Der Lymphotoxin-beta-Rezeptor (LTβR), ein Mitglied der TNF-Rezeptorfamilie, interagiert funktionell sowohl mit dem auf aktivierten B-, T- und NK-Zellen exprimierten Liganden LTα1β2 als auch mit LIGHT. Die meisten Studien beschäftigten sich bislang mit der wichtigen Funktion des LTβR bei der Entwicklung sekundärer lymphatischer Organe und der Aufrechterhaltung der lymphatischen Strukturen. Über die Bedeutung des LTβR in der Entzündungsreaktion ist bisher nicht viel bekannt. Vorangegangene Daten unserer Arbeitsgruppe haben gezeigt, dass die Aktivierung des LTβR durch LTα1β2-exprimierende T-Zellen eine wichtige Rolle in der Gegenregulation der Entzündungsreaktion im experimentellen Modell der akuten DSS-induzierten Kolitis spielt. Zudem konnte durch den Einsatz von Knochenmark-Chimären nachgewiesen werden, dass die Expression des LTβR auf Zellen hämatopoetischen Ursprungs eine entscheidende Bedeutung für die Kontrolle der intestinalen Entzündungsreaktion hat.
In der vorliegenden Arbeit wurden die zugrundeliegenden zellulären und molekularen Mechanismen, die für die verminderte Entzündungsreaktion nach der Aktivierung des LTβR verantwortlich sind, näher untersucht. Dabei konnte die Induktion des antientzündlichen Effektorgens TRIM30 als Resultat der LTβR-Signalwirkung in Makrophagen erstmalig nachgewiesen werden. TRIM30 wirkt als negativer Regulator der TLR-induzierten NFκB-Aktivierung, der einen zuvor unbekannten negativen feedback-Mechanismus vermittelt, um anhaltende und überschießende Entzündungsreaktionen zu kontrollieren. BMDM und J774-Zellen wurden als experimentelles Modell in vitro verwendet, die den LTβR auf der Zelloberfläche konstitutiv exprimieren. Die LTβR-vermittelte TRIM30-Induktion konnte in diesen Zellen nach der Stimulierung mit agonistischem anti-Maus LTβR mAK sowohl auf mRNA- als auch auf Protein-Ebene nachgewiesen werden. Zum Nachweis der TRIM30-Proteinexpression wurde ein polyklonales spezifisches anti-TRIM30 Antiserum hergestellt. In der Kokultur mit aktivierten T-Zellen, welche die Liganden des LTβR (LTα1β2 und/oder LIGHT) auf der Zelloberfläche exprimieren, konnte die TRIM30-Induktion in den LTβR-tragenden Makrophagen nachgewiesen werden.
Da TRIM30 in die LPS-induzierte Toleranz in vitro und in vivo involviert ist, wurde die Auswirkung der LTβR-Aktivierung auf die TLR-induzierte Zytokinexpression untersucht. Es konnte gezeigt werden, dass die Vorstimulierung des LTβR auf BMDM oder J774-Zellen zur signifikant reduzierten TLR-vermittelten Zytokin- und NO-Induktion in vitro führt. Diese Ergebnisse konnten in vivo bestätigt werden. Hier führte die intraperitoneale Applikation von agonistischem anti-Maus LTβR mAK ebenfalls zur Reduktion der TLR-vermittelten Entzündungsreaktion, die durch signifikant verminderte TNF-Mengen im Serum gekennzeichnet war. Zudem konnte nach der Vorbehandlung eine signifikant erhöhte TRIM30-Expression in PECs und in den myeloiden Zellen aus der Milz nachgewiesen werden. Um eine eindeutige Wirkung des LTβR-induzierten TRIM30 in der Gegenregulation der Entzündung nachzuweisen, wurden in vitro knock-down-Experimente mittels TRIM30-spezfischer siRNA durchgeführt. Durch eine gezielte Herunterregulation von TRIM30 auf mRNA- und Protein-Ebene konnte die LTβR-vermittelte Reduktion der Zytokinexpression nach TLR-Restimulierung signifikant gehemmt werden.
Zur näheren Charakterisierung der LTβR-tragenden Zellen, die für eine Hemmung der Entzündungsreaktion verantwortlich sind, wurden LTβR(flox/flox) x LysMcre-Mäuse in unserem Labor hergestellt. Diese Mäuse zeigen eine zellspezifische Deletion des LTβR auf Makrophagen/Neutrophilen. Die aus dem Knochenmark der LTβR(flox/flox) x LysMcre-Mäuse generierten BMDM waren nicht in der Lage nach Stimulierung mit agonistischem anti-Maus LTβR mAK TRIM30 zu induzieren. Zudem konnte die TLR-induzierte Zytokin- und NO-Freisetzung dieser Zellen durch eine LTβR-Vorstimulierung nicht reduziert werden. In vivo wurde ebenfalls keine Hemmung der TLR-vermittelten Entzündungsreaktion beobachtet. Mit den erhaltenen Ergebnissen konnte erstmalig gezeigt werden, dass die Aktivierung des LTβR zur Induktion eines TRIM30-abhängigen Signalwegs in Makrophagen/Neutrophilen führt, der für die Gegenregulation einer Entzündungsreaktion entscheidend ist.
In der vorliegenden Arbeit wurde außerdem der LTβR-abhängige Signalweg, der zur TRIM30-Induktion führt, näher charakterisiert. Es konnte zunächst gezeigt werden, dass die Aktivierung des LTβR die Induktion von TRIM30 direkt vermittelt und nicht indirekt von MyD88 oder TNF abhängig ist. Zudem wurde untersucht, welcher der beiden NFκB-Signalwege in den Makrophagen nach der Stimulierung des LTβR bei der Induktion von TRIM30 aktiviert wird. Die Ergebnisse zeigen, dass nach LTβR-Stimulierung sowohl der klassische als auch der alternative NFκB-Signalweg in den Makrophagen aktiviert wird. Nach retroviraler Transduktion der J774-Zellen mit spezifischen NFκB-Inhibitoren, die den klassischen NFκB-Weg (IκBα Superrepressor) oder den alternativen NFκB-Weg (dominant-negative Form von NIK oder NIK DN) hemmen, wurde die TRIM30-Expression in Abhängigkeit der beiden Signalwege untersucht. Es zeigte sich, dass die LTβR-vermittelte Induktion von TRIM30 von der Aktivierung des klassischen NFκB-Signalwegs abhängig zu sein scheint. Zur Identifizierung der beteiligten TRAF-Proteine wurden die J774-Zellen mit dominant-negativen Formen von TRAF2, 3, 5 und 6 transduziert. Es konnte gezeigt werden, dass die LTβR-induzierte TRIM30-Expression unabhängig von TRAF2, 5 und 6, aber in Abhängigkeit von TRAF3 vermittelt wird.
Zusammenfassend weisen die in dieser Arbeit erhaltenen Daten darauf hin, dass über das LTβR/Ligand-System ein neuer regulatorischer Mechanismus vermittelt wird, um frühe Entzündungsreaktionen zu kontrollieren. Die Aktivierung des LTβR auf Zellen des angeborenen Immunsystems durch LTα1β2/LIGHT, exprimiert auf Zellen des adaptiven Immunsystems, führt zur Induktion eines gegenregulierenden TRIM30-abhängigen Signalwegs, der für einen Schutz vor überschießenden Entzündungsreaktionen entscheidend ist.

The Lymphotoxin-β-receptor (LTβR), a member of the TNF receptor superfamily, interacts with the heterotrimeric ligand LTα1β2 or the homotrimeric ligand LIGHT, both expressed on activated T, B and NK cells.
So far most studies have focused on the critical role of the LTβR in the development of secondary lymphoid organs and the maintenance of lymphoid structures. Up to now very little is known about the physiological consequences of LTβR activation in inflammatory processes. However, our previous studies indicated that activation of the LTβR by LTα1β2 expressed by T cells seems to control and down-regulate intestinal inflammation in the experimental model of acute DSS-induced colitis. These results were confirmed using bone marrow chimeras demonstrating that activation of the LTβR on haematopoietic cells plays a crucial role in dampening DSS-induced intestinal inflammation. Therefore, we assume in our working hypothesis that activation of the LTβR expressed on cells of haematopoietic origin plays a distinct role in the resolution of inflammation.
While characterizing the cellular and molecular mechanisms responsible for the reduced inflammatory reaction after LTβR stimulation, we were able to identify the induction of TRIM30 as a result of LTβR signalling in macrophages for the first time. So far, TRIM30 has been reported as a negative regulator of TLR-mediated NFκB activation inducing a negative feedback mechanism to control prolonged and excessive inflammatory reactions. Using bone marrow derived macrophages (BMDM) and mouse macrophage cell line J774, we demonstrated that LTβR activation induces TRIM30 expression on mRNA and protein level. Furthermore, co-culturing of activated T cells, which expressed the ligands of the LTβR on the cell surface, results in induction of TRIM30 expression in macrophages.
Since TRIM30 is involved in LPS-induced tolerance in vitro and in vivo, we tested the impact of LTβR activation on the TLR-induced expression of pro-inflammatory cytokines. We could demonstrate that LTβR pre-stimulation on primary BMDM or on the J774 cell line results in a significantly reduced TLR-mediated induction of cytokine and NO production in vitro. These results were confirmed in vivo. Therefore, the intraperitoneally application of agonistic anti-LTβR mAb leads to a reduction of TLR-mediated inflammatory reaction characterized by a significantly decreased TNF concentration in the serum. Moreover, the TRIM30 expression is significantly enhanced in peritoneal exudate cells (PEC) and CD11b+ splenocytes after pre-treatment of mice. To confirm that LTβR-induced TRIM30 expression is responsible for counter regulation of inflammation, specific knock-down experiments using TRIM30-specific siRNA were performed. The knock-down of LTβR-dependent induction of TRIM30 on mRNA and protein level abolished the LTβR-mediated cytokine tolerance upon TLR restimulation.
In order to identify and characterize the LTβR-expressing cell population responsible for enhanced inflammatory reaction, we generated mice with specific deletion of LTβR expression on macrophages and neutrophils. Ablation of LTβR signalling in BMDM resulted in an abolished TRIM30 induction after stimulation with agonistic anti-LTβR mAb. Moreover, after pre-stimulation of LTβR reduced TLR-induced cytokine and NO production in vitro was not detected anymore.
Furthermore, LTβR(flox/flox) x LysMcre mice were characterized by an exacerbated acute inflammatory reaction in the model of TLR-mediated cytokine induction. Our data suggest that LTβR activation on macrophages/neutrophils results in the activation of a counter-regulating TRIM30-dependent signalling pathway, indispensable for protection against exacerbating inflammatory reactions.
On a molecular level the signalling pathway leading to LTβR-mediated TRIM30 induction was characterized. We demonstrated that the LTβR-induced TRIM30 expression is independent of MyD88 or TNF. Furthermore, we analyzed which NFκB signalling pathway is responsible for the TRIM30 expression. The results indicate that both the canonical as well as the non-canonical pathway is activated in response to LTβR stimulation of macrophages. Further, J774 cells, retrovirally transduced with specific inhibitors of NFκB proteins, which block either the canonical pathway (IκB alpha superrepressor) or the non-canonical pathway (dominant-negative mutant of NIK or NIK DN), were analyzed for TRIM30 expression. Hence, we showed that LTβR-induced TRIM30 expression seems to be mediated by the canonical NFκB signalling pathway. To investigate the role of TNFR-associated factor proteins (TRAF) in LTβR-induced TRIM30 expression, J774 cells were retrovirally transduced with dominant-negative mutants of TRAF2, 3, 5 or 6. Our findings indicate a crucial role for TRAF3 in LTβR-mediated TRIM30 induction.
These results support the conclusion that LTβR activation on cells of the innate immune system by LTα1β2/LIGHT expressed on cells of the adaptive immune system results in TRIM30 induction, which negatively regulates NFκB-dependent pathways. Therefore, LTβR activation dampens and controls an acute inflammatory reaction.
