In dieser Arbeit wurde die Bedeutung des second messengers Ceramid und der Einfluss der sauren Sphingomyelinase (ASM) auf die Matrix Metalloproteinase (MMP)-1 Produktion untersucht.
In einem genetischen Modell mit ASM-defizienten humanen Hautfibroblasten konnte gezeigt werden, dass die Induktion der MMP-1 nach Stimulation mit dem pro-inflammatorischen Zytokin IL-1β durch die ASM reguliert wird. Im Gegensatz dazu zeigte die Inhibition der neutralen SMase keinen Effekt auf die MMP-1 Konzentration. Die durch IL-1β induzierte MMP-1 Produktion in Fibroblasten ist dabei vollständig von der ASM-Produktion abhängig. Die Inhibition der ASM scheint daher eine mögliche Strategie zu sein, um die MMP-1 Synthese bei Entzündungsreaktionen zu vermindern.
Bei der experimentellen Kolitis kommt es zu einer Akkumulation von Ceramid und Verringerung von LPC. Eine Folge der Ceramidbildung ist die gesteigerte Produktion von MMP-1. Die Induktion von MMP-1 durch TNF oder IL-1β wird durch Hemmung der ASM vollständig blockiert. Die ASM Hemmung scheint daher eine mögliche Strategie zu sein, die überschießende MMP-1-Produktion und möglicherweise damit die Gewebedestruktion bei Colitis ulcerosa zu reduzieren.
Die MMPs spielen auch bei der Zellinvasion und Zirrhoseenstehung eine zentrale Rolle. Darüber hinaus sind die MMPs entscheidend bei der Invasion und Metastasierung von Tumorzellen beteiligt. Die pharmakologische Hemmung der ASM durch Imipramin, hat einen Einfluss auf die Invasion hepatischer Sternzellen und Karzinomzellen. Werden hepatische Sternzellen mit Imipramin inkubiert, ist die MMP-1 Produktion nach Stimulation mit IL-1β, TNF und TGF-β gehemmt und die Invasion in eine Kollagenschicht aufgehoben. Damit ergeben sich durch die Modulation der ceramidvermittelten MMP-1 Produktion auch bei hepatischen Erkrankungen wie der Leberfibrose bzw. Zirrhose und der Entstehung des hepatozellulären Karzinoms möglicherweise neue therapeutische Angriffspunkte.

The aim of the current thesis was to investigate the importance of the second messenger ceramide and the influence of acid sphingomyelinases (ASM) in matrix metalloproteinase (MMP)-1 production.
Using a genetic model with ASM-deficient human skin fibroblasts it was shown that induction of MMP-1 after stimulation with pro-inflammatory IL-1β is regulated by ASM. In contrast, inhibition of neutral sphingomyelinase showed not an effect of MMP-1 concentration. The IL-1β induced MMP-1 production is completely ASM dependent in fibroblasts. Inhibition of ASM seems to be a potential strategy to reduce MMP-1 synthesis in inflammation.
In experimental colitis, there is an accumulation of ceramide and decrease of lysophosphatidylcholine. An important effect of ceramide generation is increased production of MMP-1. Induction of MMP-1 by TNF or IL-1β is completely blocked by ASM inhibition. ASM inhibition could be a potential strategy to avoid excessive MMP-1 production and possibly reduced tissue destruction in ulcerative colitis.
MMPs play a crucial role in cell invasion and cirrhosis formation. Moreover, MMPs are important for invasion and metastasis of tumour cells. Pharmacological inhibition of ASM by imipramine influences the invasion of hepatic stellate cells and carcinoma cells. After incubation with imipramine in hepatic stellate cells, MMP-1 production is inhibited after stimulation with IL-1β, TNF and TGF-β and invasion in coated collagen layer is abrogated. Therefore, the modulation of ceramide induced MMP-1 production could lead to therapeutic strategy in disease of the liver and liver fibrosis or liver cirrhosis even development of hepatocellular carcinoma.
