Abstract (English)

A very efficiently synthetic strategy was developed aiming to obtain a series of amino acids containing the rigid azabicyclo[3.2.1]octane scaffold in few steps, multi gram scale, and starting from commercially available compounds.
Furthermore, enantiomerically pure azabicyclo[3.2.1]octane amino acids were also prepared using a very efficient protocol consisting in the use of an asymmetric synthesis or in the use of a chiral building block as resolving agent.
Studies of orthogonal deprotection of the amino and of carboxylic functions were also developed aiming to obtain useful compounds for different applications.
In fact, these interesting amino acids were tested in a binding test as a rigid mimic of GABA, as a potential ligand for transition metals, and also as catalyst in asymmetric Diels-Alder and Staudinger reactions. The most promising application was found in the use of azabicyclo[3.2.1]octane amino acids as a alpha,alpha-disobstituted amino acids. These compounds are able to induce a well defined 310 helical structure when inserted in a short peptide sequence.

Es wurde eine sehr effiziente Synthesestrategie mit dem Ziel entwickelt, eine Reihe von Aminosäuren, die das starre Azabicyclo[3.2.1]octan-Gerüst enthalten, in wenigen Stufen, im Multigramm-Maßstab, sowie unter der Verwendung kommerziell erhältlicher Ausgangsverbindungen, herzustellen.
Desweiteren wurden auch enantiomerenreine Azabicyclo[3.2.1]octan-Aminosäuren nach einer sehr effektiven Vorschrift hergestellt, welche auf einer asymmetrischen Synthese beruht oder chirale Verbindungen zur Racematspaltung nutzt.
Darüberhinaus wurden Untersuchungen zur orthogonalen Entschützung der Amino- und Carboxyfunktionen durchgeführt, um nützliche Verbindungen für unterschiedlichste Anwendungen zu erhalten.
Tatsächlich getestet wurden diese interessanten Aminosäuren im Zuge eines Bindungsassays (als starres GABA - Imitat), als möglicher Ligand für Übergangsmetalle und als Katalysator in asymmetrischen Diels - Alder und Staudinger Reaktionen.
Als vielversprechendste Anwendung stellte sich die Verwendung der Azabicyclo[3.2.1]octan-Aminosäuren als alpha,alpha-disubstituierte Aminosäuren heraus. Diese Verbindungen sind in der Lage eine gut definierte 310- helikale Struktur zu induzieren, wenn sie in kurze Peptidsequenzen eingebaut werden.
