Das Lesch Nyhan Syndrom wird durch einen Enzymdefekt der Hypoxanthin-Phosphoribosyltransferase (HPRT) hervorgerufen. Es zeigen sich klinisch erhöhte Harnsäurewerte im Blut, neurologische Veränderungen und Verhaltensabnormitäten, welche ein autoaggressives Verhalten einschließen. Das Lesch Nyhan Syndrom wird durch einen Enzymdefekt der Hypoxanthin-Phosphoribosyltransferase (HPRT) hervorgerufen. Es zeigen sich klinisch erhöhte Harnsäurewerte im Blut, neurologische Veränderungen und Verhaltensabnormitäten, welche ein autoaggressives Verhalten einschließen. Die Verbindung des Enzymdefekts zu den Verhaltensabnormitäten ist bis jetzt unbekannt. In HPRT-defizienten B103- und Neuro2a-Zellen ist die Aktivität der Nukleosid 5´-Triphosphatase (NTPase) erniedrigt, wogegen sie in Mausfibroblasten erhöht ist. Da mögliche Gene dafür die Ectonukleotidasen (NTPDasen) wären, wurde in dieser Arbeit die Genexpression der NTPDasen mittels Polymeraserkettenreaktion untersucht. Die Ergebnisse zeigten einerseits Erhöhungen und andererseits Erniedrigungen der Genexpression innerhalb der HPRT-defizienten Isoformen, welche vom Zelltyp und der Zellspezies abhängig waren. Diese Veränderungen in der NTPDase-Genexpression könnten einen (insuffizienten) Kompensierungsversuch für die Veränderungen des Nukleotidstoffwechsels widerspiegeln.

Lesch-Nyhan disease ist caused by a deficiency of hypoxanthine phosphoribosyl transferase (HPRT). The link between HPRT deficiency and the neurophysiatric symptoms is unknown. In rat B103 cell membranes and mouse Neuro2a cell membranes, nucleoside5´triphosphatase (NTPase) activitiy is substantially reduced, whereas in fibroblast membranes from HPRT knock-out mice NTPase activity is increased. ...
Lesch-Nyhan disease ist caused by a deficiency of hypoxanthine phosphoribosyl transferase (HPRT). The link between HPRT deficiency and the neurophysiatric symptoms is unknown. In rat B103 cell membranes and mouse Neuro2a cell membranes, nucleoside5´triphosphatase (NTPase) activitiy is substantially reduced, whereas in fibroblast membranes from HPRT knock-out mice NTPase activity is increased. Candidate genes for these NTPase activity changes are ecto-nucleoside 5´-triphosphate diphosphohydrolases (NTPDases). Therefore, we studied the expression of NTPDases by polymerase chain reaction. Collectively, there are complex decreases an increases in NTPDase isoform expression in HPRT deficiency that depend on the specific cell type an species studied. These changes in NTPDase expression may reflect an (insufficient) attempt to compensate for the changes in nucleotide metabolism caused by HPRT deficiency.
