Abstract (German)
Es ist bekannt, dass Bone Morphogenetic Proteins (BMPs) an der Regulation der Pro¬gression des malignen Melanoms beteiligt sind. So konnte bereits nachgewiesen werden, dass die verstärkte Expression von BMP4 in Melanomzellen im Vergleich zu Melanozyten unter anderem die Migration und Invasion der Melanomzellen fördert. Im Rahmen der vorliegenden Arbeit wurden die molekularen Mechanismen, welche in einer Induktion der BMP4 Expression im Melanom resultieren, untersucht. Es zeigte sich, dass der Verlust der Expression einer microRNA eine Signalkaskade induziert, welche über verschiedene Zwischenschritte die BMP4 Expression und die Migrationsfähigkeit der Melanomzellen fördert. Die Expression der miR-196a ist in Melanomzellen im Vergleich zu normalen Melanozyten sehr stark vermindert, was zu einer verstärkten Expression von HOX-B7 führt. Mit Hilfe von Mutationsanalysen der miR-196a Bindestelle in der 3’UTR von HOX-B7 konnte die direkte Bindung der miR-196a an HOX-B7 nachgewiesen werden. Weiterhin zeigte sich, dass das Ausschalten der HOX-B7 Expression mit Hilfe von spezifischen siRNAs in einer verminderten Expression des Wachstumsfaktors bFGF resultiert. bFGF wiederum aktiviert den Transkriptionsfaktor Ets1, welcher schließlich die BMP4 Expression induziert. Zusätzlich konnte nachgewiesen werden, dass die Induktion der Migration im malignen Melanom über die Aktivierung der BMP4-Expression vermittelt wird. Im zweiten Teil der Arbeit wurde ein bisher unbekanntes Zielgen des BMP-Signalweges im malignen Melanom identifiziert und funktionell charakerisiert. Mit Hilfe eines Microarrays wurden in einem ersten Screening diejenigen Gene identifiziert, welche nach Behandlung mit dem Smad-spezifischen BMP-Inhibitor Dorsomorphin im Vergleich zu den kontrollbehandelten Melanomzellen vermindert exprimiert wurden. Zur Verifizierung der Microarray-Daten wurde die Expressionsstärke der potentiellen BMP-Zielgene mittels qRT-PCR analysiert. Dabei zeigte sich, dass Dido1 (Death inducer obliterator 1) die stärkste Regulation aufwies. Weiterhin konnte dargestellt werden, dass Dido1 sowohl in Melanomzellen als auch in Melanomgeweben verstärkt exprimiert wird. Funktionelle Analysen zeigen, dass Dido1 einerseits sowohl das Attachment, die Migration, die Invasion sowie das matrix-unabhängige Wachstum der Melanomzellen fördert, andererseits darüber hinaus eine anti-apoptotische Wirkung auf die Melanomzellen ausübt. In der vorliegenden Arbeit konnten somit zum einen die regulatorischen Prozesse, welche zur Induktion der BMP4 Expression im malignen Melanom führen, aufgeklärt werden. Zum anderen konnte anhand von funktionellen Untersuchungen gezeigt werden, dass Dido1 als Smad-abhängiges BMP-Zielgen einen entscheidenden Beitrag zur Entstehung und Progression des malignen Melanoms leistet.

It is known that Bone Morphogenetic Proteins (BMPs) play an important role in the progression of malignant melanoma. Previously, it has been shown that BMP4 is overexpressed in melanoma cells compared to melanocytes and induces migratory and invasive potential of melanoma cells. In the first part of this work, the molecular mechanisms leading to overexpression of BMP4 in melanoma were determined. It was shown that loss of expression of a microRNA activates a signalling cascade, which finally enhances the expression of BMP4 and therefore the migratory potential of melanoma cells. Expression of the microRNA miR-196a is strongly reduced in melanoma cells compared to normal melanocytes, resulting in induced expression of the transcription factor HOX-B7. Direct binding of miR-196a to the 3’UTR of the HOX-B7 transcript was confirmed by mutagenesis analysis. Furthermore, knockdown of HOX-B7 expression using specific siRNAs led to diminished expression of the growth factor bFGF. bFGF in turn activates the transcription factor Ets-1, which subsequently induces expression of BMP4. In addition, it could be demonstrated that induction of migratory potential of melanoma cells is mediated via activation of BMP4 expression. In the second part of this study, a hitherto unknown BMP target gene was identified and functionally characterised. Treatment of melanoma cells with the Smad-specific inhibitor Dorsomorphin resulted in diminished expression of several potential BMP target genes as determined by microarray analysis. Subsequently, Death Inducer Obliterator 1 (Dido1) was identified as a BMP-specific Smad-regulated target gene, which could be confirmed by qRT-PCR and immunofluorescence staining. Further, expression of Dido1 is upregulated in melanoma cell lines and tissue samples compared to healthy melanocytes. Functional analysis revealed that Dido1 promotes attachment, migration, invasion and colony formation of melanoma cells. Additionally, Dido1 contributes to apoptosis resistance of melanoma cells. Within the scope of this doctoral thesis, the signaling cascade leading to upregulation of BMP4 expression in malignant melanoma was elucidated. In addition, functional studies revealed that the Smad-dependent BMP target gene Dido1 strongly contributes to formation and progression of malignant melanoma.
