Haspine sind Serin/Threonin-Kinasen, die aufgrund ihrer abweichenden Struktur eine distinkte Subgruppe von eukaryotischen Proteinkinasen bilden. Der Name (haploid germ cell specific nuclear protein kinase) reflektiert ihre Entdeckung in testikularen Maus-Gameten, wobei inzwischen bekannt ist, dass sie ubiquitär in proliferierenden Zellen aller bislang untersuchten Organismen exprimiert werden. In Säugern, Pflanzen, S. pombe und X. laevis phosphorylieren Haspine das Histon H3 an der Position Threonin 3. Über diese Modifikation unterstützt Haspin u.a. die Chromosomen-Kohäsion und die Positionierung des CPCs (chromosomal passenger complex) am inneren Zentromer. In S. cerevisiae gibt es – anders als in Säugern - statt einem, zwei Haspin-Homologe, Alk1 und Alk2, deren Funktion bislang weitestgehend unverstanden ist. In der vorliegenden Arbeit konnte gezeigt werden, dass in der Bäckerhefe die Haspin-Proteine das spezifisch auf die Tochterzelle gerichtete Wachstum in der Mitose beenden. Im Vergleich mit anderen morphogenetischen Ereignissen ist dieser Schritt molekular unverstanden.
Hinweise für eine solche Funktion lieferten die Analysen der Haspin-Doppelmutante und die Überexpressionsstudien von ALK2. Die Abwesenheit von ALK1 und ALK2 führte dazu, dass die Zellen nicht mehr in der Lage waren, die Asymmetrie der Mutter-Tochterzelle aufzuheben. Durch die Visualisierung einer Serie von Komponenten des polarisierten Wachstums und des Aktin-Zytoskeletts zeigte sich, dass die Polarisationsmaschinerie am Kortex nicht aufgelöst werden konnte. Folglich bildeten sich große, elongierte Tochterzellen. Darüber hinaus wirkten erhöhte Alk2-Mengen inhibitorisch auf die Assemblierung von Proteinen der Polaritätsmaschinerie und auf die Polarisation des Aktin-Zytoskeletts. Dadurch wurde spezifisch die Ausbildung einer Knospe unterbunden, während die Spindelpol-Duplikation und die DNA-Replikation in der Zellprogression fortschritten.
Weiterhin unterstützten mikroskopische Studien der räumlichen und zeitlichen Lokalisation von Alk1 und Alk2 eine Aufgabe bei der Beendigung des spezifisch auf die Tochterzelle gerichteten Zellwachstums. Dabei zeigte sich zum Einen, dass beide Homologen Zellzyklus-abhängig reguliert werden und in der G1-Phase, in der die Etablierung der Zellpolarität beginnt, nicht vorhanden sind. Zum Anderen wurde beobachtet, dass Alk1 inhomogen im Zytoplasma und am mütterlichen Kortex lokalisiert, Alk2 hingegen im Kern, an der nukleären Spindel und am Knospenhals. Nachdem der Kern den Knospenhals passierte, war Alk2 und auch Alk1, im Metaphase-arretierten Zustand, verstärkt am Knospenkortex zu detektieren. Durch die Untersuchung verschiedener N- und C-terminaler Verkürzungen von Alk2 konnte gezeigt werden, dass der N-terminale Bereich hinreichende Regionen für die nukleäre, kortikale als auch für die Knospenhals-Lokalisation beinhaltet.
Zudem ergab sich, dass die asymmetrisch lokalisierende Kinase Kin4 die kortikale und nukleäre Lokalisation von Alk2 steuert. Auf diese Weise wird Alk2 zum richtigen Zeitpunkt zur Beendigung des polaren Wachstums an den Kortex der Tochterzelle gelenkt. Erhöhte Kin4-Mengen führten zu einem Verlust der kortikalen Alk2-Lokalisation. Entsprechend bewirkte die Abwesenheit des Mutterzell-spezifischen Kin4s eine Zunahme des Alk2-Signals am mütterlichen Kortex. Zusätzlich nahm in kin4Δ-Mutanten das nukleäre Signal von Alk2(1-676) und des N-terminalen Subfragments Alk2(1-333) ab, während das der Verkürzungen Alk2(291-330) und Alk2(112-333) unverändert in seiner Intensität war. Aus diesem Grund scheint der Einfluss von Kin4 auf die Kernlokalisation von Alk2 indirekt über den Bereich der Aminosäuren 1 bis 112 vermittelt zu werden.
Des Weiteren zeigte sich, dass Haspin die Lokalisation von Bem2 steuert. In der alk1Δ alk2Δ-Mutante war das am Zellwachstum beteiligte Protein nicht mehr am Kortex zu detektieren, während es nach ALK2-Überexpression dort verstärkt lokalisierte. Möglicherweise stellt Bem2 das potentielle Zielprotein von den Haspin-Homologen zur Beendigung des polaren Wachstums dar.
Genetische Interaktionen der Haspin-Doppelmutante mit dem Gen des Kinesin-verwandten Motorproteins Cin8 wiesen auf eine Verbindung zum Mikrotubuli-Zytoskelett hin. Bei der Visualisierung der Spindel der Haspin-Doppelmutanten zeigte sich, dass die Abwesenheit beider Proteine die Orientierung der Spindel stört, was in falsch ausgerichteten Mikrotubuli-Strukturen resultierte. Es konnte gezeigt werden, dass Alk1 und Alk2 keine Bestandteile bekannter Spindelorientierungswege sind, jedoch mit diesen in Verbindung stehen. Zudem wurde eine Rolle von Haspin in der Mikrotubuli-Dynamik immer wahrscheinlicher, da genetische Interaktionen von alk1Δ alk2Δ mit Mutanten, die eine veränderte Mikrotubuli-Dynamik aufwiesen, beobachtet wurden.
Weiterhin wurde deutlich, dass Haspin in S. cerevisiae – anders als in Säugerzellen - seine Funktionen nicht über eine Histon H3 Threonin 3 Phosphorylierung ausübt. Zum Einen zeigte eine Histon-Mutante, bei der diese Stelle unmodifizierbar war, nicht die Phänotypen der Haspin-Doppelmutante. Zum Anderen wurde in der Bäckerhefe die in der Literatur beschriebene, über Haspin-vermittelte CPC-Lokalisation nicht festgestellt.
Zusammengenommen weisen diese Daten darauf hin, dass die Haspin-Homologen Alk1 und Alk2 in S. cerevisiae das asymmetrisch auf die Tochter gerichtete Wachstum in der Mitose beenden. Dabei stellt nicht, wie aus anderen Organismen bekannt, das Histon H3 Threonin 3 das relevante Substrat dar, sondern womöglich das Rho-GAP Bem2.

Haspins are serin/threonine kinases which form, due to their different structure, a distinct subgroup of eukaryotic protein kinases. The name haploid germ cell specific nuclear protein kinase (Haspin) reflects their discovery in testicular mice germ cells, however it is known, to date, that they are ubiquitaryly expressed in all probed organisms. In mammals, plants, S. pombe and X. laevis, Haspins phosphorylate histone H3 at position threonine 3. In addition to other functions Haspin supports the cohesion of chromosomes and the positioning of the CPC (chromosomal passenger complex) to the inner centromere using this modification. Unlike the situation in mammals, in S. cerevisiae, there are two instead of one Haspins, Alk1 and Alk2, whose function is almost unknown up to now. In this work it was shown that Haspins in budding yeast terminate the specific daughter cell directed growth in mitosis. Unlike other morphogenetic events this step has been hardly investigated on a molecular level.
Details for such a function were obtained through analysis of the Haspin-double mutant and the overexpression of ALK2. In the absence of ALK1 and ALK2 the cells were not able to break down the asymmetry between the mother-daughter cell. Through visualization of a series of polarity growth components and the actin cytoskeleton, this work describes that the polarity machinery at the cortex could not dissolve and consequently, large, elongated daughter cells emerged. In addition, an elevated amount of Alk2 appeared to inhibit the assembly of the polarity machinery and the actin cytoskeleton. Thereby, the formation of the bud was specifically prevented, while the duplication of the spindle pole body and the DNA-replication proceeded in the cell cycle.
Furthermore, microscopy studies of the spatial and temporal localization of Alk1 and Alk2 support their function in determing the polarized growth. On the one hand it could be shown that both Haspin proteins are cell-cycle dependent regulated and are absent in G1-phase, where the polarity establishment begins. On the other hand it could be shown that Alk1 is localized in the cytoplasm and the mother cortex, while Alk2 was present in the nucleus, at the nuclear spindle and at the budneck. After the nucleus passed the budneck, Alk2 and Alk1 were strongly detected at the bud cortex. Analysis of various Alk2 constructs, C-terminal as well as N-terminal truncated, could delimit the area of the nuclear, cortical and the budneck localization to the N-terminal region.
Moreover the asymmetric localizing kinase Kin4 directs the cortical and nuclear localization of Alk2 and navigates therefore Alk2 at the right time to the daughter cortex. An increased amount of Kin4 led to a loss of the cortical Alk2 localization. In addition, the absence of the mother specific Kin4 resulted in an increased cortical Alk2 signal. Furthermore, the nuclear signal of Alk2(1-676) and Alk2(1-333) was reduced in kin4Δ mutants, while the signal of Alk2(291-330) and Alk2(112-333) was unaltered. For this reason the influence of Kin4 on the nuclear localization of Alk2 seems to be mediated indirectly through the amino acids 1 – 112.
Moreover it was shown that Haspin regulates the localization of Bem2. In the alk1Δ alk2Δ mutant the polarity growth protein was no longer detectable at the cortex, while after the overexpression of ALK2, Bem2 localized there more intensely. It is possible that Bem2 is the potential substrate of Haspin for terminating the polarized growth.
Genetic interactions of the Haspin double mutant with the gene of the kinesin-related motorprotein Cin8 indicated a connection to the microtubule network. By visualization of the spindle in the Haspin double mutants, it could be shown that the simultaneously absence of Alk1 and Alk2 disturbed the spindle orientation, resulting in misorientated microtubule structures. This work discovered that Alk1 and Alk2 do not perform their function through one of the common spindle orientation pathways, however they are connected to them. In addition, a role of Haspins in microtubule dynamics became more likely, because genetic interactions of alk1Δ alk2Δ with mutants, which had altered microtubule dynamics, could be observed.
Moreover, unlike the situation in mammals, Haspins in S. cerevisiae do not perform their function through histone H3 threonine 3 phosphorylation. On the one hand, a histone mutant, which was not modifiable at this site, did not phenocopy the Haspin double mutant. On the other hand, the CPC-localization interceded by Haspin described in the literature, could not be determined in budding yeast.
Taken together, the results strengthen the opinion that the Haspin-homologues Alk1 and Alk2 in S. cerevisiae terminate the daughter cell directed growth by breaking down the mother-daughter cell asymmetry. Thereby, unlike in other organisms, the relevant target protein is not histone H3 threonine 3, but potentially Rho-GAP Bem2.
