Abstract (German)

In der vorliegenden Arbeit sollte das Abbauverhalten der verwendeten Keramiken in An- und Abwesenheit von BMP-7 untersucht werden, sowie die ektope Knochenneubildung.
Hierzu wurden in 10 Göttinger Minischweine ektop 3 verschiedene Keramiken (HA; HA60; TCP) implantiert. Nach 3-monatiger Einheilzeit, in deren Verlauf in-vivo CT’s angefertigt wurden sowie eine Fluoreszenzmarkierung durchgeführt wurde,wurden die Tiere getötet und die Keramiken explantiert. Die angefertigten Präparate wurden histomorphometrisch, fluoreszenzmikroskopisch und histologisch analysiert und ausgewertet.
Die HA und HA60-Keramiken zeigten ein osteoklastenabhängiges Abbauverhalten. Bei den TCP-Keramiken wurden ausschließlich Degradationsprozesse gefunden. Eine statistische Signifikanz fand sich im Vergleich von An- und Abwesenheit von BMP-7. Bei den mit
BMP-7 beimpften Keramiken war das Abbauverhalten höher (p<0,001).
Alle verwendeten Keramiken zeigten Knochenneubildung, wobei in Anwesenheit von BMP-7 diese statistisch Signifikant höher war als in Abwesenheit (p<0,001). Die HA60 Keramik zeigte die größte Knochenneubildungspotenz.
BMP-7 ist ein bereits lang bekannter und erforschter Wachstumsfaktor. Der positive Effekt auf die Knochenneubildung, welcher in der vorliegenden Studie gezeigt wurde, konnte auch von vielen anderen Autoren beschrieben werden. Es konnte in dieser Studie desweiteren der beschleunigende Einfluss auf den Abbau der Kalziumphosphatkeramiken nachgewiesen werden.

Translation of the abstract (English)

The aim of this study was to evaluate the chemical degradation pattern plus the ectopic ossific
characteristics of three different bone substitution materials in the presence and non-presence
of BMP-7.
In 10 Göttinger minipigs, three different bone substitution materials (TCP,HA,HA60) were
ectopically implanted. After three months, the minipigs were sacrificed and the ceramics were
explanted. During those three months, computed tomographies and fluorescent markers were
performed.
The fabricated biological preperations were analysed histomorphometrically and
histologically, in addition to evaluation using fluorescence microscopy.
HA and HA60 ceramics showed an osteoclast dependent resorption. TCP showed only
degradation. The comparison of the chemical degregation pattern in presence and nonpresence
of BMP-7 was stastically significant (p≤0,001).
All three bone substitution materials showed ectopic bone formation.
The presence of BMP-7 showed statistical significance (p≤0,001).
BMP-7 is an already known and long-studied growth factor. The positive effect on bone
formation, which was shown in the present study, could also be described by many other
authors. Furthermore, this study demonstrated the accelerating influence on the degradation
of calcium phosphate ceramics.
