Bei der rheumatoiden Arthritis (RA) handelt es sich um eine Multisystemerkrankung, deren komplexe Pathogenese noch immer nicht völlig verstanden ist. Immunsystem, Nerven- und endokrines System des Körpers spielen für den Verlauf der Erkrankung eine Rolle. In dieser Arbeit wurde der Einfluss des sympathischen Nervensystems auf den Interleukin-7 Rezeptor (IL-7Rα) auf B-Zellen untersucht. Da im Maus-Modell der Kollagen Typ-II-induzierten Arthritis eine Verschlechterung der Erkrankung nach Transfer von IL-7-stimulierten B-Zellen gezeigt werden konnte, bestand die Annahme, dass der IL-7R auf B-Zellen einen proinflammatorischen Einfluss im Kontext der Arthritis ausübt. Wurden die B-Zellen jedoch in Anwesenheit des Neurotransmitters Noradrenalin aktiviert, war der proinflammatorische Effekt nach Stimulation von B-Zellen mit IL-7 nicht mehr vorhanden. In der vorliegenden Arbeit wurde deshalb, neben dem Nachweis von IL-7Rα auf humanen und Maus B-Zellen, untersucht, weshalb eine Aktivierung der B-Zellen unter dem Einfluss von Noradrenalin die Wirkung der IL-7 Stimulation aufhebt. Die erste Hypothese war, dass durch die adrenerge Stimulation eine Veränderung der Rezeptor-Expression auf der Oberfläche von B-Zellen erfolgt. Es konnte jedoch gezeigt werden, dass zwar die Aktivierung von B-Zellen mit anti-CD40 und IL-4 eine Erhöhung der IL-7Rα Expression auf B-Zellen bewirkt, dass dies jedoch durch einen adrenergen Stimulus nicht beeinflusst wird. Als nächstes wurde die Hypothese untersucht, dass eine Aktivierung von B-Zellen in der Anwesenheit von Noradrenalin den Signalweg unterhalb des IL-7R beeinflusst. Hierzu wurde eine Methode zur intrazellulären Bestimmung von pSTAT5 und pERK mittels Durchflusszytometrie nach Stimulation von B-Zellen mit IL-7 etabliert. Es zeigte sich, dass die Stimulation mit β2-adrenergen Agonisten während der Aktivierung der B-Zellen mit anti-CD40/IL-4, die IL-7 induzierte Phosphorylierung von STAT5 verminderte, wohingegen die Phosphorylierung von ERK zunahm. Diese Ergebnisse deuten darauf hin, dass die intrazelluläre Bildung von second messengern unterhalb des IL-7R durch Stimulation der β2-AR auf B-Zellen verändert wird. Dies könnte auch eine Erklärung dafür bieten, dass IL-7Rα + B-Zellen, die in Anwesenheit des sympathischen Neurotransmitters Noradrenalin aktiviert werden, ihre proinflammatorischen Eigenschaften im Maus-Modell der Arthritis verlieren. Das tiefere Verständnis dieser Zusammenhänge kann eventuell für die Entwicklung neuer therapeutischer Prinzipien bei der RA verwendet werden.

Rheumatoid arthritis (RA) is a multi-systemic disease whose complex pathogenesis is still not fully understood. The immune system, the nervous and the endocrine system play a role in the process of the disease. In this study, the influence of the sympathetic nervous system on the interleukin-7 receptor (IL-7Rα) on B-cells was determined. Since it could be shown that in the collagen type II-induced arthritis (CIA) in mice there was an aggravation in the disease after transfer of IL-7 stimulated B-cells, it was expected that IL-7R on B-cells will mediate proinflammatory effects in the context of arthritis. When B-cells where activated in the presence of the neurotransmitter norepinephrine, the proinflammatory effect after stimulation of B-cells with IL-7 in CIA was lost. Therefore, we wanted to determine why activation of B-cells under the influence of norepinephrine counteracts the proinflammatory effect of IL-7 stimulation, alongside with the detection of IL-7Rα on human and mouse B-cells. The first hypothesis was, that adrenergic stimulation changes receptor-expression on the surface of B-cells. It could be shown that activation of B-cells with anti-CD40 and IL-4 causes an increase in IL-7Rα expression on B-cells, but this increase was not influenced by adrenergic stimuli. Therefore, we tested the hypothesis, that activation of B-cells in the presence of norepinephrine impacts the pathway downstream of IL-7Rα. For this purpose a method for intracellular detection of phosphorylated signal transducer and activator of transcription 5 (pSTAT5) and phosphorylated extracellular signal-regulated kinase (pERK) was established. The results showed that stimulation with β2-adrenergic agonists during activation of B-cells with anti-CD40 and IL-4 decrease the IL-7 induced phosphorylation of STAT5, whereas the phosphorylation of ERK was increased. These findings suggest that intracellular production of second messengers following the IL-7R is changed by stimulation of β2-AR on B-cells. This could also be an explanation for the fact that IL-7Rα positive B-cells, which are activated in the presence of norepinephrine, lose their proinflammatory effect in CIA. The deeper understanding of these correlations could eventually be used for the development of new therapeutic principles in RA.
