Abstract (German)

In der vorliegenden Arbeit konnte zum ersten Mal der ontogenetische Zelltod in der Retina bei Mäusen auch an älteren Tieren untersucht werden, da bei einem vollständigen Knock-out des TßRII die Tiere bereits intrauterin bzw. kurz nach Geburt an diversen Miß- oder Fehlbildungen versterben. Dies erfolgte durch den konditionellen Knock-out des TßRII, von dem nur bestimmte Zellen der Retina betroffen sind. Zusätzlich wurde eine verstärkte Aktivierung des TßRII Signalwegs durch einen konditionellen Knock-out von Smad7 und seine Auswirkung auf den ontogenetischen Zelltod in der Retina untersucht.
Nach Bestätigung des konditionellen Knock-out für den TßRII durch immunhistochemische Färbungen, Western Blot Analysen und in TUNEL-Analysen wurde gezeigt, dass es durch das Fehlen des TGF-ß Signalwegs zu einer gesteigerten Apoptose im Bereich der inneren Körnerschicht (INL) an d 4, d 7, d 10 kam. Diese TUNEL-Daten wurden durch die Detektion freier Histone (ELISA) an d 10 nochmals validiert. Auch wenn kein offensichtlicher Phänotyp bei α-Cre; TGF-ß-R2fl/fl Tieren im Vergleich zu TGF-ß-R2+/+ Tieren in Erscheinung trat, so war die innere Körnerschicht (INL) an d 15 nach Geburt bei α-Cre; TGF-ß-R2fl/fl Tieren tendenziell dünner und erreichte an manchen Stellen signifikantes Niveau. Im Bereich der Außensegmente der Photorezeptoren zeigte sich bei α-Cre; TGF-ß-R2fl/fl an d 15 nach Geburt auch eine Tendenz zu dünneren Außensegmenten, allerdings wurde nur an einer Stelle ein signifikantes Niveau erreicht, was auch zu erwarten war, da die Photorezeptoren kaum am ontogenetischen Zelltod teilnehmen.
Der konditionelle Knock-out von Smad7 konnte in Western Blot Analysen und quantitativer real-time Analysen bestätigt werden. Der Phänotyp von α-Cre; Smad7fl/fl Tieren wurde mit dem von Smad7+/+ an d 15 nach Geburt miteinander verglichen, wobei keine Unterschiede festgestellt wurden. Im Bereich der inneren Körnerschicht (INL) kam es zu keiner Hyperzellularität bei α-Cre; Smad7fl/fl Tieren. In TUNEL-Analysen an d 7 und d 10 konnte gezeigt werden, dass es bei einem konditionellen Knock-out von Smad7 zu einer signifikant verminderten Apoptose im Bereich der inneren Körner-schicht (INL) kommt.
Entgegen der bisherigen Meinung zum ontogenetischen Zelltod in der Retina wurde herausgearbeitet, dass TGF-ß einen protektiven Effekt auf die Neurone während des ontogenetischen Zelltodes in der Retina darstellt.

The ontogenetic cell death could be investigated first in the retina of older mice because of the intrauterine deathness of the complete, systemic knock-out of the TßRII. This was possible through the conditional knock-out of the TßRII which influences only particular cells in the murine retina. Additionally we designed mice with a conditional knock-out of Smad7 (with a stronger activation of the TGF-ß pathway) and investigated the influence of the Smad7 on the ontogenetic cell death. We confirmed the conditional knock-out of the TßRII with immunhistochemistry and Western Blot. TUNEL staining showed an increasing number of apoptotic cells on d 4, d 7 and d 10 predominantly in the inner nuclear layer (INL) through an inactivation of the TGF-ß pathway. A validation of the TUNEL data happened through the detection of free nucleosome (ELISA) on d 10. There was no obvious phenotype in α-Cre; TGF-ß-R2fl/fl mice compared to TGF-ß-R2+/+ mice, although the inner nuclear layer (INL) was significantly thinner on some places on d15. The outer segments of the photoreceptors of α-Cre; TGF-ß-R2fl/fl mice were only on one place significantly thinner than these of TGF-ß-R2+/+ mice because the photoreceptors succumb rarely on the ontogenetic cell death.
We proofed the conditional knock-out of Smad7 in Western Blot and quantitative real time analysis. The phenotype of α-Cre; Smad7fl/fl mice was compared to the phenotype of Smad7+/+ mice on d 15 without an obvious difference. There was no hypercellularity in the inner nuclear layer (INL) of α-Cre; Smad7fl/fl mice. TUNEL staining on d 7 and d 10 showed a significantly decreasing number of apoptotic cells in α-Cre; Smad7fl/fl mice compared to Smad7+/+ mice predominantly in the inner nuclear layer (INL).
Contrary to former papers we showed that TGF-ß have a protective function to neurons in the retina during the ontogenetic cell death.
