Abstract (German)

Den Einfluss des TGF-β Signalwegs auf den Programmierten Zelltod in der Retina während der Embryonalentwicklung zu untersuchen, war Gegenstand vorliegender Arbeit. Durch konditionellen Knockout des TGF-β-Rezeptors2 wurden die Voraussetzungen dafür geschaffen. Den Einfluss des TGF-β Signalwegs auf den Programmierten Zelltod in der Retina während der Embryonalentwicklung zu untersuchen, war Gegenstand vorliegender Arbeit. Durch konditionellen Knockout des TGF-β-Rezeptors2 wurden die Voraussetzungen dafür geschaffen. In TUNEL-Analysen konnte unter Blockade des TGF-β Signalwegs eine signifikant höhere Apoptoserate am Embryonaltag E12.5, E14.5 und E16.5 in der Retina festgestellt werden, was folgern lässt, dass TGF-β antiapoptotische Wirkung auf retinale Neurone hat. Durch BrdU-Färbungen, Zellzählungen und Dickenmessungen der Retina ließ sich eine Veränderung der Proliferationsrate ausschließen und schlussfolgern, dass der fehlende offensichtliche Phänotyp durch sehr geringe nummerische Werte der apoptotischen Zellen zustande kommt. Lediglich eine Tendenz zu geringeren Retinadicken der transgenen Tiere konnte detektiert werden.
Um die Ergebnisse unter konditionellem Knockout des TGF-β-Rezeptors2 zu bestätigen, wurde eine zweite Mauslinie unter konditionellem Knockout von Smad 7 herangezogen. Auch in diesem Fall ergab die Phänotypanalyse der α-Cre;Smad7fl/fl Mäuse keine Unterschiede zu Smad7+/+ Mäusen. Die Untersuchungen mittels TUNEL-Analysen am Embryonaltag E16.5 ergaben eine signifikante Reduzierung der retinalen Apoptose unter Verstärkung des TGF-β Signalwegs durch fehlendes negatives Feedback von Smad 7 und bestätigten die Annahme, dass TGF-β während der Embryonalentwicklung antiapoptotisch auf die Neurone der Retina wirkt.

The aim of this dissertation was to investigate the influence of the TGF-β signaling pathway on programmed cell death in the mouse retina during embryonic development. The aim of this dissertation was to investigate the influence of the TGF-β signaling pathway on programmed cell death in the mouse retina during embryonic development. Therefore, TGF-β-R2fl/fl mice were crossed with α-Cre mice to generate a conditional inactivation of TGF-β-R2 in optic cup-derived cells of the neural retina. In consequence of TGF-β signaling pathway inhibition, TUNEL analyzes showed a significantly higher rate of apoptosis at embryonic day E12.5, E14.5 and E16.5 in the retina. This allows the conclusion that TGF-β has anti-apoptotic effects on retinal neurons during development. Based on BrdU staining, cell counting within the retina and measurements of the retinal thickness, a change of the proliferation rate could be excluded. Thus, the most likely explanation for the lack of a morphometric and morphologic phenotype might be that the number of cells undergoing apoptosis is too low compared to the total number of cells in the retina.
To further validate the results of the conditional knockout of TGF-β-R2, a second mouse line with a conditional inactivation of Smad7 was used. Phenotype analysis of α-Cre; Smad7fl/fl mice did not reveal any morphological differences when compared to Smad7+/+ mice. TUNEL analysis at embryonic day E16.5 resulted in a significant reduction of apoptotic cells in the retina. This protective effect is most likely due to the desinhibition of the TGF-β signaling pathway, caused by a lack of the inhibitory Smad7 protein. These results confirmed the hypothesis that TGF-β has an anti-apoptotic effect during embryonic development on the neurons of the mouse retina.
