Schäden und Verlust des Gelenkknorpels stellen eine immense Belastung für den individuellen Patienten und für die Volkswirtschaft dar. Das moderne Tissue Enginee-ring verspricht eine zukunftsweisende Behandlung für fokale, meist traumatisch entstandene Knorpeldefekte zu werden. Obwohl das Hormonmilieu sowohl bei Spen-derzellentnahme als auch bei Transplantation des TE-Konstruktes determiniert ist, sind Studien zur Beeinflussung der Chondrogeneseleistung rar und der zugrundeliegende intrazelluläre Signalweg noch unbekannt. Ziel dieser Arbeit war es, Kurzzeitwirkungen von 17β-Estradiol auf die Chondrogenese humaner mesenchymaler Stammzellen zu untersuchen und den MAPK pathway als möglichen Signalweg zu analysieren.
Die Syntheseleistung konnte durch eine nur eintägige Kultivierung mit 17β-Estradiol beeinflusst werden. Eine tendenzielle Konzentrationsabhängigkeit wurde beobachtet, jedoch ohne statistische Signifikanz. In der neu etablierten Western Blot-Analyse konnte die Aktivierung der MAPK (p38, ERK) nachgewiesen werden. Diese war einwirk-zeit- und konzentrationsabhängig. Ein alleiniger Effekt durch membranständige Rezeptoren konnte nicht bewiesen werden.
Die vorliegende Arbeit belegt die Hormonsensitivität des verwendeten Modells auch für kurze Einwirkzeiten. Die Aktivierung der MAPK ist ein erster Schritt zur Entschlüsse-lung des Signalweggeflechts.
Die Ergebnisse weisen auf das Potenzial der Sexualhormone beim modernen Tissue Engineering hin und rücken den Hormonstatus zukünftiger Patienten in den Fokus.

Translation of the abstract (English)

Articular cartilage damage causes pain and limited mobility for patients and contributes to high economic costs. Over the last years, cell-based tissue engineering has been considered an effective regenerative approach regarding the repair of chondral defects.
Objective of this thesis was to examine short term effects of 17β-estradiol on chondrogenesis of bone marrow derived human MSCs and to study the mitogen activated protein kinases (MAPK) pathway as a possible signalling mechanism - with consideration of sex-dependent differences in cartilage repair.
After only one day of incubation with 17β-estradiol, an influence on chondrogenesis of human MSCs appeared. This effect turned out to be dose dependent, however, without statistical significance. The newly established western blot analysis showed an increase of activity of the MAPK (ERK, p38) dependent on E2 concentration and incubation period. An exclusive effect mediated by membrane-associated receptors was not proven.
This thesis confirms hormone sensitivity of the 3-dimensional aggregate culture even for short incubation periods. Activation of the MAPK is the first step to handle the complexity of the E2 signalling mechanisms.
These findings emphasize the potential of sex hormones regarding guided tissue engineering and set the focus on the hormonal status of future patients.
