Abstract (German)

Im Rahmen dieser Untersuchung sollte die transkriptionelle Regulation des ABCA1-Promotors durch den Transkriptionsfaktor HIF1 in den für den Lipidstoffwechsel relevanten Zellen HepG2 und RAW 264.7 nachgewiesen werden. Mittels DNA-Mikroarrays konnte bereits eine ABCA1-Regulation unter hypoxischen Bedingungen und durch Überexpression einer konstitutiv aktiven Form von HIF1 alpha; in menschlichen Endothelzellen [54] und unter hypoxischen Bedingungen in RPTEC-Zellen [69] gezeigt werden.
Die Ergebnisse dieser Arbeit lassen insgesamt darauf schließen, dass HIF1 eine Rolle bei der Regulation des ABCA1-Promotors spielt. Durch Luciferase- Promotor-Assyas konnte eine vermehrte ABCA1-Expression in HepG2- und RAW264.7-Zellen durch den Transkriptionsfaktor HIF1 bestätigt werden. Außerdem zeigte sich eine direkte Bindung des Promotors durch die funktionelle Einheit, bestehend aus HIF1 alpha und ARNT, im Gelshift-Assay an die E-Box. Auch Krishnamurthy et al. konnten eine direkte Regulation des ABCG2-Promotors durch HIF1 mittels EMSA an das proximale HRE-Element (-116bp) des Promotors nachweisen. Da beide Promotoren ein E-Box Element besitzen, könnten auch andere Promotoren, die diese Sequenz enthalten, durch HIF1 reguliert werden.

Translation of the abstract (English)

ABCA1 is one of the major regulators of hepatic and peripheral cholesterol metabolism. Changes in ABCA1 expression greatly influence cholesterol production in the liver and reverse cholesterol transport from peripheral tissues back to the liver, which is closely related to manifestation of atherosclerosis. Atherosclerotic lesion associated local as well as systemic hypoxia adversely affects lesion development.
In this study, the regulation of the ABCA1 gene by the hypoxia sensitive HIF-1 nuclear receptor complex was analyzed. We found that HIF-1 complex specifically binds to the E-box motif of the ABCA1 promoter (-229/-210), which induces ABCA1 transcription in HepG2 and RAW 264.7 cells. Basal ABCA1 promoter activity was also found low in macrophages and intensive in liver cells, which could be induced by the upregulation of the HIF-1 complex in both cell types. The expression of VEGF was induced in both cell types by HIF-1alpha upregulation, as control.
