Bone morphogenetic proteins (BMPs) sind pleiotrope Morphogene aus der TGF-ß Superfamilie, die über einen Konzentrationsgradienten vielfältige Effekte wie Proliferation, Differenzierung, Apoptose und Migration unterschiedlicher Zelltypen regulieren.

Sie spielen eine Schlüsselrolle in mehreren Stadien der embryonalen Hämatopoese, etwa bei der Bildung erster Blutinseln aus ventralem Mesoderm durch BMP-4. Der Einfluss von BMPs auf postnatale hämatopoetische Stammzellen ist demgegenüber weit weniger klar.

In der vorliegenden Arbeit wurde der Einfluss von BMP-2, BMP-4 und BMP-7 auf eine primitive CD34+/CD38- Lin- Stammzellpopulation aus humanem, mit G-CSF stimulierten, peripheren Blut untersucht. Die Charakterisierung der hämatopoetischen Vorläuferzellen erfolgte sowohl anhand von Oberflächenproteinen als auch in funktionellen Assays (CFC-Kurzzeitkultur, LTC-IC-Langzeitkultur). Die Kapazität dieser Zellpopulation, auf BMP-Signale zu reagieren, wurde durch den Nachweis von BMP-Rezeptoren gezeigt. Im Vergleich der generierten Zellpopulationen vor und nach einwöchiger Kultur mit BMP-2, BMP-4 oder BMP-7 in unterschiedlichen Konzentrationen, ergaben sich Hinweise auf eine Hemmung des zu erwarteten Verlusts von LTC-IC durch BMP-2 in niedriger Konzentration. Weiterhin scheinen BMP-2 und BMP-4 in diesem Zusammenhang synergistisch zu wirken.

Bone morphogenetic proteins (BMPs) are pleiotropic morphogens belonging to the TGF-ß superfamily that cause multiple effects �like proliferation, differentiation, apoptose or migration- of different cell types.

They play a crucial role during embryonic hematopoesis: BMP-4 governs the development of primitive blood islands from ventral mesoderm. The influence of BMPs on postnatal hematopoetic stem cells is still elusive.

In this study, the influence of BMP-2, BMP-4 and BMP-7 on primitive, peripheral blood derived, human CD34+/CD38- Lin- hematopoetic stem cells (HSC) was investigated. Characterization of HSC was accomplished by flow cytometry and functional assays (CFC short-time culture, LTC-IC long-time culture). Detection of BMP receptors in the applied cell population showed the capacity of these cells to react on BMP-signalling. Comparing the features of the generated cell populations before and after seven days of culture with BMP-2, BMP-4 or BMP-7 in distinct concentrations gave evidence of BMP-2 inhibiting the loss of primitive LTC-IC. Furthermore, there was indication of the capacity of BMP-2 and BMP-4 to act syergistically.
