Kopplungsfaktoren im circadianen System von Drosophila geben muss. Des Weiteren konnte herausgefunden werden, dass ektopisches PDF in der akzessorischen Medulla stark verlangsamend auf die innere Uhr wirkt und kein Splitting verursacht, d.h. weiterhin die nachgeschalteten Zellen miteinander koppelt.
Als Resumee meiner Versuche darf man vermuten, dass von den lLNv ausgeschüttetes PDF in der akzessorischen Medulla über die PDF-Rezeptoren der sLNv auf die innere Uhr zurückwirkt und diese verlangsamt. Gleichzeitig koppelt es über die dorsalen Terminalen der sLNv einen Teil der morgendlichen lokomotorischen Aktivität an restliche Aktivität.

The circadian Rhythm in Drosophila is a vast field of research, which has contributed a lot to the understanding of endogenous clocks in the past. Unfortunately this is only partly true in the fields of the transduction of the major rhythm to subordinate structures and the communication of pacemakers amongst each other. This basic research study intended to reveal more about the functions and properties of the neuropeptide Pigment Dispersing Factor (Pdf), which has been proposed as a potential coupling and output factor. Various approaches had to be combined in order to get a clearer picture of the role of Pdf in the circadian network.
The most striking result of this study is the fact that nocturnal activity occurs in high temperatures if the flies lack Pdf and at room temperature, when the sLNv have been ablated. This might be due to a decoupling of this rhythm component from the rest of the activity in the dorsal brain. This result triggers the realization that there has to be another coupling factor in the circadian system of Drosophila. It was revealed, that ectopic Pdf in the accessory medulla has a strongly retarding effect on the endogenous clock, without causing any Splitting. This means that the coupling effect still works on the downstream neurons.
Those results indicate that Pdf which is liberated by the lLNv into the accessory medulla retroacts to the endogenous clock via Pdf-receptors on the sLNv and thus is slowing the clock down. At the same time, Pdf is coupling parts of the locomotory morning activity to the rest of the activity via the dorsal terminals in the sLNv.
