Abstract (German)
Die X-gebundene juvenile Retinoschisis (XRS) ist eine Netzhauterkrankung die hauptsächlich Männer betrifft (George et al. 1995). Genetische Ursache der XRS sind Mutationen im Retinoschisin (RS1) Gen, welches für ein 24 kDa Protein kodiert (Sauer et al. 1997). Das Protein des Retinoschisins (RS1) wird als homo-oktamerischer Komplex sezerniert und ist extrazellulär an den Bipolarzellen und Photorezeptoren der Netzhaut lokalisiert.
Ein wesentlicher Teil des RS1 Proteins wird von der konservierten Discoidin Domäne gebildet, welche verantwortlich für eine Bindung an Komponenten der extrazellulären Matrix (EZM) ist (Alexander et al. 1992). Die konkreten Bindungspartner des RS1 sind noch nicht vollständig geklärt, ebenso wie die eigentliche zelluläre bzw. extrazelluläre Funktion des Proteins. Aufgrund bekannter Funktionen des Discoidin Moduls bei anderen Proteinen wird für das RS1 eine Funktion als Gerüstprotein diskutiert (Molday et al. 2001; Wu et al. 2005).
Speziell die Bindung des RS1 Proteins an Zellmembranen wurde durch eine eigens im Laufe dieser Arbeit entwickelte Methode näher untersucht. Hierbei wird rekombinantes RS1 Protein mit homogenisiertem Mausgewebe inkubiert, anschließend wird der lösliche Überstand abgetrennt und mittels Western Blot analysiert. Die Methode wurde des Weiteren modifiziert durch den Einsatz von mutanten RS1 Proteinen und der Zugabe von Peptiden und Monosacchariden. Die Bindung zu potenziellen Bindungspartnern wie Phospholipiden und Kollagenen wurde mittels ELISA untersucht. Die Bindung des RS1 wurde auch zu den alpha 3/beta 2 Untereinheiten der Na/K ATPase mittels Immunopräzipitation untersucht, sowie deren strukturelle Voraussetzungen für diese Bindung.
Die Ergebnisse dieser Arbeit zeigen, dass RS1 unabhängig von seiner oligomeren Form nur an die Membranen der Netzhaut bindet. Die Oktamerstruktur des RS1 Proteins scheint nicht für die Bindung an die Netzhaut notwendig zu sein und erfüllt wahrscheinlich eine andere, bisher nicht bekannte, Funktion. Drei früher charakterisierte Polypeptidschlaufen (Spike 1 bis Spike 3) der Discoidin Domäne sind vermutlich an der Bindung des RS1 Proteins an die Netzhaut beteiligt, da Blockierungsexperimente mit synthetischen Peptiden die Bindung behindern. Die Bindung wird allerdings nicht ausschließlich über die Spikes vermittelt, da Galactose die Bindung ebenfalls blockiert, obwohl es bekanntermaßen nicht direkt an die Spikes der Discoidin Domäne bindet. Eindeutig als Bindungspartner ausgeschlossen wurden die Phospholipide und die Kollagene I - V. Co-Präzipitationsexperimente des RS1 Proteins mit den Untereinheiten der Na/K ATPase, insbesondere mit der extrazellulären Domäne der beta 2-Untereinheit, bestätigen die Ergebnisse einer kanadischen Arbeitsgruppe (Molday et al. 2007) und zeigen dass tatsächlich eine direkte Interaktion zwischen Retinoschisin und der Na/K ATPase existiert (Molday et al. 2007). Das Bindungsverhältnis zwischen RS1 und der Na/K ATPase kann aufgrund der vorhandenen Daten auf etwa 1:8 geschätzt werden. In diesem Zusammenhang ist eine Funktion des RS1 als Gerüstprotein mit möglichen Aufgaben in der Positionierung und Interaktion mit der Na/K ATPasen zu diskutieren.
Der zweite Schwerpunkt dieser Arbeit lag auf der altersabhängige Makuladegeneration (AMD). Die AMD ist in den westlichen Industrienationen eine der häufigsten Ursachen von Erblindungen im Alter. Ihre Entwicklung wird von Umwelt- aber auch genetischen Faktoren beeinflusst und gilt daher als eine komplexe Erkrankung. Vor kurzem wurde durch Kopplungsstudien ein Genort auf Chromosom 10q26 näher charakterisiert, der signifikant mit der AMD assoziiert ist (Fisher et al. 2005; Rivera et al. 2005). Dieser Genort enthält das ARMS2 (age-related maculopathy susceptibility 2). Besonders auffällig war eine ARMS2 Variante, die eine mRNA destabilisierende Insertion/Deletion enthielt, was einem funktionellen Polymorphismus entsprechen könnte (Fritsche et al. 2008).
Daher wurde im Rahmen dieser Arbeit ein polyklonaler anti ARMS2 Antikörper hergestellt und dessen Epitop feinkartiert. Die endogene Expression des ARMS2 Proteins wurde mittels dieses Antikörpers in humanen Geweben ermittelt. Diese Ergebnisse zeigen zum einen, dass ARMS2 ein proteinkodierendes Gen ist. Des Weiteren wurde die Expression der vermuteten funktionellen ARMS2 Varianten in humanem Plazentagewebe bestimmt. Dabei wurde festgestellt, dass die Insertion/Deletion ARMS2 Variante nicht exprimiert wird. Dies scheint die Rolle der risikoassoziierten Variante des ARMS2 im Krankheitsprozess der AMD zu unterstützen.

Genetic mutations are the cause of the XRS in Retinoschisin (RS1) gene, which encodes a 24 kDa protein (Sauer et al. 1997). The protein of the Retinoschisins (RS1) is secreted as homo-oktamerischer complex and is localized extracellularly in the bipolar cells and photoreceptors in the retina.
An essential part of the RS1 protein is formed by the conserved discoidin domain, which is responsible for binding to components of the extracellular matrix (ECM) (Alexander et al. 1992). The specific binding partner of the RS1 are not yet fully understood, as well as the actual cellular or extracellular function of the protein. Based on known functions of the discoidin module with other proteins is a function of the RS1 discussed as a scaffold protein (Molday et al, 2001;.. Wu et al 2005).
Specifically, the binding of RS1 protein on cell membranes was examined by a specially developed method in this work. This recombinant RS1 protein is incubated with homogenized mouse tissue, and then the soluble supernatant was separated and analyzed by Western blot. The method was further modified by the use of mutant RS1 proteins and peptides, and the addition of monosaccharides. The binding of potential binding partners, such as phospholipids and collagen was examined by ELISA. The binding of RS1 was also studied at the alpha 3 / beta 2 subunit of the Na / K ATPase by immunoprecipitation, and their structural requirements for this binding.
The results of this study show that RS1 binds regardless of its oligomeric form only in the membranes of the retina. The octameric structurer of the RS1 protein does not appear to be necessary for binding to the retina and probably fulfills a different, hitherto unknown, function. Three previously characterized peptide spikes (spike 1 to spike 3) of the discoidin domain are probably involved in the binding of RS1 protein in the retina, since blocking experiments with synthetic peptides inhibit the binding. However, the binding is not mediated exclusively through the spikes, because the galactose binding also blocked, although it is not known directly to the studs of the discoidin domain binds. Clearly ruled out as binding partners are the phospholipids and the collagens I - V. Co-precipitation experiments of RS1 protein with the subunits of the Na / K ATPase, in particular with the extracellular domain of the beta 2-subunit, confirming the results of a Canadian research group (Molday et al . 2007) and show that indeed there is a direct interaction between Retinoschisin and the Na / K ATPase (Molday et al. 2007). The binding relationship between RS1 and the Na / K ATPase can be estimated from the available data to about 1:8. In this context, a function of the RS1 is to discuss as a scaffold protein with potential roles in the positioning and interaction with the Na / K ATPases.
The second focus of this work was on the age-related macular degeneration (AMD). AMD is in the western industrial nations, a leading cause of blindness in old age. Their development is influenced by environmental factors but also genetic and is therefore considered a complex disease. Recently, linkage analysis was carried one gene locus on chromosome 10q26 characterized in detail, which is significantly associated with AMD (Fisher et al, 2005;.. Rivera et al 2005). This locus contains the ARMS2 (age-related maculopathy susceptibility 2). Particularly striking ARMS2 one variant, which contained an mRNA destabilizing insertion / deletion, which could correspond to a functional polymorphism (Fritsche et al. 2008) was.
Therefore, in this study, a polyclonal alpha ARMS2 antibody was produced and mapped its epitope. The endogenous expression of ARMS2 protein was determined by this antibody in human tissues. These results show firstly that ARMS2 is a protein-coding gene. Furthermore, the expression of the presumed functional ARMS2 variants was determined in human placental tissue. It was found that the insertion / deletion variant ARMS2 is not expressed. This is the role of risk-associated variant of the disease process ARMS2 seems to support the AMD.
