Abstract (German)

Wir konnten mit dieser Arbeit zeigen, dass das Flavonoid Luteolin ein potenter Modulator der Aktivierung, der Zellform und der Funktionen von Mikroglia ist. Sowohl in ruhender wie auch in LPS-aktivierter Mikroglia verursacht Luteolin globale Veränderungen des Transkriptoms, was zu einem M2-ähnlichen Phänotyp mit anti-inflammatorischen und neuroprotektiven Eigenschaften führt. Wir konnten mit dieser Arbeit zeigen, dass das Flavonoid Luteolin ein potenter Modulator der Aktivierung, der Zellform und der Funktionen von Mikroglia ist. Sowohl in ruhender wie auch in LPS-aktivierter Mikroglia verursacht Luteolin globale Veränderungen des Transkriptoms, was zu einem M2-ähnlichen Phänotyp mit anti-inflammatorischen und neuroprotektiven Eigenschaften führt. Dies geschieht mittels einer Modulation von mehreren, auch von NFkB unabhängigen Signalwegen. Damit bieten unsere Ergebnisse eine Grundlage für die Entwicklung von immun-modulatorischen und neuroprotektiven Therapien neurodegenerativer Erkrankungen.

Translation of the abstract (English)

We have shown that the flavonoid luteolin is a potent modulator of microglial activation, cell shape, and effector functions. Luteolin induced global changes in the transcriptome of resting or LPS-activated microglia leading to a polarized M2-like phenotype with anti-inflammatory and neuroprotective characteristics. We have shown that the flavonoid luteolin is a potent
modulator of microglial activation, cell shape, and effector
functions. Luteolin induced global changes in the
transcriptome of resting or LPS-activated microglia leading
to a polarized M2-like phenotype with anti-inflammatory
and neuroprotective characteristics. Luteolin’s
mechanisms of action appear to target several independent
pathways independent of NFkB. Our results provide
a basis to develop immuno-modulatory and
neuroprotective therapies for the treatment of neurodegenerative
disoders.
