HINTERGRUND Den Zytokinen der Tumornekrosefaktorsuperfamilie (TNFSF) obliegt in hohem Maße die Regulation entzündlicher Prozesse im Organismus. LIGHT, als unlängst charakterisiertes Mitglied dieses Systems, besitzt große Bedeutung in der Pathogenese von Entzündungsreaktionen, wie sie kennzeichnend für rheumatoide Arthritis oder chronisch entzündliche Darmerkrankungen sind. In physiologischer Hinsicht erfüllt der proinflammatorisch wirkende Mediator hauptsächlich Funktionen bei der Virusabwehr, der Apoptoseinduktion und der Aufrechterhaltung der mukosalen Immunhomöostase an der inneren Grenzfläche des Körpers. Wegen seiner pathophysiologischen Relevanz in fehlgelenkten Entzündungsreaktionen und der komplexen Steuermechanismen innerhalb eines eigenen Signalnetzwerkes zieht LIGHT das besondere Interesse der Zytokinforschung auf sich. LIGHT wird ausschließlich auf Leukozyten, vornehmlich auf aktivierten T-Lymphozyten und dendritischen Zellen exprimiert. Ein nachgeschaltetes, verzweigtes System aus Ligand-Rezeptor-Wechselwirkungen beeinflusst die Intensität von Entzündungsreaktionen. Störungen dieses Netzwerkes durch konstitutive Überexpression von LIGHT verursachen Inflammation, Gewebsdestruktion und Defektbildung. Folglich könnte spezifisches pharmakotherapeutisches Targeting von LIGHT als neue Behandlungsstrategie bei chronischen Entzündungserkrankungen erfolgreich sein.
METHODEN Die Untersuchung der Einflussnahme einer LIGHT-neutralisierenden Behandlung auf Entzündungsprozesse erfolgte im Maus-Modell der akuten DSS-induzierten Colitis. Für die Experimente standen zwei monoklonale Anti-Maus-LIGHT-Antikörper (9D10 und 15B2) zur Verfügung, welche gemäß Standardhybridomtechnik nach Immunisierung von LIGHT-defizienten Mäusen mit rekombinantem Maus-LIGHT generiert wurden. Bei der Charakterisierung der Antikörpereigenschaften lag besonderes Augenmerk auf der Bindungsanalyse gegenüber der Zielstruktur in löslicher sowie membrangebundener Form und auf der Austestung einer biologisch funktionellen Antagonisierung von LIGHT. Die Prüfung der Antikörperbindungsfähigkeit erfolgte mittels Dot Blot, Western Blot und Immunpräzipitation gegenüber dem Molekül im löslichen Zustand – sodann in durchflusszytometrischen Analysen mit LIGHT-exprimierenden Zellen gegenüber dem membranständigen Protein. Zur Untersuchung der Antikörperwirkung in vivo wurde bei Mäusen durch Gabe von Dextran-Sulfat-Sodium (DSS) eine Darmentzündung induziert. Die LIGHT-Expression im Darm ist während DSS-Colitis gesteigert.
Über die Versuchsdauer hinweg wurde Tieren der Experimentalgruppe täglich Anti-LIGHT-Antikörper injiziert – die Mäuse der Kontrollgruppe hingegen erhielten eine identische Menge an IgG aus Mausserum. Der Vergleich der Entzündungsintensität zwischen den beiden Versuchsgruppen geschah anhand folgender Parameter: 1) Gewichtsverlauf, 2) Colonlänge, 3) Histologischer Score, 4) Myeloperoxidase-Aktivität in der Darmwand und 5) Zytokinexpression aus Milzzellen.
ERGEBNISSE Wichtige Aussagen über Bindungsfähigkeit und Funktionalität liefert die Charakterisierung der Antikörper in vitro. Dot Blot und Immunpräzipitation bescheinigen die Bindung an rekombinantes, lösliches Maus-LIGHT. Klon 15B2 erweist sich hierbei zusätzlich spezifisch gegen das humane Ortholog. Durchflusszytometrische Analysen mit Hilfe von transfizierten, Maus-LIGHT auf der Oberfläche exprimierenden Zellen bestätigen, dass eine Bindung der hergestellten Antikörper an das Zytokin im membrangebundenen Zustand nicht zu Stande kommt. Ihre LIGHT-neutralisierende Wirkung und damit biologische Funktionalität stellen beide Antikörperklone im zellbiologischen Stimulationsexperiment unter Beweis. Die Applikation von Antikörper 15B2 während Induktion der DSS-Colitis führt in vivo zu einer signifikant geringeren Entzündungsreaktion im Intestinum. Beim Vergleich der zwei Versuchsgruppen ergeben sich für Tiere, die keine Anti-LIGHT-Behandlung erhielten, deutlich negativere Ergebnisse bezüglich der zur Klassifikation der Erkrankungsschwere bestimmten Parameter. Damit gelingt eine Linderung der Entzündungssymptomatik in akuter DSS-induzierter Colitis durch den monoklonalen Anti-Maus-LIGHT-Antikörper 15B2.
SCHLUSSFOLGERUNG Spezifisches Targeting des proinflammatorischen Liganden aus der TNFSF, LIGHT, mit monoklonalen Antikörpern bewirkt eine signifikante Besserung der Entzündungsreaktion im Tiermodell der akuten DSS-induzierten Colitis. Dieser antiinflammatorische Behandlungseffekt präsentiert sich trotz des Unvermögens der verwendeten Antikörper das Zytokin in seiner membrangebundenen Form zu neutralisieren. Die Ergebnisse betonen die große Bedeutung von löslichem LIGHT bei der Ausbreitung akuter Entzündungsprozesse. Da sich unkontrollierte Expression von LIGHT als charakteristisch für autoreaktives Inflammationsgeschehen zeigt, könnten spezifische Eingriffe in dieses Signaltransduktionsnetzwerk zukünftig neue Therapieoptionen bei verschiedenen Autoimmunerkrankungen eröffnen.

BACKGROUND The cytokines of the TNFSF are involved in all inflammatory processes in the organism. LIGHT, the most recent characterized member of this system, has great importance in the pathogenesis of inflammatory reactions, e.g. in rheumatoid arthritis or inflammatory bowel disease. In physiological terms this mediator mainly meets functions in virus defense, induction of apotosis and maintenance of the mucosal immune homeostasis at the inner boundary of the body. Because of its pathophysiological relevance in inaccurate inflammatory responses and the Gordian control mechanisms within a particular signaling network, LIGHT is of special interest in cytokine research. LIGHT is expressed exclusively on leukocytes, primarily on activated T-lymphocytes and dendritic cells. A subsequent, complex system of ligand-receptor interactions influences the intensity of inflammatory events. Disruption of this network by constitutive expression of LIGHT causes chronic inflammation, tissue destruction and organ dysfunction. For this reason specific pharmacotherapeutic targeting of LIGHT could achieve success as novel treatment strategy in inflammatory diseases.
METHODS The effects of a LIGHT-neutralizing treatment on inflammatory processes were studied by a DSS-induced colitis model in mice. Two monoclonal antibodies (9D10 and 15B2), that had been generated according to standard hybridoma technique after immunization of LIGHT-deficient mice with recombinant mouse-LIGHT, were used for the experiments. The characterization of the antibodies’ properties focused on compound analyses towards the target in soluble and membrane-bound state as well as on the testing of a biologically functional antagonism of LIGHT. The examination of the antibody binding capacity to the molecule in soluble state was performed by Dot Blot, Western Blot and immunoprecipitation. Flow cytometric analyses with LIGHT-expressing cells were accomplished to evaluate the molecular behavior adverse the membrane-bound protein. In order to investigate the effects of an antibody treatment in vivo, an acute colitis was induced in mice by dextran sulfate sodium (DSS). Individuals suffering from DSS-colitis exhibit an increased expression of LIGHT in the gut.
The animals of the experimental group got daily injections of anti-LIGHT-antibody, while the mice of the control group obtained an identical amount of IgG from mouse serum. The intensity of inflammation and colitis between the two groups was compared using the following parameters: 1) weight loss, 2) length of the colon, 3) histological score, 4) myeloperoxidase activity in the intestinal wall, and 5) cytokine expression of spleen cells.
RESULTS The characterization of the antibodies in vitro provides important evidence concerning compound capacity and functionality. Dot Blot and immunoprecipitation confirm binding to recombinant soluble mouse-LIGHT. Clone 15B2 proves to be additionally specific for the human ortholog. Analyses by flow cytometry using cells, which express mouse-LIGHT on their surfaces, attest to the monoclonal antibodies inability of binding to the cytokine in membrane-bound state. Both antibodies demonstrate their LIGHT-neutralizing capability – and thus biological functionality – in cellular stimulation assays with the soluble protein. The application of antibody 15B2 during induction of DSS-colitis causes in vivo a significantly decrease of the intestinal inflammatory response. At the end of the test those animals, which did not get an anti-LIGHT-treatment, exhibit significantly more negative results regarding the parameters for classification of disease severity. Consequently, the symptoms of inflammation can be eased in acute DSS-induced colitis by the monoclonal anti-mouse-LIGHT antibody 15B2.
CONCLUSION Specific targeting of the proinflammatory ligand LIGHT, a member of the TNFSF, with monoclonal antibodies results in a significant alleviation of inflammatory reactions in the model of acute DSS-induced colitis. This anti-inflammatory effect occurs despite the incapacity of the antibodies used to neutralize the cytokine in its membrane-bound state on cells. The results emphasize the importance of soluble LIGHT in dissemination and increase of inflammation. Uncontrolled expression of LIGHT is characteristic of many auto-reactive inflammatory processes. In the future, specific interventions of this signaling network could open up novel options of medical treatment of various autoimmune diseases.
