Abstract (German)
Im Rahmen dieser Dissertation sollte die Rolle des sezernierten Proteins MIA (�Melanoma Inhibitory Activity�) bei der Entstehung und Progression des malignen Melanoms näher aufgeklärt werden. Dabei wurde die Regulation der Transkription und Aktivität von MIA auf molekularbiologischer Ebene analysiert, um ein besseres Verständnis von der Entstehung des Melanoms zu erlangen.
Es ist bekannt, dass MIA mit Proteinen der extrazellulären Matrix sowie mit Integrinen interagieren kann und so zum Ablösen der Zellen von der Matrix führt. Auf der Suche nach weiteren Interaktionspartnern von MIA wurde mittels SELDI-TOF-Massenspektrometrie und Ko-Immunpräzipitation Cadherin-7 als neuer Bindungspartner identifiziert. Das klassische Zell/Zell-Adhäsionsmolekül Cadherin-7 wird von Melanomzellen gebildet, wobei eine verstärkte Expression im Primärtumor im Vergleich zu metastatischen Zellen festgestellt wurde. Eine Verminderung der Expression von Cadherin-7 in einer Zelllinie aus einem Primärtumor führte dabei zu einer verringerten Migration der Zellen. Die Migrationseffekte, die MIA im Melanom ausübt, konnten durch Überexpression von Cadherin-7 aufgehoben werden. Interessanterweise konnten wir durch Cadherin-7 auch eine Regulation der mRNA-Expression von MIA feststellen, wobei Cadherin-7 die Expression von MIA hemmt. Cadherin-7 kann sowohl die Aktivität als auch die Expression von MIA regulieren und somit die Migration von Melanomzellen beeinflussen. Die Daten deuten darauf hin, dass Cadherin-7 die frühe Entwicklung des Melanoms fördert und dagegen im Laufe der Tumorprogression die Funktion von MIA an Bedeutung gewinnt.
Um die transkriptionelle Regulation von MIA weiter zu untersuchen, wurde der Ko-Repressor CtBP1 analysiert. Von unserer Arbeitsgruppe wurde bereits gezeigt, dass CtBP1 in Melanozyten exprimiert wird und hier die Expression von MIA hemmt. In CtBP1-exprimierenden Zellklonen konnte die Expression von MIA und das Metastasierungspotential der Melanomzellen reduziert werden. In weiteren Analysen entdeckten wir im Melanom eine Spleißvariante des Transkriptionsfaktors CtBP1. Es zeigte sich hierbei, dass diese Spleißvariante nicht mehr an TCF4, das die Expression von MIA reguliert, und an Snail binden kann. Somit kann in Melanomzellen die Repressorfunktion von CtBP1 nicht mehr ausgeübt werden.
Um die Effekte von CtBP1 besser verstehen zu können, haben wir mögliche Zielgene untersucht, die im Melanom stark exprimiert werden. Basierend auf Microarray-Daten haben wir uns auf 14 Gene fokussiert, wobei von den meisten Genen bereits bekannt ist, dass sie an der Metastasierung von Tumoren beteiligt sind. Es ist sehr interessant, dass elf Gene Expressionsmuster besitzen, die mit der Invasivität von Melanomzellen in Einklang gebracht werden können. Dieser Teil der Arbeit zeigte, dass eine verminderte Expression von CtBP1 mit dem Potential zur Migration und Invasion von Melanomzellen korreliert, wobei die Spleißvariante von CtBP1 die Expression von MIA nicht inhibieren kann.
Weitere Untersuchungen im Rahmen dieses Projekts zeigten, dass die Aktivität von CtBP1 durch Hypoxie reguliert wird. Der Transkriptionsfaktor HIF ist der entscheidende Regulator für die Effekte bei Sauerstoffmangel, wobei HIF auch bei der Tumorprogression von Bedeutung ist. Interessanterweise stellte sich heraus, dass im Melanom im Vergleich zu anderen Tumoren bereits unter normalen Sauerstoffbedingungen eine hohe Aktivität eines HRE-Reporter-Konstrukts, das mehrere HIF-Bindestellen enthält, vorliegt. Durch Versuche mit siRNAs und Inhibitoren stellte sich heraus, dass HIF-1alpha für eine Hypoxie-unabhängige Regulation von Genen im Melanom verantwortlich zu machen ist. Das Protein HIF-1alpha weist dabei im Melanom neben vermehrter Aktivität auch eine hohe Stabilität auf.
Um die Analysen der Hypoxie-unabhängigen Regulation von HIF-1alpha im Melanom zu vertiefen, schlossen wir eine Behandlung der Melanomzellen mit Radikalfängern an. Es zeigte dabei eine deutliche Verringerung der Aktivität von HIF-1alpha, so dass eine Beteiligung von reaktiven Sauerstoffspezies (ROS) an diesem Effekt aufgezeigt wurde. Über weitere Analysen wurde festgestellt, dass ROS den Transkriptionsfaktor NFkappaB, der im Melanom stark exprimiert wird, induzieren und so für die hohe Aktivität von HIF-1alpha von Melanomzellen unter Normoxie verantwortlich ist. Diese hohe Aktivität von HIF-1alpha kann die Tumorprogression des malignen Melanoms fördern.
Zusammenfassend konnte diese Promotionsarbeit weitere Aspekte der Regulation der Transkription und Aktivität von MIA im malignen Melanom aufklären. Zusätzlich konnten weitere wichtige Regulationsmechanismen, die an der Entwicklung des Melanoms beteiligt sind, aufgeklärt werden.

In this work the role of the secreted protein MIA (�Melanoma Inhibitory Activity�) in development and progression of malignant melanoma should be further elucidated. Thereby, the regulation of transcription and activity of MIA was analysed on molecular level. It is known that MIA can interact with integrins and proteins of the extracellular matrix.
We identified cadherin-7 as a new MIA-binding protein using surface-enhanced laser desorption/ionization-mass spectrometry technology and co-immunoprecipitation. We demonstrated enhanced expression of the classical cell-cell adhesion molecule cadherin-7 in primary tumor cells compared to metastatic cells. Downregulation of expression in cells derived from primary melanomas resulted in reduced cell migration. In addition, MIA effects on cell migration were abrogated in cell clones overexpressing cadherin-7. Interestingly, overexpression of cadherin-7 resulted in a decreased MIA mRNA expression. In conclusion, these findings suggest that cadherin-7 regulates the expression and activity of MIA and the migration of melanoma cells playing a role in tumor development of malignant melanoma.
To further investigate the regulation of MIA on transcriptional level, the co-repressor CtBP1 was analysed. We recently revealed that CtBP1 expression is lost in melanoma cells and MIA expression is subsequently increased. In CtBP1 expressing cell clones the expression of MIA and the metastatic potential of melanoma cells was reduced. We also detected an alternative splice product of CtBP1 with unknown function. This splice variant can not bind to TCF4, which regulates the expression of MIA, revealing a loss of repressor function of CtBP1 in melanoma cells.
To understand the effect of CtBP1 we identified putative LEF/TCF target genes found to be strongly expressed in melanoma using DNA microarray analysis. We focused on fourteen genes that are expressed in melanoma cells. Detailed analysis revealed that most of these genes were known to be involved in tumor metastasis. Eleven genes had expression profiles associated with melanoma cell invasiveness. In summary, this part of the study revealed that reduction of CtBP1 expression is correlated with migratory, invasive potential of melanoma cells, in which the splice variant of CtBP1 can not inhibit the expression of MIA.
Further analysis showed that the activity of CtBP1 could be regulated by hypoxia. The hypoxia-inducible factor 1 (HIF-1) is a transcription factor that plays a major role in gene regulation and tumor progression under hypoxia. Interestingly we showed an increased HIF activity in melanoma cells in comparison to other tumor types under normoxic conditions. Transfection experiments with siRNAs and inhibitors demonstrated that this effect is due to HIF-1alpha. The protein HIF-1alpha showed also high stability in melanoma cells. The inhibition of reactive oxygen species (ROS) with scavengers decreased the HIF activity and the HIF1alpha protein expression. Interestingly, the inhibition of the NFkappaB pathway also reduced the accumulation of HIF-1alpha assuming a context between ROS and NFkappaB. Additionally, we identified an increased HIF-1alpha accumulation in melanoma under normoxia mediated by ROS and the NFkappaB pathway. This increased activity of HIF-1alpha can further promote the progression of malignant melanoma.
Overall, this work could further elucidate the regulation of transcription and activity of MIA as well as the development of malignant melanoma.
