Gegenstand der vorliegenden Dissertation ist die Analyse der Bedeutung einer neuen Klasse kleiner, nicht-codierender RNAs, den microRNAs (miRNAs), bei der Entstehung und Progression des malignen Melanoms.
Im Rahmen dieser Arbeit konnten zwei miRNAs identifiziert werden, welche als wichtige Tumorsuppressoren im Melanom fungieren. Eine dieser miRNAs ist let-7a. Es konnte gezeigt werden, dass die Regulation der Expression von Integrin beta3 in Melanomzellen auf posttranskriptioneller Ebene durch diese miRNA erfolgt, während Promotor-abhängige Mechanismen nur eine untergeordnete Rolle spielen. Der Verlust von let-7a im Melanom führt zur Induktion der Integrin beta3 Expression und trägt dadurch wesentlich zum invasiven Potenzial von Melanomzellen bei. Das Einbringen synthetischer let-7a Moleküle führt zu einer starken Reduktion der Invasivität von Melanomzellen und könnte somit als neuartiger therapeutischer Ansatz verfolgt werden.
Weiterhin konnte gezeigt werden, dass die Expression einer zweiten miRNA, miR-196a, im malignen Melanom stark vermindert vorliegt. Dies führt zu einem Anstieg des in normalen humanen Melanozyten durch miR-196a reprimierten Homeobox-Transkriptionsfaktors HOX-C8 in Melanomzellen. Durch die verstärkte Expression von HOX-C8 werden mindestens drei weitere, Tumor-relevante Gene, nämlich Osteopontin, Cadherin11 und Calponin-1, in Melanomzellen fehlreguliert. Durch die Reprimierung von HOX-C8 und dem daraus resultierenden indirekten Einfluss auf eine Vielzahl Tumor-assoziierter Gene, kommt miR-196a im Melanom eine Rolle als potenter Tumorsuppressor zu. Dies konnte in preliminären Mausexperimenten, in welchen die Re-Expression von miR-196a in Melanomzellen die Tumorbildung in vivo signifikant verzögerte, bestätigt werden.
Im dritten Teil dieser Arbeit wurden miRNA-Expressionsprofile von normalen humanen Melanozyten und etablierten Melanomzelllinien generiert. Durch Abgleichen der miRNA-Expressionsprofile und unter zusätzlicher Verwendung von in vitro Melanom-Modellsystemen, konnte eine große Anzahl miRNAs identifiziert werden, deren Fehlexpression mit verschiedenen Schritten der Melanomprogression assoziiert ist. Interessanterweise wurde dabei der überwiegende Teil deregulierter miRNAs in Melanomzellen verstärkt exprimiert. Der Großteil der identifizierten miRNAs wurde bisher nicht in Verbindung mit Prozessen der Tumorentstehung oder -progression im Allgemeinen und der Tumorigenese des malignen Melanoms im Speziellen diskutiert. Weitere, funktionelle Analysen dieser miRNAs könnten neue Einsichten in die Mechanismen der Entstehung und Progression des malignen Melanoms ermöglichen.

Translation of the abstract (English)

Object of this dissertation is the characterization of the role of a new class of small, non-coding RNAs – the microRNAs (miRNAs) – in the formation and progression of malignant melanoma.
Within the scope of this research two miRNAs were identified which act as important tumour suppressors in melanoma. One of those miRNAs is let-7a. It was shown that the expression of integrin beta3 is regulated on a posttranscriptional level by this miRNA whereas promoter-dependent mechanisms play only a minor role. The loss of let-7a in malignant melanoma results in an induction of integrin beta3 expression and, therefore, essentially contributes to the invasive potential of melanoma cells. Transfection of synthetic let-7a molecules results in a strong reduction of the invasiveness of melanoma cells and could thus build the fundament for new therapeutic approaches.
During this work it was further shown that the expression of another miRNA, miR-196a, is strongly reduced in malignant melanoma. This leads to an increased expression of the transcription factor HOX-C8 which in normal human melanocytes is repressed by miR-196a. As a consequence of the elevated levels of HOX-C8 the expression of at least three further tumour-relevant genes, namely osteopontin, cadherin11, and calponin-1 is deregulated in melanoma cells. Due to the repression of HOX-C8 and the resulting indirect effect on a multitude of tumour-associated genes, miR-196a can be said to be a potent tumour suppressor in malignant melanoma. This fact was underlined by preliminary mouse experiments in which re-expression of miR-196a in melanoma cells significantly reduced their tumour-forming potential in vivo.
As a third part of this thesis miRNA expression profiles of normal human melanocytes and established melanoma cell lines were generated. By a detailed comparison of the miRNA expression profiles created and through additionally utilizing several in vitro melanoma model systems a large number of miRNAs was identified whose misexpression is associated with different steps of melanoma progression. Interestingly, expression of most of those miRNAs was enforced in melanoma cells. The majority of miRNAs identified has neither been discussed to be related to tumour formation and progression in general nor to the tumourigenesis of malignant melanoma in special before. In the future, further functional analyses of those miRNAs could provide new insights into the mechanisms contributing to formation and progression of malignant melanoma.
