Wir stellten uns zum einen die Frage, ob auch PXR-Agonisten, wie Rifampicin oder LCA, in intestinalen Zellen FGF19 hochregulieren. Fragestellung: Bislang war bekannt, dass FXR-Agonisten, z.B. CDCA, einen positiven regulierenden Effekt auf die Synthese von FGF19 ausüben. Wir stellten uns zum einen die Frage, ob auch PXR-Agonisten, wie Rifampicin oder LCA, in intestinalen Zellen FGF19 hochregulieren. Zum anderen sollte anschließend die PXR responsive Promotorregion des FGF19-Gens näher charakterisiert werden.
Methoden: Im ersten Schritt wurden LS174T-Darmzellen mit verschiedenen Konzentrationen zum einen des typischen FXR-Agonisten CDCA als Positivkontrolle, zum anderen mit den PXR-Agonisten LCA in einer Zeitreihe stimuliert. Anschließend wurde mit Taqman real-time RT-PCR die relative mRNA-Expression von FGF19 bestimmt.
Im zweiten Schritt wurden verschiedene Deletionskonstrukte der FGF19- Promotorregion in Luciferase-Reporterplasmide integriert. In anschließenden Stimulationen mit Rifampicin und LCA wurde die Reaktion auf Rifampicin und LCA mittels luciferase assays gemessen.
In einem dritten Schritt wurden die Stimulationsansätze um Co-Transfektion mit den Expressionsplasmiden pSG5-hPXR und pSG5-hRXR erweitert.
Ergebnisse: Sowohl CDCA als auch LCA regulieren FGF19 dosis- und zeitabhängig hoch. Mithilfe von luciferase assays und verschieden langer Deletionskonstrukte konnten zwei mögliche proximale PXR-Binderegionen im FGF19-Gen identifiziert werden. Schließlich zeigten Rifampicin- und LCAStimulationen bei gleichzeitiger Überexpression von PXR und dessen Dimerisationspartner RXR eine deutliche Induktion der FGF19- Promotoraktivität.
Schlussfolgerung: Es ist wahrscheinlich, dass LCA in einer negativen Feedback-Schleife durch PXR die Expression von FGF19 induziert und so über den enterohepatischen Kreislauf in die Regulation der Gallensäuresynthese eingreift.
Translation of the abstract (English)
AIM: To study the effect of the toxic secondary bile acidlithocholic acid(LCA) on the expression of fibroblast growth factor 19 (FGF19) in intestinal cells and to characterize the pregnane-X-receptor (PXR) response of the FGF19 promoter region. AIM: To study the effect of the toxic secondary bile acidlithocholic acid(LCA) on the expression of fibroblast growth factor 19 (FGF19) in intestinal cells and to characterize the pregnane-X-receptor (PXR) response of the FGF19 promoter region.
METHODS: The intestinal cell line LS174T was stimulated with various concentrations of chenodeoxycholic acid and lithocholic acid for several time points. FGF19 mRNA levels were determined with quantitative realtime RT-PCR. FGF19 deletion promoter constructs were generated and the LCA response was analzyed in reporter assays. Co-transfections with PXR and RXR were carried out to study FGF19 regulation by these factors.
RESULTS: LCA and CDCA strongly up-regulate FGF19 mRNA expression in LS174T cells in a time and dose dependent manner. Using reporter gene assays with several deletion constructs we found that the LCA responsive element in the human FGF19 promoter maps to the proximal regulatory region containing two potential binding sites for PXR. Overexpression of PXR and its dimerization partner retinoid X receptor (RXR) and stimulation with LCA or the potent PXR ligand rifampicin leads to a signifi cant induction of FGF19 promoter activity in intestinal cells.
CONCLUSION: LCA induced feedback inhibition of bile acid synthesis in the liver is likely to be regulated by PXR inducing intestinal FGF19 expression.
